Abstract

Carotenoids are one of pigment which needed for development of post-larvae tiger shrimp (Penaeus monodon), but the shrimp unable to synthesis carotenoids by themselves in the body so it takes a feed containing carotenoids. Carotenoids can be obtained from synthetic feed or natural feed. One of the natural feed is microalgae Chlorella vulgaris. β-carotene is the highest content of carotenoid in C. vulgaris, this content can be improved by protoplast fusion. The purpose of this study was to determine the amount of β-carotene in fusan intraspecies C. vulgaris and it’s application as supplement in the post-larvae stadia 10th tiger shrimp (P. monodon) Measurement of the amount of  β-carotene using UV-Vis at a wavelength of 460nm. The feeding is done three times a day. The results showed that the amount of β-carotene on fusan C. vulgaris is greater than the wildtype of C. vulgaris. β-carotene in the fusan C. vulgaris is14.88µg/g while the wildtype of C. vulgaris is 7.14 µg/g. Additional applications C. vulgaris by 10 x 104 in 0.0225 g synthetys feed post-larvae stadia 10th shrimp (P. monodon) compared to the post-larvae shrimp fed only control showed that the feed given additional fusan of C. vulgaris increases the weight of shrimp at 39.9%, while the given wildtype of C. vulgaris increasing shrimp weight by 10.1%. Keywords: β-carotene, Chlorella vulgaris, protoplast fusion, post larvae shrimp

Abstract

Peat soil is present in many areas of Indonesia, one of them is in Sampit regency Central Kalimantan. Peat soil is a type of soil which has an extreme characteristic  of pH  around 3-5. However, it is presumed that there is bacteria that can produce growth hormones, such as  IAA. This study aimed to determine the ability of bacterial  isolated from peat garden and burned area in Sampit regency which could  produce IAA. Several tests were carried out including bacterial isolation, purification isolates, growth at different pH conditions, as well as IAA production with the addition of L-Tryptophan precursors with different concentrations. The  growth and concentration of IAA was measured  using spectrophotometry with wavelength 600 nm to 530 nm for growth and IAA, respectively. Results showed that  five isolates produced IAA, three isolates from peat garden area and two isolates from burned areas. All isolates showed the highest growth at pH 6, and the highest IAA production  at  43.311 ppm was produced by TGK6  isolate with the addition of 300 ppm L-Tryptophan. Keywords: Indole Acetic Acid, Peat Soil, L-Tryptophan

Abstract

Molds are widely distributed in nature, they even present in extreme environment, such as hot and dry soil. Molds which can grow in extreme environment has been adapted to xeric environment by producing enzymes  with  special  characteristics.  The  aim  of  this  study  to  determine  the  diversity  of  molds  from A.americana L. rhizosphere at West Sukolilo Village, Labang district, Bangkalan Regency, Madura and also to examined their cellulolytic and proteolytic activities. Isolation of molds were done by using spread and dilution methods. Molds identification were done by macroscopic and microscopic examined. The molds diversity was calculated using Shannon-Wiener index. Semiquantitative enzyme examination were done using Carboxymethil Cellulose (CMC) agar for cellulolytic ang liquid gelatin 12% for proteolytic. Results of this study showed that mold diversity isolated from   A. americana L. rhizosphere was moderate (1,8-2,7) with total of 43 species, comes from 7 genus i.e. Aspergillus, Penicillium, Fusarium, Aureobasidium, Byssochlamys, Paecilomyces, and Trichoderma.  The  highest  index  of  cellulolytic  produced  by  Aspergillus  sydowii  (2,35),  while  highest proteolytic activity produced by Aspergillus flavus (86%). Keywords : molds, diversity index, xeric, cellulolytic, proteolytic

Abstract

Inulin is an abundant compound in the nature after starch. Hydrolytic enzyme called inulinase is able to catalyse hydrolysis reaction of inulin polysaccharides into fructose and fructooligosaccharide (FOS). Yeast isolateDUCC Y-015 is one of the microbes that is able to produceinulinase. The action of inulinase enzyme associated with the growth of the isolate. Specific growth rate is one of the measurement of growth parameters; itis defined as the increase in cell biomass per unit time in the growth phase of the microbes. This study aims to examine theinulinase production and specific growth rate of yeast isolate DUCC Y-015 on the medium with various concentrations of dahlia tuber flour. The measurement of growth was done by weighing the cell dry weight and inulinase activity test was done by calculating the reduction sugar using DNS method. The used concentrations of dahlia tuber flourin the medium were 2%, 3%, 4% and 5%. Inulinase activity in each treatment was 0,111 IU/ml, 0,147 IU/ml, 0,113 IU/ml and 0,119 IU/ml, respectively, whereas the specific growth rate was 0,0062 hour-1, 0,0081 hour-1, 0,0077 hour-1, 0,0052 hour-1, respectively. The resultswere tested using the Mann-Whitney Test and theKruskal-Wallis Test. According to the results, the most optimuminulinase production and specific growth rate (µ) of yeast isolate DUCC Y-015 on dahlia tuber flour medium was treated with 3% concentration of dahlia tuber flour. Keywords: Dahlia Tuber Flour, Yeast Isolate DUCC Y-015, Specific Growth Rate, Inulinase Activity
