Estimating carbon stocks of three traditional agroforestry systems and their relationships with tree diversity and stand density. Biodiversitas 23: 6137-6146. One of the important sectors as a strategy for climate resilience and low carbon development in Indonesia is Forestry and Other Land Uses (FOLU) including the agroforestry sector. Carbon stock and tree diversity in an ecosystem are dynamic over time and still need to be investigated. This study was aimed: i) to estimate aboveground carbon stocks in the traditional agroforestry of Mount Halimun Salak National Park (MHSNP), in Kampung Birang and Kampung Merabu, East Kalimantan, as well as ii) to analyze the correlation between carbon stock with tree diversity and stand density in traditional agroforestry of Mount Halimun Salak National Park (MHSNP), in Kampung Birang and Kampung Merabu, East Kalimantan. The total plots were 75 plots (a plot size of 50x50 m), and carbon stock estimation was focused on pole and tree levels. The average carbon stock in the traditional zone of MHSNP, Kampung Birang, and Kampung Merabu respectively was 51.02 Mg C/ha, 96.25 Mg C/ha, and 82.17 Mg C/ha. The carbon stock in traditional agroforestry of all study sites was very significantly affected by the basal area (m2/ha) (p-val<0.05) with a strong correlation (r = 80% and 97%). The carbon stock was affected by stand density (p-val<0.1) with a very low correlation (r = 27.3%). The carbon stock was very significantly affected by total species (p-val<0.05). However, it had a weak correlation (r = 28.0%). In agroforestry systems, how to manage the agroforestry, socio-economic aspects, ecological site, and market are the main factor in species selection and species maintenance by landowners that indirectly influence biodiversity conservation effort and carbon stock value. The agroforestry system has the potential to become a target and priority location for Indonesia’s FOLU Net Sink 2030 by providing non-timber forest products, which can avoid leakage in forest conservation efforts and carbon stock.

Biodiversitas 23: 6397-6403. The high demand for spiny lobster seeds has placed intense pressure on the lobsters’ wild stock in Indonesia. To address this issue, a hatchery was developed to produce scalloped spiny lobster larvae. However, the dietary requirements after the yolk sac has been depleted have proven to be a significant challenge. Thus, the present study aimed to identify the potential live diet of scalloped spiny lobster larvae by identifying the stomach contents of lobster larvae captured in the wild. Fifteen scalloped spiny lobsters at the post-larval stage were collected from three fishing grounds: Gerupuk Bay (Lombok Island), Tawang Bay (East Java), and Prigi Bay (East Java), Indonesia. The stomach of each scalloped spiny lobster was dissected under a dissecting microscope and its contents were observed under a binocular microscope for plankton identification and abundance. The stomach contents of scalloped spiny lobster resulted in five identified plankton species collected from Prigi Bay [Tintinnopsis sp. (37.5%), Grammatophora sp. (25%), Synedra sp. (18.8%), Phormidium sp. (4.3%), and Rhizisolenia sp. (4.3%)]; six plankton species collected from Tawang Bay [Ochromonas sp. (32.3%), Synedra sp. (20.6%), Tintinnopsis sp. (14.7%), Uronema sp. (14.7%), Coscinodiscuss sp. (2.9%), and Planktoniella sp. (2.9%)]; and six plankton species collected from Gerupuk Bay [Synedra sp. (33.3%), Chlorococcum sp. (33.0%), Phormidium sp. (13.3%), Gymnidinium sp. (6.6%), unidentified Cirripedia (3.3%), Rhizosolenia sp. (3.0%)]. Of these plankton, Synedra sp. and Rhizolenia sp. were the most common representatives in the stomach of all lobster samples. Thus, assessing these two plankton genera is highly recommended for future studies.

The antioxidant activities and chemical compounds of Aquilaria crassna, Aquilaria microcarpa, and Gyrinops versteegii leaves growing in Langkat, North Sumatra, Indonesia. Biodiversitas 23: 6619-6628. Agarwood has been cultivated for its wood and leaves in Indonesia. This research aims to determine the potential of the leaves of Aquilaria crassna, A. microcarpa, and Gyrinops versteegii that grow in Langkat as raw materials for antioxidant-rich tea based on their chemical compounds and antioxidant activities. The leaf powder was extracted using ethanol. Bioactive compounds were analyzed by GC-MS, while the antioxidant activity was analyzed by the DPPH method. The results showed that A. crassna leaves contained alkaloids, flavonoids, tannins, steroid/triterpenoid dan glycosides, whereas A. microcarpa leaves contained alkaloids, tannins, saponins, steroid/triterpenoid dan glycosides alkaloids, tannins, and triterpenoids. In comparison, G. versteegii leaves contained flavonoids, tannins, saponins, steroid/triterpenoids, and glycosides. The IC50 values of ethanol extract of A. crassna, A. microcarpa, and G. versteegii leaves were 30.0079, 29.3263, and 31.6304 µg/mL, respectively. The highest chemical compounds of A. crassna, A. microcarpa, and G. versteegii were Cyano, -acetic acid, 1,2,3,4-Cyclopentane Petrol, and Oxacycloheptadec-8-en-2-one Ambrettolide The three species had high antioxidant activity; therefore they are potential sources of antioxidants.

A supportive growing environment and the presence of microbes in the soil are considered good habitats for plant growth. Endophytic microbes live in symbiosis (mutual benefit) with their host plants, both in the stem and root tissues of plants, providing defense services in biotic and abiotic stress situations. This study was aimed to identify various microbes that exist in various agricultural cultivation ecosystems in Central Sulawesi. Microbial sampling was obtained from healthy plant tissues (endophytes) and rhizosphere in lowland rice and shallot plantation ecosystems. The sample obtained was then weighed as much as 5 g and surface sterilized for further isolation by serial dilution method to be spread on NA (Nutrient Agar) media for bacteria, and PDA (Potato Dextrose Agar) for fungi. The microbes that had grown were re-isolated to obtain pure isolate culture of microbes. The results showed that 8 isolates from the rhizosphere and 4 isolates as endophytes in the ecosystem of organic rice fields were recorded. The fungal isolates from the shallot plantation “Lembah Palu” found as many as 12 isolates from rhizosphere and 4 isolates as endophytes. The results of macroscopic and microscopic characteristics on fungal isolates identified as the species of Trichoderma, Fusarium, Penicillium, Gliocladium and Aspergillus.

Ornamental fish is a potential commodity necessarily optimized to improve the welfare of society. One of the Indonesian provinces producing diverse species of ornamental fish to export is Southeast Sulawesi. This study aimed to evaluate the ornamental fish potentials and create an evaluation possibly implemented as a consideration to determine business development strategies. The method was a case study, collected data through observation, interviews, literature study, and internal and external conditions were analyzed using SWOT analysis. The study analyzed the potential, trade, and strategy for business development in ornamental fish in Southeast Sulawesi. The study discovers that 38 species are traded, and nine of them are dominantly traded. Moreover, there are six main species potentially cultivated due to the demand and stable production; they are Betok Kupang (Chrysietera cyanea), Capungan (Apogon sp.), Angel KKO (Cirrhilabrus sp.), Clown fish (Amphiprion ocellaris), Keling Rub (Cirrhilabrus sp.), and Letter six (Paracanthurus hepatus). The development of ornamental fish cultivation can be implemented with the S-O (strengths–opportunities) strategy that manages and protects the environment of ornamental fish, strengthens the capacity of fishers and stakeholders for environmentally friendly activities and export market orientation, and builds cooperation of fisherman, exporters, and stakeholders to develop business in ornamental fish by emphasizing local species with high economic values and market demands. These results can be a consideration in making decisions related to the development of ornamental fish.

The partial contribution of CO2-emission losses from subsidence in small-holder oil palm plantation on a tropical peatland in West Kalimantan, Indonesia. Biodiversitas 23: 6539-6545. Carbon storage in tropical peat ecosystems over thousands of years, especially within peat soil, is in huge quantity. Degradation of peat ecosystems is generally caused by human factors, whether intentional or not, damaging the carbon storage function of tropical peatlands, where forest clearing, drainage development, and burning of land converted to agriculture and plantations result in significant greenhouse gas emissions. Tropical peat in the Kubu Raya District of West Kalimantan, which has a relatively large area of peat, has been degraded as a cause of uncontrolled drainage and land fires caused by a lack of management after its forest cover was lost. The main impact is an increase in peat CO2 emissions due to changes in land use, especially lowering groundwater levels. Subsequently, the subsidence process also occurs after land clearing. This study aims to obtain the proportion of carbon biomass loss due to the CO2 emission process from reducing the peat layer due to subsidence. Data collection was executed for two years, where CO2 emission was monitored bi-weekly, and the subsidence was measured bi-monthly. The results demonstrate groundwater levels dictate the peat CO2 emission and subsidence. Lowering GWL 30 to -85 cm increases CO2 by more than three times, approximately. The rate of peat subsidence shows similar trends to the emission. The proportion of peat biomass loss on CO2 emission was between 58.9 to 73.5%, except for GWL ~5 cm, where the proportion was the highest at 82%. The results of this study are beneficial in explaining the part of the subsidence that impacts the sources of CO2 emissions from the small-holder oil palm and GWL management on peatlands.

Biodiversitas 23: 6164-6170. Domesticated dog populations have grown throughout Indonesia, notably in Bitung City. Dogs are frequently kept as home guardians and reside with their owners. Dogs are typically attacked by ectoparasites, parasites that adhere to the skin's surface and potentially spread the disease microorganisms to humans, animals, and the environment. As a specific host, ticks are frequently found in dogs. This research aims to identify and analyze the molecular character bacterial of Rhipicephalus sanguineus ticks in Bitung City. A hypervariable region of 16S rRNA called V3-V4 was selected for the DNA barcode area. Dogs' ticks have been collected from several areas in Bitung. The microbes inside the digestive tract were analyzed using genomic DNA with a gSYNC DNA extraction kit, and PCR amplification using KOD FX Neo and MyTaq HS red mix. At First Base Singapore, gel electrophoresis and sequencing were carried out. The collected sequencing data were analyzed using MEGA X and the BLAST, connected online with the NCBI's GenBank. The nine distinct bacteria were identified inside the digestive tract of ticks. Pseudomonas stutzeri (35%), Brevundimonas sp. (21%), and Aquamicrobium lusatiense (17%) were the pathogens that were most frequently found. These results enhance knowledge of the pathogen microbes that may impact either humans or animals.

Biodiversitas 23: 6227-6237. Medicinal plants have long been used by indigenous communities, including Gayo people in Aceh, Indonesia. The conservation and sustainable utilization of medicinal plants can be integrated with ecotourism activities. Kedah Rainforest Lodges is an ecotourism area in the Protected Forest area of Forest Management Unit (FMU) Region V Aceh, Indonesia, which is an important source of medicinal plants, clean water and nature tourism. However, biodiversity in the area is threatened by excessive human activity, including the long-standing tradition of Gayo people using medicinal plants. Therefore, this study aimed to investigate medicinal plants used by the local community living around Kedah Rainforest Lodges using an ethnobotanical approach and to identify a strategy for the conservation of medicinal plants using strengths, weaknesses, opportunities and threats (SWOT) analysis. A combination of field observations, interviews, documentation and focus group discussions were employed for data collection. The result of the ethnobotanical study identified 44 species belonging to 31 families of medicinal plants used by Gayo people in the studied area. Asteraceae is the family with the largest number of medicinal plant species, while herbs are the most common habitus of the medicinal plants and leaves are the most used plant part. Based on SWOT analysis, there are several strategies to conserve medicinal plants in Kedah Rainforest Lodges, including (i) the rehabilitation of degraded areas through the enrichment of medicinal plants to create living pharmacies; (i) the development of facilities to support medicinal plants ecotourism; and (iii) partnerships with related stakeholders to obtain funding and enhance the capacity of the area manager and staff.

Molecular identification of anaerobic fungi isolated from buffalo rumen with their growth rate, cellulase enzyme activity, and fermentation products characteristics. Biodiversitas 23: 6448-6455. Anaerobic fungi are essential microbes in the degradation process of feed fiber in the rumen with the ability to produce fibrolytic enzymes and rhizoids that can penetrate feed particles. The activity of cellulase enzymes in fungi is influenced by several factors such as the type of feed and livestock used as a source of isolate. Therefore, this research aimed to analyze the types of anaerobic fungi isolated from buffalo based on their DNA nitrogenous bases composition and also to evaluate the growth rate, cellulase enzyme activity, and fermentation product concentration of anaerobic fungi. The growth rate of fungi was examined by measuring the biomass formed, while cellulase enzyme activity was carried out using CMC, Avicel, and Filter Paper as substrates. The fungal fermentation products were analyzed using the HPLC and GC methods. The results showed that the fungi isolated from buffalo rumen were closely related to anaerobic fungi type Piromyces sp., Caecomyces sp., and Neocallimastix frontalis, with different growth rate, cellulase enzyme activity, and the fermentation products concentration in each type of anaerobic fungi. Therefore, it can be concluded that anaerobic fungi isolated from buffalo rumen can degrade cellulose. It was also discovered that fungi Neocallimastix frontalis had a higher growth rate, cellulase enzyme activity (CMCase, avicelase, and FPase), and fermentation products than Piromyces sp. and Caecomyces sp.

Abstract. Herliyana EN, Oktavianto P, Siregar UJ. 2022. Identification and characterization of Pestalotiopsis spp. causing leaf spot and leaf blight on jabon (Neolamarckia spp.) in Indonesia. Biodiversitas 23: 6547-6556. Jabon (Neolamarckia spp.) is one of the fast-growing trees with high economic value for industrial plantations. Its monoculture planting system is faced with certain problems, such as the leaf spot and leaf blight diseases caused by Pestalotiopsis sp. Therefore, the aim of this research was to describe the morphology of Pestalotiopsis species through macroscopic and microscopic observations, followed by the identification of fungus using DNA barcoding causing leaf spot and leaf blight on the tree. A total of 11 Pestalotiopsis isolates were used for this research. The morphology was identified by observing mycelium colonies and fungal conidia, while DNA barcoding was initiated after DNA isolation using cetyltrimethylammonium bromide (CTAB) method, amplification with ITS 1 and 4 primers, followed by sequencing process. The results of DNA barcoding showed that 5 species of Pestalotiopsis, namely Pseudopestalotiopsis theae, Pestalotiopsis microspora, Pestalotiopsis palmarum, Neopestalotiopsis clavispora, and Pestalotiopsis virgatula were found associated with leaf spot and blight disease. All the five species were further classified into 3 clades using phylogeny according to their median cells and apical appendage shape. The clade X consisted of P. palmarum, N. clavispora, and P. virgatula with versicolorous median cells, whereas clade Y included P. microspora with concolorous median cell, and clade Z consisted Ps. theae with concolorous median cell and knobbed apical appendage.
