Evaluation of suitability of plant species for lowland forest landscape restoration in Lepan watersheds, Langkat District, North Sumatra, Indonesia. Biodiversitas 20: 2903-2909. Lepan watersheds have high management priority in North Sumatra due to healthy biophysical, socio-economic conditions and issues. Many human activities, such as conversion of forest land into settlements and oil palm plantations have resulted in decreasing the ecological functions of the forest landscape. Therefore, it is necessary to initiate efforts and actions to preserve the environment in order to prevent further damage. The effort required is to restore the already damaged ecosystem with the cultivation of species of plants in accordance with the land and soil conditions of the Lepan watersheds. Four criteria are used for determining suitable plants that can be planted as well as cultivated by the community. They are plant species found in natural forests, plant species meeting the physical and soil characteristics, plant species cultivated by community and plant species based on community perceptions. Species matching was determined by scoring and weighting method. This study found that the most suitable species for restoration activities based on these four criteria is Pterospermum subpeltatum.

Catharanthus roseus (L.) is an important medicinal plant of the family Apocynaceae used to treat many of the diseases. C. roseus also possess good antioxidant potential. The purpose of this research was 1) to determine the variability of C. roseus based on the morphological, anatomical characters and chlorophyll contents, 2) to know the differences in morphological, anatomical characteristics and chlorophyll contents of C. roseus found in Banyumas Regency. The research used a survey method, with a purposive sampling technique. Preparation of leaf for anatomical analysis was based on paraffin method. The results showed that C. roseus in Banyumas Regency were grouped into eight varieties. The morphological characters observed consisted of 21 characters, 8 quantitative characters, and 13 qualitative characters. Most of the quantitative morphological and anatomical characters were significantly diverse (p <0.05) except for petiole length, corolla number, stomata width and number of trichomata. The eight varieties of C. roseus have different chlorophyll a, chlorophyll b, and total chlorophyll contents but their anthocyanin contents were not significantly different.

Prediction of genetic gain in Ficus variegata Blume progeny trial based on breeding values. The aim of this study was to estimate the breeding value based on components variance and genetic gain prediction of Ficus variegata Blume using REML/BLUP. A total of 17 families of Ficus variegata Blume were evaluated at three years after planting at progeny trial in Mangunan, Bantul, Yogyakarta laid out in Randomized Completely Block Design (RCBD). Estimation of variance family (height and diameter) were ± 5% of total variance. Narrow-sense individual heritability was moderate (0.27 and 0.21 respectively) indicated promising genetic improvement. Strong genetic correlation of height and diameter (rG=0.9) indicated that improving a trait will improving others. BLUP analysis for diameter trait showed that the best individual value from best family selection was appropriated with restriction 10 family and 5 individual per family and genetic gain prediction was 15.83% gave acceptable rate of inbreeding 2% per generation.

Abstract. Saab R, Siregar PG, Tatang Mitra Setia TM. 2019. Implementation of animal welfare in tiger sanctuary, Barumun Nagari Wildlife Sanctuary, North Sumatra, Indonesia. Biodiversitas 20: 2790-2795. The accelerated growth in the development of economic and social infrastructure has drastically diminished the population of Sumatran tiger (Panthera tigris sumatrae) and its habitat. As a result, conflicts between tigers and humans are increasing rapidly in recent years. The wild tigers in conflict, such as those that attack humans and livestock, are usually killed or captured, while those who cannot return to the forest due to injury or illness are then translocated to the zoo. The increasing number of human and tiger conflicts raises the need for special facilities, such as sanctuaries, to recover the condition of the tigers, before they are released to the wild. This study aimed to examine the aspects of welfare management and assess the level of welfare of the tigers kept in the sanctuary at the Barumun Nagari Wildlife Sanctuary (BNWS) in North Sumatra, Indonesia. The study was carried out from March to April 2019, while data collection was carried out by literature studies, field observation on implementation of five animal welfare parameters, and self-assessment by the manager of the sanctuary. The results showed that, on a score from 1 to 5, the animal welfare parameter scores were 4.88 (freedom from hunger and thirst), 4.81 (freedom to express natural behavior), 4.79 (freedom from discomfort), 3.81 (freedom from fear and distress) and 3.69 (freedom from pain, injuries, and diseases). The total achievement implementation value of animal welfare in BNWS was 89.13, or very good. However, some improvements still have to be made by the sanctuary manager to enhance the animal welfare of tigers in BNWS.

Biodiversitas 20: 2781-2789. Ciletuh Geopark has high biodiversity but has not been supported by local knowledge about the use of animals to support sustainable development. This study aims to investigate: (i) the number of species that are used, (ii) distribution of local knowledge about animals using, and (iii) the dependency of communities on creatures in Ciletuh Geopark, Ciemas Subdistrict, Sukabumi. Mixed methods of qualitative (semi-structured interviews) and quantitative methods (proportional random sampling) were applied in this study. The community uses 153 species, consisting 58 Avifauna species, 13 mammals species, nine Herpetofauna species, three insects species, 16 species of freshwater fish, 43 species of seawater fish, two Mollusca species, and nine crustacean species. Utilization of fauna is not distributed in each village. Animals using of Avifauna, Herpetofauna, insects, and mammals tend to be utilized by people of Girimukti Village. Seawater fish, Mollusca, and crustaceans tend to be utilized by Ciwaru Village, while freshwater fish tend to be used in each village, except Girimukti and Cibenda Village. Utilization of fauna in Ciemas Subdistrict has begun to be abandoned, although several people still use them.

Seafood is very popular among Indonesian people, especially in coastal areas. In Bantul Yogyakarta, blood clams have become one of tourist's favorite, either cooked or raw. Blood clams are filter feeders that cause the clams to be vulnerable to contamination of pathogenic bacteria that can cause foodborne disease, including Escherichia, Pseudomonas, and Vibrio. The 10-20% cases of foodborne disease transmitted through seafood caused by Vibrio spp. Three species of Vibrio can cause foodborne disease in humans, i.e., V. cholerae, V. parahaemolyticus, and V. vulnificus. The purpose of this study was to determine the character of Vibrio using morphological and molecular identification and pathogenicity on blood clam (Anadara granosa). Blood clams samples were collected from Depok, Goa Cemara, and Kwaru beaches in Bantul, Yogyakarta, Indonesia. Isolation of Vibrio spp. from blood clams was done using selective differential Thiosulfate Citrate bile salts sucrose (TCBS) culture medium. The morphological characters of the isolate colonies were determined based on the color, shape, texture, and size of the colony. Hemolysis test was also performed to evaluate the pathogenicity by using blood agar media. Molecular identification of Vibrio species was made using 16S rRNA sequence. Phylogenetic analysis was performed using Neighbor-Joining method in Mega X software. Samples for the analysis came from DNA sequences of this study and those from the GenBank database. Of the total 15 isolates obtained, four isolates showed positive ?-hemolysis, namely, isolate P2S2 -1bH, P3S1-1aH, P2S1-1aK and P2S2-1aK, and one isolate had positive ?-hemolysis (P3S2-1aK). Seven species of Vibrio were identified as V. algynolyticus, V. parahaemolyticus, V. diabolicus, V neocaledonicus, V. azureus, V. natrigens, and V. cholerae.

Biodiversitas 20: 2891-2899. Green mussels (Perna viridis) have filter feeder properties that allow pathogenic bacteria from the water environment to accumulate in relatively high levels. About 20% of foodborne diseases are caused by large quantities of seafood contaminated with bacteria. The purpose of this study is to determine the morphological characteristics, pathogenicity, identity, and the kinship of Vibrio species on green mussels in Yogyakarta coastal tourism areas. Vibrio spp. were grown on selective differential TCBS media. In this media, the suspected Vibrio spp. would produce yellow or green colonies. The ability of hemolysis of Vibrio was blood agar media, the species was molecularly identified using 16S rRNA gene sequence, and the phylogenetic relationship of the Vibrio spp., was analyzed using MEGA X Neighbor-Joining program. Based on morphological analysis, we obtained 23 bacterial isolates suspected to be Vibrio spp. Two Isolates (L1K2 6 and L2K2 13) were positive for ?-hemolysis activity and 4 isolates (L1K1 3, L2K1 8, L2K2 16, and L3K2 22) were positive for ?-hemolysis activity. The molecular analysis involved 18 Vibrio species, and 4 of them represented the Vibrio genus and 14 species represented 97-99% similarity species in accordance with the 16S rRNA sequence in database, namely: Vibrio alginolyticus, Vibrio parahaemolyticus, Vibrio cholerae, Vibrio neocaledonicus, Vibrio mimicus, Vibrio azureus, Vibrio diabolicus, Vibrio tapetis, Vibrio natriegens, and Vibrio owensii. The most dominant number of Vibrio isolates was V. alginolyticus, while the lowest was V. owensii. The highest number of Vibrio species in green mussels was found in Goa Cemara beach while the lowest was in Kwaru beach. Vibrio spp bacteria found in green clams in coastal tourism areas in Yogyakarta have close phylogenetic relationships with other Vibrio in seafood in Indonesian coastal waters.

Short Communication: Intraspecific genetic diversity and population subdivision of rabbitfish (Siganidae: Siganus canaliculatus) in urbanized reefs of Jakarta Bay, Indonesia. Biodiversitas 20: xxxx. Indonesian coral reefs and seagrass are heavily influenced by human activities through pollution and habitat loss, and sea-level rise or the increase of ocean temperature due to the global change. In the Jakarta Bay and Seribu Islands, marine communities have been impacted by an increase in eutrophication and sedimentation levels. As a result, these habitats became degraded and could affect their associated organisms such as rabbitfish. Genetic diversity and population structure can provide information about a long-term condition in an aquatic organism. The study was conducted to assess genetic diversity and population structure of rabbitfish (Siganus canaliculatus) across an environmental gradient in Jakarta Bay and Seribu Islands. A total of 31 individuals were collected using gillnet in study sites. A molecular marker mitochondrial Cytochrome oxidase 1 (CO1) was used to amplify DNA. The result of the phylogenetic tree formed two major clades, which generally separate northern and southern region of Seribu Islands populations. AMOVA analysis showed significant genetic differentiation between the population of Siganus canaliculatus in the north zone (National Park) and south zone Seribu Island (FST = 0.38, p<0.005). The highest haplotype diversity (Hd) of Siganus canaliculatus contained was observed in the north (Pulau Semak Daun) and south (Pulau Damar) in both as moderate (0.50). Nucleotide diversity ( ) of Siganus canaliculatus was categorized as low (0.076) to moderate (0.500). Environment and humans seem to affect the value of genetic diversity, and as the study suggests, a significant population subdivision makes this population vulnerable.

Abstract. Situmorang ROP, Kuswanda W. 2019. Network mapping in the development of elephant conservation center in North Sumatra, Indonesia. Biodiversitas 20: 2858-2867. A project in the Aek Nauli Elephant Conservation Camp (ANECC) is being carried out to conserve Sumatran elephant handled by several institutions within the Ministry of Environmental and Forestry of Republic of Indonesia and other supporting stakeholders. A social networking analysis (SNA) had been carried out to analyze the social interactions of the stakeholders in the development of the ANECC. This research aimed at finding out the important actors in the network and describing at the social network gap in the development of ANECC objectives as conservation, and research, and ecotourism center. From the research, we found four institutions engaged in conservation, research, and animal health surveillance as the core actors and 19 other institutions as the periphery actors consisting of authorities, local government, businessmen, and local people. The study also found that the Environmental and Forestry Research and Development Institute Aek Nauli is the most influential actor in the network, and the National Planning Agency and Girsang Sipanganbolon Sub-district as the bridging actors who connect the supporting actors to the core actors. Conservation aspect is the most communicated in the network, followed by ecotourism, and research. Our research also found that conservation and research aspects have been handled by the appropriate institutions, however, ecotourism has not been handled by certain agencies responsible for the development of ecotourism. Hence, the study suggests that the core actors can consider binding cooperation with potential entrepreneurs on tourism around the site to develop ecotourism sector in ANECC.

Biodiversitas 20: 3005-3010. Sugarcane (Saccharum officinarum L.) belongs to the Poaceae family which are widely used for sweeteners production. In Indonesia, the need for sugarcane continues to increase along with its increased use in the production of bioethanol from molasses, a by-product of sugar production. However, the supply for sugar is still far from enough. Improvement of sugarcane productivity can be done by selecting superior varieties to form a new superior hybrid. Maintaining high genetic diversity in a population is essential. Selection of varieties is important because it can increase productivity without increasing the cost of crop production. Even the selected pest-resistant varieties can reduce the cost of crop production. Indonesia had produced many superior sugarcane plant varieties, but studies on their characters and genetic relationship are still lacking. Several molecular markers have been used to elucidate the genetic diversity among sugarcane varieties. Random Amplified Polymorphism DNA (RAPD) is one of the molecular markers that have been proven to describe genetic diversity among sugarcane plants. This study aimed to determine the genetic diversity among 22 superior sugarcane varieties in Indonesia based on eight RAPD molecular markers. The superior sugarcane varieties in Indonesia in this study were obtained from Indonesian Sweetener and Fiber Crops Research Institute (ISFRI), Malang and Madukismo Sugar Factory, Yogyakarta. DNA products were visualized by using gel electrophoresis and resolved bands were scored as binary data as presence (1) and absence (0). Linkage distance was done based on Jaccard’s similarity coefficient, and clustering analysis was performed based on the unweighted pair- group average (UPGMA) method. Polymorphism Information Content (PIC) analysis showed a high degree of polymorphism ranging from 0.76 to 0.92. Cluster analysis showed that 22 sugarcane varieties were divided into two main clusters and four sub-clusters based on their origin.
