Abstract

Leptin is a hormone produced by adipocytes and plays an important role in wound healing. The objectives of this research were to identify leptin and total leukocyte after incision on rat fed high fat diet and zinc topicalapplication. Sixteen male Sprague Dawley rats at 3 months of age were used in this study. Rats were randomly allotted into 4 groups (A,B,C and D) of 4. Rats in group A and B were fed normal diet, while rats in group C and D were fed high fat diet. After 2 months of treatment, skin incision was done at the back side of the rat. Incision wound was then closed with single interrupted suture. The wound of the rats in group A and C was treated with vaseline, while in group B and D was treated with zinc 10%. Three days after surgery, blood was collected from each rat for leptin analysis and total leukocyte. Leptin level and total leukocyte were analyzed statistically using ANOVA for factorial experiment (2x2). The results showed that leptin level was significantly higher in rats fedhigh fat diet compared to that of the rats fed normal diet (P<0.05). There was no significant different on total leukocyte. Therefore, it was concluded that leptin level was affected by diet, while total leukocyte was notaffected neither by diet nor topical application. However, total leukocyte tends to be lowered in the rat with zinc topical application.Keywords: high fat diet, leptin, zinc, total leucocyte, Sprague Dawley rats

Abstract

In the Java Sea, there are a lot of dolphins, that often snagged fishing nets. While In Indonesia, bottlenosedolphins are mammals protected water. The aims of this research was to study the erythrocytes profile inbottlenose dolphins before and after the captivity. Seven dolphins were used in this research. Blood samples weretaken out from the superficial vein of tail for examination of the number of erythrocytes, hemoglobin levels andvalue of hematokrit. Sampling was done when the dolphins appointed from the waters of the Java Sea and afterexperiencing dolphin captivity in PT. WSI, Kendal, Central Java. The data before and after captivity were compared and tested with the statistical analysis on the level of significance of the paired t-test 95%. The results showed the significant increase in the number of erythrocytes, hemoglobin levels and the values of hematokrit significantly. It was concluded that captivity done by WSI Corporation, Kendal, Central Java did increase the profile of erythrocytes in bottlenose dolphins.Keyword : Bottlenose dolphins, captivity, profile erythrocytes, superficial vein of tail, paired t-test

Abstract

Newcastle disease (ND) is caused by Avian paramyxovirus, family Paramyxoviridae, one of the major diseases in chickens. This research was aimed to find lesions in chicken's embryo organs macroscopically andmicroscopically, infected by pathogenic ND virus. Embryonic chicken eggs (ECE) were inoculated by the ND Salatiga virus and ND La Sota virus as a control avirulent virus. Aquabidestilata used as a negative control. ECEwich showed the death of the embryos, stored in the refrigerator. The allantois fluid was collected, for further examination of viral growth. Chicken embryos that died then observed macroscopically. The organs of chicken embryos were made into histopathologic preparations stained with Hematoxylin and Eosin (H&E) for microscopic analysis. The identification of ND virus growth on isolates was done by haemagglutination andhaemagglutination inhibition test using an anti-ND serum. The chicken embryos that were infected by the ND Salatiga virus died approximately 26 hours post-inoculation. Macroscopic lesions were visible as haemorrhagein the skin. Microscopic lesions indicated the congestion and haemorrhage in lungs, inflammation and congestion in the skin, congestion in intestines, liver, kidneys and heart. There was also mild congestion on theskin in chicken embryos infected by ND La Sota virus. The microscopic lesions showed congestion in lungs, liver, kidneys and heart, also the inflammation and congestion on the skin. The macroscopic and microscopic lesions of chicken embryos infected by the ND Salatiga virus were more severe than lesions caused by ND La Sota virus.Key words: Newcastle disease, chicken embryos, macroscopic lesions, microscopic lesions, La Sota

Abstract

In the present study, Dactylogyrus sp. was examined in the gill(s) of the ornamental marine fish due to the fact that this kind of parasite is very often found in those fishes without any pathognomonic clinical signs orpathologic anantomic lesions. The gill(s) samples collection and examination were performed in the Laboratory of Balai Karantina Ikan, Pengendalian Mutu dan Keamanan Hasil Perikanan. during Janury-July 2011. At first, the gills were examined natively and were subsequently processed for histopathologic examination. In the native examination, it was found the Dactylogyrus sp. with their size(s) ranging from 200-500 um. At necropsy, the gills were swollen and looked darker than normal. Based on the histopathologic lesions, the epithelial cells of the gills lamellas either primary or secondary became hyperplasia and fusion. The fusion of the gills is primarily thought to cause fatal to the fish due to significant oxygen depletion.Keywords: ornamental marine fish, gill, Dactylogyrus sp., native examination, hiperplasia and fusion.

Abstract

Campylobacter jejuni is responsible for about 90% of cases of campylobacteriosis in human with gastroenteritis. Efforts to prevent the incidence of campylobacteriosis in human should be started with anassessment on its level in poultry. This study aims to determine the factors associated with the presence of C. jejuni on village chicken in Mataram, Nusa Tenggara barat. Association between C. jejuni as the dependent variable with independent variables were analyzed using logistic regression. Prevalence of C. jejuni from a total of 216 samples in Mataram at chicken level was 35.6% and while at the farm level was 70.5% (31/44). Variables showed a positive association of C. jejuni occurrence in chicken consist of age of chicken more than 3 months, the number of chicken ownership, and cage with soil floor. While the variable showed a negative association to the incidence of C. jejuni was drinking water of chicken originated from wells. Model obtained using logistic regression in association with occurence of C. jejuni on village chicken in Mataram was Y = -2.033 + 0.271 (age of chicken more than 3 month) + 0.103 (number of chicken ownership) + 0.741 (cage with soil floor) - 0.890 (drinking water of chicken originated from wells)Keywords: Campylobacter jejuni, logistic regression, village chicken, drinking water, wells

Abstract

Osteogenic activity  in  normal fracture  healing will start  early  at  around  1 week after  the  repair ,  but not necessarily occur in fracture healing with the aid of grafts. Demineralized freeze dried bovine bone xenograft (DFDBBX) is a bone graft material made from cows that are    widely produced recently to assist the process of bone  grafting  in  humans,  but  has  not  been  used  on  animals.  It  does  not  know  how  it  works  in  helping  to  repair  the bone.  T wenty male SD rats  weighing 150 g, were divided into  2 groups of 10 rats  each. Group I  was the  control group, the  rats  were fractured  and reconnected  immediately  using intramedullar  pin fixation,  Group II  was a group  of  rats  that  were    fractured  and  reconnected  immediately  using  pin  fixation  intramedular  plus  graft material of freeze dried bovine bone xenograft. Fracture of the left femur was made in diaphysis part of bone. Evaluation  of  joints  performed  regularly  at  1  week  ,  2  weeks,  4  weeks,  6  weeks  and  8  weeks  post  surgery respectively by selecting at random, each group of 2 rats. Rats at each observation were collected two  head / group,      rats    were  then  killed,  the  bone  parts  to  be  joined  was  taken  and  stored  in  10%  of  formalin  for histopathologic  examination.  Some  pieces  of  the  bones  were  used  for  the  analysis  of  bone  morphogenetic  protein 2 (BMP-2 ) by immunohistochemical method. The results were then analyzed descriptively to see the changes directly on the X-rays and tissue. Overall results of this study showed that the graft material of DFDBBX were osteocondusive  rather  than osteoinductive  properties.  Nevertheless,  the  biomaterial  is  useful  for  orthopedic purposes  primarily  of  handling  cases  with    partial  fracture  of  bone  tissue  which  is  lost.  Due  to  the  nature osteocondusive  of  DFDBBX  will  make  cells  to  grow  and  to  unify  the  separate  bone  fragments.Keywords  :  fracture,  osteogenic  activity ,  demineralized  freeze  dried  bovine  bone  xenograft,  bone morphogenetic  protein-2,  immunohistochemical  analysis

Abstract

Kerapu macan  (Epinephelus fuscoguttatus)    is  one of the  kerapu  fishes  that  has been successfully  bred by the  farmers.  The  survival  rate  of  the  fish  reaches  up  to  the  40%  at  the  fish  hatchery  of  the  home  industry  of  in  Bali. The disease in which may cause significant mortality in kerapu fish, especially for the larvae and juveniles is viral nervous necrosis (VNN). We therefore developed and applied the immunological diagnostic approach of immunocytochemistry technique of streptavidin biotin (SB) for early detection of  VNN. Mucous samples of VNN  infected-  kerapu  fish  in  vivo  were   firstly  detected  by  reverse  transcriptase-polymerase  chain  reaction  (R T -PCR)  for  the  presence  of  DNA  VNN.  If  it  is  RT -PCR-positive  VNN,  the  mucous  samples  were  then  tested  by  SB. Results of the present study indicated that the VNN virus could be detected with SB technique within only 24 hours post infection.  It  was concluded that  rapid  and accurate SB technique  is  suitable and appropriate to  be applied  for  routine  control  and  prevention  national  program  in  the  Fish  Quarantine  for  Indonesia  because  no  need of  fish  sacrification,  and  scientific,  law  and  international    accepted,  and  even  no  hazardous  environmental contamination.Keywords:  kerapu  macan  (Epinephelus  fuscoguttatus),VNN,  mucous,  SB,  early  detection

Abstract

The objectives of this study were to reveals factors associated with level of herd immunity against rabies. Four hundred and eighteen blood samples were collected from five subdistricts and 14 villages using stratified and  household  cluster  methods. The  result  in  ELISA  test  is    ≥  0,5  IU/ml,  dog  titers  sample  was  stated  as protective  to  rabies.  The result  showed  that  the  prevalence  of  protective  antibody  titers  was very  low  i.e.  3.35%. Linear regression  analysis reveals  subdistrict of    Nusaniwe, subdistrict of Baguala, dog owner education, post-vaccination  period  of  0  -  6  months, sex,  and  dogs  kept  as  a  guard  house  contribute  to  protective  level  of  antibody titers. Unweighted logistic regression revealed that subdistrict of Baguala (OR = 0.05), subdistrict of Sirimau (OR = 0.09), dog kept as a guard house (OR = 3.96), dog owner education (OR = 12.29), and post-vaccination period  of  0  -  6  months  (OR  =  27.08)  are  significantly  associated  with  dog's  immunity .  V accination  programs  need to  be  improved  by  considering  the  four  factors  that  emerged  in  these  two  regression  results.Keywords  :  rabies,  level  of  dog's  immunity ,  ELISA,  Ambon,  post-vaccination

Abstract

Abstract         One of potencies of marine waters that is developed, and starts to have the international market is grouper fish. Tiger grouper culture can not be separated from disease factors that may attack and destroy the cultivation. One of the important diseases that can cause mass mortality in fish, especially in grouper larvae and juveniles stadium is viral nervous necrosis (VNN). Examination by using immunocytochemical method of streptavidin biotin (SB) undertaken to develop early detection of VNN in order to be applied in preventing spread of the VNN disease in seeds of tiger groupers that are already distributed worldwide in Indonesia. And also, this SB methodology can also be applied to the tiger groupers without any injuring or even killing of the fish for the purpose of sampling. In the present study, the preliminary test was done to determine the infectivity level of the inoculum (VNN virus) owned by the Office of Fish Quarantine, Quality Control, and Fishery Safety Class I Denpasar, Bali. Whereas, the main test was done by collecting water sample from tiger grouper fish farming that was infected with VNN. The water sample was examined by using  immunocytochemical method of SB after being confirmed by RT-PCR-positive test that was previously purified. Based on both SB and PCR tests having VNN positive results, it is concluded that  the immunocytochemical method of SB could be applied as early, rapid and accurate detection of VNN  in only a 24 hours post infection by using water as the sample. Keywords: VNN, water sample, streptavidin biotin, RT-PCR, tiger grouper

Abstract

Infectious myonecrosis virus (IMNV) is one of the shrimp viral diseases caused by a double-stranded RNA virus, non-enveloped, and belongs to the family group Totiviridae. Weakness diagnosis of RNA viral diseases due to the RNA viruses are unstable and highly reactive. So far the diagnostic methods for detecting IMNV  with high accuracy is based on biomolecular test, such as reverse transcription polymerase chain reaction (RT-PCR). The RT PCR generally requires 4-8 hours of testing time, and also requires thermal cycler with a commercial kit materials which is relatively expensive. The purpose of the present study is to find and determine an alternative isolation of RNA from the samples and to determine whether the reverse transcription loop-mediated isothermal amplification (RT-LAMP) method based on biomolecular approach  can be an alternative testing method for diagnosis of IMNV in the white shrimps(L.vannamei).The RT-LAMP is more simple, straightforward, but it has a high sensitivity. Direct and indirect RNA extraction methods from samples and FTA cards give good results as a template in RT-PCR and RT-LAMP IMNV. The FTA card as one of the RNA samples of  storage media provides an alternative storage method that is easy, simple and has a shelf life of RNA in the long term (up to 6 months). Incubation time of RT-LAMP IMNV at 400C is between 15-30 minutes. Amplification time of RT-LAMP IMNV at 56o C is between 70-110 minutes. In the present study, the white shrimps (L.vannamei) that are positive IMNV-RT-LAMP provides 100% compliance results when tested by the RT-PCR method. The time of RT-LAMP amplification IMNV ranged from 70-110 minutes shorter than that of conventional PCR (180-240 minutes). Overall the RT-LAMP method for  IMNV can be applied as a method of testing for confirmative IMNV diagnosis in the white shrimps (L.vannamei). Keywords: IMNV, RT-LAMP, RNA, FTA cards,  white shrimp (L.
