Abstract

Tea is one of the most popular beverages in the world. Produced from Camellia sinensis leaves, tea has been studied to provide health benefits due to the content of important metabolites. This study aimed to investigate the catechin contents, antioxidant, and antibacterial activities of Indonesian tea varieties, namely green tea, black tea, and white tea. Tea infusion was prepared by extracting 1 g of each sample into 10 mL of distilled water and incubated at 80 °C for 60 min. The catechin and epigallocatechin gallate (EGCG) contents of tea extracts were determined using high-performance liquid chromatography (HPLC). Antioxidant activity was measured using the free radical method with 2,2-diphenyl-1-pycrylhidrazyl (DPPH), while antimicrobial activity was assesed using paper disc diffusion assay. The results indicated that green tea had the highest contents of catechin (646 ± 17.14 mg/L) and EGCG (997.8 ± 36.72 mg/L), and antioxidant activity with IC50 of 5.65 µg/mL. Furthermore, green tea and white tea extracts showed inhibitory activity against Gram-positive bacteria such as Micrococcus luteus, Bacillus subtilis, and Staphylococcus aureus whereas black tea had no activity against all bacterial strains tested. Generally, we concluded that white tea and green tea contributed to the higher content of catechins and exhibited strong antioxidant and antibacterial properties.

Abstract

Cancer, a non-communicable illness, is the leading cause of death worldwide. In 2030, cancer is expected to exceed 21 million cases and 13 million cancer deaths globally. One of the most prevalent and significant types is prostate cancer (PCa). It is commonly associated with adenocarcinoma, which develops from the mucous glands within the organ. This review highlights available detection systems, and therapeutic options in PCa management. One prevention of deadly PCa is early diagnoses, such as prostate-specific antigen (PSA) screening or genomic profiling. Further testing like MRI or CT scan may also be needed to detect cancers that have progressed to other body regions. There are several possible treatments for PCa, including watchful cancer waiting, surgery, radiotherapy, hormone therapy, and chemotherapy. Based on current studies, androgen deprivation therapy (ADT) combined with docetaxel therapy enhanced great results to treat advanced PCa. The latest development, called theranostics, is a single entity that can perform both diagnostic and therapeutic functions. It can detect disease borders, track therapy in real-time, and provide prognostic data. The FDA has already authorized two prostate-specific membrane antigen (PSMA) positron emission tomography (PET) devices: including Gallium 68 PSMA-11 (Ga 68 PSMA-11) and Pylarify (piflufolastat F 18).

Abstract

Recombinant human epidermal growth factor (rhEGF) has been developed to provide the protein for therapeutic uses. Isopropyl β-d-1-thiogalactopyranoside (IPTG)-induced expression in Escherichia coli BL21(DE3) has shown the most effective system among other inducers systems in a similar host. However, suitable conditions related to IPTG concentration, incubation time, and incubation temperature are different depending on the amino acid content of the recombinant protein. This study aimed to evaluate the effects of various IPTG concentration, incubation time, and incubation temperature on rhEGF concentration. According to each analysis of those factors, induction with 0.05 mM IPTG for 2 h at 23°C was the most appropriate condition to obtain the highest concentration of rhEGF. The rhEGF was positively confirmed with a monoclonal antihEGF antibody and purified in high purity reaching 95.2%, yet recovery was low (1.44%) due to loss in fractions containing endogenous proteins. Therefore, further studies related to type of matrix, column length, and sample concentration in applying size exclusion chromatography are requested for higher recovery

Abstract

Sunda porcupine (Hystrix javanica F. Cuvier 1823) is an endemic fauna from Indonesia that its quills are believed to have medical benefits by local people in some regions of Indonesia. However, the benefits have never been well recorded nor proven scientifically. Local people believe the Sunda porcupine’s quills have efficacy to relieve property for treating toothache. There is limited research on Sunda porcupine’s quills, especially the active compounds, which may affect toothache. This research aims to perform basic pharmacological experiments on Sunda porcupine’s quill samples, which includes screening for the active compounds and determining the LC50 toxicity using brine shrimp lethality test (BSLT) method. Sunda porcupine’s quills were first prepared into simplicia powder (60 mesh in size) and then extracted with 70% ethanol by maceration to produce crude extract. We found that the crude extract of Sunda porcupine’s quills contains some active compounds, including alkaloids, flavonoids, saponins, triterpenoids, steroids, and peptides. The LC50 value of the crude extract was 2,683.19 ppm; thus, categorized as non-toxic. These findings can be used to identify the active compounds in Sunda porcupine’s quills and can be used as a background for further research.

Abstract

The toxic fermentation inhibitors in lignocellulosic hydrolysates pose significant problems for the production of second-generation biofuels and biochemicals. Among these inhibitors, 5-(hydroxymethyl)furfural (HMF) and furfural are specifically well known. This study investigated the furfural and 5-HMF tolerance in Candida strains, which could be used for the development of advanced generation bioethanol processes. The 10 isolates of Candida were selected based on the cell growth and bioethanol production on YPD medium containing several concentrations of furfural and 5-HMF by using spectrophotometer and HPLC. Candida parapsilosis Y80 could grow and produce bioethanol in the medium that contains furfural and 5-HMF with a concentration of 60 mM and 40 mM, respectively. Based on the results, C. parapsilosis Y80 has potential activity in the development of bioethanol fermentation.

Abstract

Probiotic has been widely used in functional food because of numerous advantages for health. MRS broth is commonly used as standard medium in studying lactobacilli. However, in some communities - like muslim and vegetarian society, components in MRS broth/medium become an issue. Beef extract and peptone – animal derived substances as nitrogen sources in the MRS medium should be avoided for the vegetarian. Meanwhile, for the muslim society, all components must be halal-certified including those animal derived ingredients. Therefore, several alternative sources for beef extract and peptone substitution were studied. Combination of alternative nitrogen sources was applied. In order to increase the effect of the alternative nitrogen sources, alternative carbon sources were also included. This is the first report about effects of L. brevis media components on cells growth to expression level of surface layer protein (Slp). Whey, lactose, sucrose, and galactose showed high contribution to L. brevis growth. However, the tested concentration of those substances were not sufficient to obtain equal bacterial growth and Slp expression than that of MRS broth. In addition, yeast extract appeared necessary to maintain cell wall and Slp expression.

Abstract

The growth and development of plants are determined by photosynthesis, which ultimately results in sucrose. Sucrose is synthesized in the source then translocated to all parts of the plant (sink). The translocation process of sucrose from source to sink is controlled by sucrose protein called sucrose transporter. SoSUT1 is a gene that encodes a sucrose transporter 1 (SUT1) protein in sugarcane. Rice transformation with the SoSUT1 causes overexpression SUT, which is expected to increase the translocation of sucrose into the seed of rice plants. This research was conducted by introducing SoSUT1 in rice plants Inpari 14 SS. Transformation using Agrobacterium tumefaciens vector in apical bud explant Indica rice cv. Inpari 14 SS results in 26 events positive rice contains genes SoSUT1. This study aims to elucidate the inheritance of transgene in the next generation and to characterize its effect on the morphology and the yields of a subsequent generation. The study is conducted by planting the seeds of T1 and T2 plants in media containing Hygromycin and using PCR analysis for further analysis. As the results, from 26 events on the T1 plant, only 3 events of T3 plants were confirmed on the T3 plant. The overexpression of the SoSUT1 gene could increase the number of tillers, the number of productive tillers, panicle length, and panicle exit length, also increasing the number of spherical grains, reducing the number of empty grains and increasing the weight of 1000 grains.

Abstract

The study aimed to determine the effectiveness of metabolites from Lactic Acid Bacteria (LAB) derived Masin (fermented sauce from Sumbawa) as antioxidant, antidiabetic, and antibacterial compounds. The LAB isolates were isolated from various strains of Staphylococcus piscifermentan which consisted of Staphylococcus piscifermentans strain CIP103958 (code: 2), strain BULST54 (code: 17), strain SK03 (code: 11), strain ATCC 51136 (code: 34), strain PCM 2409 (code: 28) and strain PU-87 (code: 5). The Metabolites of LAB were analyzed by the bioprospecting test to indicate antidiabetic, antioxidant and antibacterial activities. The isolate (Code: 5) at 500 ug/ml showed the most effective antioxidant activity up to 71%. The isolate (code: 28), at 300 ug/ml revealed to have the most antidiabetic activity up to 43 %. The isolate (code: 2) showed moderate antibacterial activity with the inhibition zone of 5.59 mm. The results of the antidiabetic, antioxidant and antibacterial activity showed that the secondary metabolites produced by LAB from the Masin have broad activities as an antidiabetic, antioxidant and antibacterial.

Abstract

Bacterial leaf blight (BLB) caused by Xanthomonas oryzae pv. oryzae (Xoo) is a serious disease in rice plants worldwide. Yield losses caused by Xoo can be as high as 50% in some parts of Asia. Xa7 gene can potentially confer a broad resistance to BLB. Evaluation of disease resistance characteristics in early breeding generations of rice is important to develop varieties with better resistance. This study reports the evaluation of 167 Indonesian rice germplasms against three BLB isolates/pathotypes in a green house setting and the genotyping of 56 Indonesian rice germplasm using 12 SSR markers linked to Xa7 BLB resistance gene. The majority of the indigenous rice germplasms was found to be susceptible to three BLB isolates/pathotypes tested. Sate Liko from Bantul, Yogyakarta, Horeg from Cirebon, West Java and Sijem from Malang, East Java revealed consistent resistance to three isolates/pathotypes tested based on BLB evaluation in a greenhouse, UPGMA analysis, and genotyping. Pathotype XII displayed more virulence to Indonesian rice varieties tested compared to pathotypes VI and V. The association analysis using the General linear model identified six markers associated with BLB resistance and two markers were highly associated (RM20589 and RM20590). This information will be useful for future studies of BLB resistance in rice plants.
