Abstract

ABSTRACT For the past three decades, PUFA has been recognized as importnant energy sources and cell membrane component. PUFA also plays key roles in many cellular events such as gene regulation and leptin activation. The role of this fatty acid is still unknown especially in vitro. The purpose of this research was to examine the effect of PUFA of the corn oil in increasing leptin concentration by PPARγ activation. Preadipocyte culture from rabbit was incubated by PUFA of corn oil. PUFA have been given on doses 0, 4, 8 and 16 µM. PPARγ protein and leptin levels were analyzed by ELISA. PPARγ expression  was  analyzed  by  immunocytochemistry.  The research  result  showed  that  every  treatment  has  increased PPARγ levels inconsistently. PPARγ concentration increased significantly on 4 µM PUFA treatment compared to control, 8 and 16 µM PUFA of corn oil. But there were no significant difference between control and 8 µM PUFA treatments. Leptin levels increased significantly between administration on 4 µM PUFA and 16 µM PUFA of corn oil administration. Leptin levels was not significantly different between control and 8 µM PUFA. There was also strong correlation between PPARγ expression and leptin levels. PUFA of corn oil increases leptin levels through PPARγ activation. Key words: PUFA, preadipocyte, leptin, PPARγ

Abstract

ABSTRACT Permanent  visual  damage  caused  by  methanol  intoxication  has  been  associated  with  prolonged  exposures  to  blood formate  concentrations.  Formate  accumulation  disrupt  mitochondrial  energy  production,  which  subsequently  lead  to caspase 9, 3 activation and finally causes apoptosis.  Folic acid (FA) has been used as adjunctive therapy to enhance the rate  of  formate  metabolism.  Extract  Ginkgo  biloba  (EGb)  the  most  popular  herbal  therapy  possess  is  antioxidant  and antiapoptotic capacity. On the other hand, Methyl cobalamin, a kind of endogenous co enzyme B 12 can promote nucleic acid and protein synthesis. To compare the effect of FA, (EGb), and Methyl Cobalamin (MC) to Apoptotic Index (AI) and Caspase 3 expression of  retinal ganglion cell induced by methanol intoxication (MI). Experimental study was conducted in Pharmacology and Biomedic laboratory of Brawijaya University for two weeks. Fifteen rats were included, and divided into five groups: (a) control, (b) Methanol Intoxication only, (c) Folic Acid 1 mg day per os, (d) Extract Ginkgo biloba 50 mg/kb  per  os,  and  (e)  Methyl  Cobalamin  500  microgram/kg  intraperitoneally  for  three  days.  MI  were  done  by  giving methanol 4 g/kg in the first day, and 2 g/kg for two days after. Abstract

ABSTRACT Guillain Barre Syndrome is one of the leading diseases from Acute Flaccid Paralysis after the decrease in poliomyelitis. InGuillain Barre Syndrome, the motoric muscles becomes weak and it could also attack the respiratory muscle which is lifethreatening to patients. The objective of this study is to report Guillain Barre Syndrome cases from June 1999 until June2005 based on the clinical manifestation and laboratory in children care unit of dr. Saiful Anwar hospital. In this researchthere were 38 Guillain Barre Syndrome cases which were diagnosed using the Gilroy and Meyer criteria. 22 patients(57,9%)  were  male,  16  patients  (42,1%)  were  female.  18  patients  (43,36%)  were  below  5  years  of  age,  13  patients(34,21%) were between 5 to 10 years of age and 7 patients (18,42%) were between the age of 10 to15 years. The mostcommon infection that precedes was upper respiratory infenction which amounts to 32 cases (84,21%). The latent phasebetween 8 to 14 days were 11 patients (28,94%) and 15 to 21 days were 22 patients (57,89%). The progresive phase 1 to7 days occured in 17 patients (44,73%), 8 to 14 days also in 17 patients (44,73%).The clinical features were paresthesiasfollowed by weakness to 23 patients (60,52%) and weakness only in 15 patients (39,47%). Abstract

ABSTRACT Death  caused  by  cardiovascular  and  cerebrovascular diseases  are  still  the  highest.  Atherosclerosis  due  to hypercholesterolemia is a predispositioning factor of both diseases. Curcumin has an antioxidant character that can inhibit lipid peroxidase to atherogenesis process. The aim of this study was to observe the effect of curcumin in decreasing total cholesterol level, LDL-cholesterol, the number of  F2-isoprostan and foam cell of aorta’s wall of white rats strain wistar after giving atherogenic diet for 10 weeks. There were 6 groups in this study (n=24): atherogenic diet group (group I as positive control), atherogenic diet group + curcumin 50 mg/kg b.w. per day (group II), atherogenic diet group + curcumin 100 mg/kg b.w. per day (group III), atherogenic diet group + curcumin 200 mg/kg b.w. per day (group IV), atherogenic diet group + curcumin 400 mg/kg b.w. per day (group V), normal diet group (group VI as negative control). After 10th, the level of total cholesterol and LDL-cholesterol were measured by spectrofotometri. Staining F2-isoprostan was done with Avidin Biotin Complex imunohystochemistry method, and staining foam cell with HE-oil Red O. The number of  F2-isoprostan and foam cells were counting semi quantitatively using  light microscope. The highest total cholesterol level was found in group I (275,15 ± 10,01; Mean ± SD), the highest LDL-cholesterol level was found in group I (158,15 ± 12,19). Statistically total cholesterol level and LDL-cholesterol level in group I were significantly higher (p ≤ 0,05) than other

Abstract

ABSTRACT Recently it has been reported that some women have suffered from illness complication as the effect of the use the liquid silicone to beautify themselves. Knowledge deprivation and eagerness to achieve complete beauty in an easy and cheap way makes them undertake a short cut which is getting the injection. The research is intended to know some factors underlying a person’s motivation to use liquid silicone injection in Malang in 2001. The research was a descriptive cross sectional method. The population was person who use liquid silicone injection in Malang 2001, this assume were 20 persons during the research (June – December 2001). This research showed that 15 respondents (75 %) of the samples knew the benefits of liquid silicone injection but did not know the effect of the injection, the education of this respondents were secondary and tertiary level 13 respondents (65 %), 6 respondents (30 %) of the sample working as beauticians, 6 respondents (30 %) of the samples having salary  Rp. 250.000,00 - Rp.500.000,00 per month,  9 respondents (45 %) of the samples feeling more beautiful and self confident after getting liquid silicone injection, 6 respondents (30 %) of the samples getting family permission, 12 respondents (60 %) of the samples having a social network to use the injection, 5 respondents (25 %) of the samples getting secondary health services and 4 respondents (20 %) of the samples getting

Abstract

ABSTRACT Previous study on human bone marrow culture showed that leukotrien B-4 (LTB-4) which was added to granulocyte-macrophage and erytroid colony, suppressed the growth of colonies. This study assessed the correlation between LTB-4 and marrow cellular. Marrow aspirate which was taken from 60 adult patients (26 men and 34 women) suffering from various haematopoietic problems was examined for amount of LTB-4 and the degree of marrow cellular. ELISA principle was applied for LTB-4 measurement and marrow cellular assessed by counting the member of nucleated cells per cubic-mm of marrow aspirate using microscopic counting chamber principle. Existing difference of the strength of correlation of LTB-4  and  marrow  cellular  was  analized  using  manova and  path  analysis  and  expressed  as  path  coefficient.The difference of confounding factors such as gender, age and type of diseases (haematopoitic problems) were controlled and homogenized during statistical analysis using SPSS 10 program. Path coefficient in the multi cellular marrow (n = 60) was – 0.092 (p = 0.495) and path coefficient in hypo cellular marrow (n = 28) was – 0.406 (p = 0.013). This study concludes that the increase of amount of LTB-4 in hypo cellular marrow is associated with the decrease of marrow cellular. Keywords : Leukotrien B-4, Hypocelular marraw, Apoptosis, Hematopoisis

Abstract

ABSTRACT The aim of this experiment is to study the effect of BCG vaccination on phagocyte activity and ROI secretion in cat peritoneum macrophages which infected with M.tuberculosis. The experiment used twenty four healthy cats. The animals were divided in 2 groups, 12 cats in each group. Group I were vaccinated with BCG, group II were control group which unvaccinated. BCG vaccination was done twice in two weeks interval. Two days after vaccination each cat was infected by 105 cfu M.tuberculosis intraperitoneally. The activity of macrophages were measured  at 1st, 2nd, 12th, and 24th, after infection using in vitro latex bead phagocyte and NBT reduction assay. Three cats were used to measure the macrophage activity in each period, using triplicate sample for each cat. The results of the experiment showed that the phagocyte activity and ROI secretion increased significantly in vaccination group (P<0.01) compared with the control group, and these activities reached to the plateau level at 2 weeks after infection. Although these enhanced activities were gradually diminished thereafter, higher levels of these activities were consistently observed until the end of experiment compared with control group.
