Abstract

The corpus luteum is a temporary endocrine gland formed from the ruptured follicle after ovulation, which plays an essential role in maintaining pregnancy in mammals. Persistent corpus luteum (PCL) is a condition in which the corpus luteum fails to regress and remains in the ovary for a prolonged period, thereby becoming one of the causes of reduced productivity in female cattle due to reproductive disorders. The clinical symptom commonly observed in PCL is prolonged anestrus, causing the reproductive process to cease. The CYP11A1 gene is known to play a role in the regulation of prostaglandin hormones, which are important for corpus luteum (CL) regression. This study aimed to quantitatively evaluate the CYP11A1 gene in cattle affected by PCL. DNA band analysis was performed to determine the presence of this gene in PCL cattle. The results showed an increased expression of the CYP11A1 gene in cattle with PCL compared to normal cattle. These findings are expected to provide information regarding the potential of CYP11A1 as a marker for the diagnosis of cattle with PCL, so that it may be used as a selection marker for productive or superior female cattle.

Abstract

Antibiotic resistance in livestock is an increasingly significant issue that affects animal health, food safety, and public health. Staphylococcus aureus is an important pathogenic bacterium in layer chickens and has the potential to serve as a source of resistant bacterial spread. This study aimed to determine the antibiotic resistance profile of S. aureus isolated from cloacal swabs of layer chickens in Bangli Regency, Bali. A total of 20 cloacal swab samples from clinically healthy layer chickens were cultured on Mannitol Salt Agar and identified through Gram staining, the catalase test, and mannitol fermentation characteristics. Antibiotic susceptibility testing was performed using the Kirby–Bauer disk diffusion method and interpreted according to Clinical and Laboratory Standards Institute. The results showed that 12 isolates (60%) were identified as S. aureus, with a higher prevalence in layer chickens (80%) compared with pre-layer chickens (40%). The highest resistance was observed against erythromycin (91.67%), followed by ciprofloxacin and sulfamethoxazole–trimethoprim (each 41.67%), while all isolates were sensitive to chloramphenicol. These findings indicate that S. aureus from layer chickens in Bangli Regency exhibits a high level of resistance to erythromycin, whereas chloramphenicol remains effective. The results highlight the need for prudent antibiotic use, strengthened biosecurity, and routine resistance monitoring to prevent the spread of resistant bacteria in layer farms.

Abstract

Bacillus thuringiensis (Bt) is widely used as a microbial agent in bioinsecticide formulations and is generally regarded as safe for non-target organisms. However, clinical reports of corneal ulceration following ocular exposure to Bt-based products have raised concerns regarding its potential toxicity to the eye. This study aimed to evaluate corneal epithelial integrity, histopathological changes, neutrophil infiltration, and the expression of inflammatory and oxidative stress markers, including tumor necrosis factor-alpha (TNF-α) and malondialdehyde (MDA), in Wistar rats following topical exposure to Bt bioinsecticide. A post-test-only control group design was applied using 24 male Wistar rats divided into one control and three treatment groups receiving Bt concentrations of 8 g/L, 10 g/L, and 12 g/L. Corneal epithelial defects were assessed using fluorescein staining, while histological examination and semi-quantitative measurements of neutrophils were performed on enucleated eyes. TNF-α levels were measured using ELISA, and MDA levels were determined via the thiobarbituric acid (TBA) assay. There was epithelial desquamation, increased epithelial thickness, and higher neutrophil infiltration in the treatment groups compared to controls, although these parameters did not reach statistical significance. Notably, TNF-α levels demonstrated a significant increase in the 10 g/L treatment group (p = 0.023), indicating activation of local inflammatory pathways. MDA levels showed a non-significant upward trend. Topical exposure to Bt can elicit corneal inflammatory responses even in the absence of clear structural damage, highlighting the need for caution regarding ocular contact with Bt-based bioinsecticides

Abstract

Abstract

Research objective: to determine the diversity and infection levels of gastrointestinal nematodes in Capra hircus based on age differences in Desa Oesao, Kabupaten Kupang. A total of 30 fecal samples were collected, consisting of 15 samples from young goats and 15 from adult goats, without sex differentiation. Grazing field temperature was also recorded. Samples were examined using sedimentation and flotation methods to identify helminth eggs, while infection intensity was determined using the McMaster method and expressed as eggs per gram (EPG) of feces. The results showed similar nematode diversity in both age groups, except for Chabertia ovina and Ostertagia sp., which were not detected in adult goats. T-test analysis indicated that infection levels in young goats were significantly higher (p < 0.05) than in adults. In young goats, infection levels were highest for Strongyloides papillosus (1,318 EPG) and Haemonchus contortus (700 EPG), followed by Chabertia ovina (67 EPG), Trichostrongylus sp. (63 EPG), Oesophagostomum sp. (43 EPG), Trichuris sp. (43 EPG), and Ostertagia sp. (20 EPG). In adult goats, lower EPG values were recorded for the same species except Chabertia ovina and Ostertagia sp. Infection diversity and intensity were influenced by immunity, age, feed sources, and grazing environment
