Abstract

Kratom (Mitragyna speciosa Korth.) is a tropical plant native to Southeast Asia and has demonstrated antibacterial activity; however, information regarding its activity against biofilm formation by Cutibacterium acnes remains limited. This study aimed to evaluate the antibacterial and biofilm formation inhibitory activities of ethanolic kratom leaf extract against C. acnes. Antibacterial activity was evaluated using the disc diffusion assay. Biofilm formation inhibition was assessed using a microtiter plate crystal violet assay with optical density measurement at 595 nm. The extract was tested at 1%, 0.5%, 0.25%, and 0.125% (w/v). The 1% extract produced the largest antibacterial inhibition zone among the extract treatments (10.8 mm). In the biofilm assay, the 1% extract produced 70.67% inhibition after 24 h and 58.75% after 48 h. The lowest tested concentration producing at least 50% biofilm inhibition was 0.5% (w/v) at both time points. These findings indicate concentration-related antibacterial and biofilm formation inhibitory activities of kratom leaf extract against C. acnes. The specific active compounds responsible for these effects remain to be identified.

Abstract

Majapahit (Crescentia cujete L.) is a medicinal plant traditionally used for various therapeutic purposes and known to contain diverse secondary metabolites with potential pharmacological activities. This study aimed to characterize the phytochemical composition and evaluate the analgesic activity of a 70% ethanol extract of majapahit stem bark in male Swiss Webster mice. The stem bark was extracted by maceration using 70% ethanol. The resulting extract was characterized based on loss on drying, moisture content, total ash content, qualitative phytochemical screening, and quantitative determination of selected secondary metabolites by UV–Vis spectrophotometry. Analgesic activity was evaluated using the hot plate method at extract doses of 125, 250, and 375 mg/kg body weight, with 0.5% CMC-Na and mefenamic acid used as the negative and positive controls, respectively. Loss on drying, moisture content, and total ash content of the crude plant material were 2.90%, 9.73%, and 12.51%, respectively, while the extract yield was 3.29%. Phytochemical screening detected alkaloids, flavonoids, saponins, and tannins. UV–Vis analysis yielded measured levels of 3.31 mg/kg flavonoids, 2.40 mg/kg tannins, 1.09 mg/kg alkaloids, and 0.74 mg/kg saponins. The 375 mg/kg dose produced the highest analgesic response among the extract-treated groups, with 45.0% response protection and 98.0% analgesic activity. Statistical analysis indicated significant differences among treatment groups (p < 0.05), although the Tukey test showed no significant difference among the extract-treated groups and the positive control. These findings indicate that the ethanol extract of majapahit stem bark has potential analgesic activity in male Swiss Webster mice.

Abstract

Encapsulation is a method for embryogenic tissue coating system by Na-alginate which has synthetic seed preservation purpose. To produce sugarcane synthetic seed need Coleoptillar embryogenic callus and apical bud that it is encapsulated by encapsulation media (MS0 + 4% Na-alginat+ 100 mM CaCl2). In this study, PEG (0%, 1%, 2%, 3%) and PBZ (0 ppm, 1 ppm, 2 ppm, 3 ppm) add to media encapsulation which are give desiccation effect (PEG) and Giberellin synthesis blockade system. Both of PEG or PBZ is used to sugarcane synthetic seed growth retardation that it is implied as storage period. The result are show that PEG 2% is the most effective concentration which is added in encapsulation media. Dessiccation effect of PEG 2% can make synthetic seed storage period until 3 weeks, synthetic seed regeneration produce 89,32 planlets and planlets clorophill concentration is 26,03 ????g/mL. PBZ 1 ppm is the most effective concentration to make storage period until 5 weeks, synthetic seed regeneration produce 39 planlets and planlets clorophill concentration is 35,81 ????g/mL.
