Abstract. Zulhalifah, Syukur A, Santoso D, Karnan. 2021. Species diversity and composition, and above-ground carbon of mangrove vegetation in Jor Bay, East Lombok, Indonesia. Biodiversitas 22: 2066-2071. Mangroves play a very important role to mitigate global warming. This study aimed to assess the species diversity and composition as well as the above-ground biomass and carbon content of mangroves in Jor Bay (Teluk Jor), Lombok Island, Indonesia. A purposive sampling method was implemented using transects and square plots as data collection techniques. The data was analyzed to reveal the frequency, density, dominance, above-ground biomass, and carbon content. The results of this study found seven mangrove species, namely Avicennia marina, Ceriops tagal, Rhizophora apiculata, Rhizophora stylosa, Sonneratia alba, Sonneratia caseolaris, and Lumnitzera racemosa. In general, Sonneratia alba was the most important species in terms of frequency, density, and dominance followed by Rhizophora apiculata. In total, mangrove vegetation in Jor Bay stored 697.45 ton C/ha, or equivalent to carbon monoxide uptake of 2559.63 CO2/ ha. In accordance with species composition, Sonneratia alba had the largest contribution to total carbon content with 453.76 tons C/ha), followed by Rhizophora apiculata with 74.47 tons C/ha. These findings suggest that mangrove vegetation in Jor Bay stored large amount of carbon in the form of above-ground biomass, implying the importance to preserve this area for carbon conservation.

Abstract. Chapagain N, Pandey N, Chitrakar P. 2021. Ichthyofaunal diversity in snow-fed Mardi Stream of Gandaki River, Central Nepal. Biodiversitas 22: 2940-2948. This study assessed site-wise and season-wise diversity and investigated the potential environmental correlates of ichthyofaunal assemblage in snow-fed Mardi Stream of central Nepal by extensive fieldworks from June 2018 to April 2019. The ichthyofaunal diversity and community structure in the hill streams of the Nepal Himalaya are understudied. A total of 1850 individual fish belonging to 3 orders, 3 families, 9 genera, and 10 species were collected. Cypriniformes emerged as the most species-rich order, accounting for 92.6% of the total species. Based on Similarity Percentage (SIMPER) analysis, the major contributing species were Schizothorax richardsonii (27.58%), Garra annandalei (26.3%), and Naziritor chelynoides (10.82%). Diversity indices were found higher in summer season and downstream areas. Results from Generalized Linear Model (GLM) and Canonical Correspondence Analysis (CCA) revealed that pH, Dissolved Oxygen (DO), water temperature and water velocity had significant relationship with fish assemblages. The fish assemblages along the gradient of the stream may be interrupted due to climate factors and anthropogenic disturbances. This study provides baseline data on hill stream fishes of Mardi Stream which might be helpful for implementation of conservation approaches.

Abstract. Hammadi H, Hammouda-Bousbia D, Chaib G, Tir A. 2021. Genetic diversity in several genotypes of Algerian lentil using biochemical markers. Biodiversitas 22: 3494-2500. Studying seed storage proteins in legumes (Fabaceae) is very important besides its economic value; seed storage proteins play an important role in plant taxonomy because of the stability of their contents. The grain protein content and protein profiles of 12 lentil genotypes (Lens culinaris Medik.) were characterized based on protein profiling through SDS-PAGE. The obtained gel has shown forty bands ranged from 13 to 21 with molecular weight 10 to 120 KDa. Of which six monomorphic bands, thirty-four polymorphic bands and five unique bands in Flip90-31, Idlep3, idlep2, Idlep4 and Radjas genotypes. The protein banding pattern polymorphism (84%) revealed an inter-genotypic variability. The Idlep3 genotype is characterized by specific bands (or a specific marker), followed by the Flip90-31 genotype. So these two genotypes can be considered as valuable gene resources for further breeding programs.

Abstract. Masri M, Sukmawaty E, Aditia L. 2021. Novel chitinolytic bacteria from the shrimp shell processing waste. Biodiversitas 22: 2672-2681. Chitinase is an enzyme that hydrolyzes chitin (?-1,4 Nsetyl-D-glucosamine polymer). Several types of microorganisms produce chitinase that contributes to basic science and application. The role of chitinase enzyme is widely utilized in various fields, such as health as an anti-tumor agent and agriculture as biological control agents in plant disease caused by fungus. The purpose of this study was to isolate novel chitinolytic bacteria from shrimp shell waste in Makassar, Indonesia, using the scatter plate method on chitin agar selective media incubated at 37°C for 3 days. The clear zone formation around the colony was marked as an indicator of chitinase activity performed by the certain isolate. Selection was carried out by recycling all isolates on chitin agar media, which were incubated at 37°C for 3 days and measured the chitinolytic index. Two of ten isolates produced the highest chitinolytic index, namely, KLA-2 (45) and KLA-4 (15) isolates. The molecular identification results showed that the KLA-2 and KLA-4 isolates were Lysinibacillus fusiformis strain WH22 and Brevibacillus reuszeri strain CMB-15, respectively. Therefore, novel chitinolytic bacteria that can produce chitinase enzyme from shrimp shell waste are L. fusiformis and B. reuszeri

Abstract

Traditionally, the Sundanese village people in West Java, to treat various common diseases, use traditional medicines made from various plants. The purpose of this study was to elucidate the knowledge of the Sundanese village people on various diseases, species of medicinal plants and parts of plants used to treat various diseases, sources of medicinal plant species, methods of processing and utilization of medicinal plant species, and source of knowledge on processing and utilization of medicinal plants. The method used in this research was mixed-methods, a mixture of qualitative and quantitative. Data collection techniques were carried out using semi-structured interviews with informants, structured interviews with respondents, and field observation. The results showed that there were 22 common diseases suffered by the people of Cibeurih Hamlet, Nagarawangi Village; 36 species of plants were recorded with the most dominant part of the leaf organ being used as medicine by the community; source of medicinal plants species mainly collected from the home garden; as well as the method of processing medicinal plants most often used by the community by boiling them, this knowledge is passed on from generation through parents.

Orchid species, including Dendrobium spp., originated from Bali are threatened to extinction due to habitat destruction, illegal hunting, small population size, and the effects of global environmental change. Conservation strategies are therefore required to preserve the remaining germplasm, one of which is through in vitro seed propagation. This research aimed to investigate the pollination compatibility (i.e., pollination that produce fruits) of Dendrobium orchids originated from Bali, and to determine the effects of adding organic matters (i.e., coconut water, tomato extract, and peptone) to the base media on orchid seed germination under in vitro culture condition. Self-pollination and sibling pollination were implemented to seven orchid species, namely Dendrobium macrophyllum, D. heterocarpum, D. fimbriatum, D. linearifolium, D. spathilingue, D. secundum, and D. plicatile. The results showed that only four species, i.e., D. macrophyllum, D. heterocarpum, D. fimbriatum, and D. linearifolium, that produced fruits after being pollinated. The subsequent in vitro propagation of seeds produced by the four species showed that the addition of organic matters of tomato extract, coconut water, and peptone significantly affected the germination of Dendrobium forma Bali orchid seeds, indicated by different germination rates. The addition of 20% coconut water to Vacin and Went (VW) media gave the highest percentages of seed germination and protocorm formation for D. macrophyllum, D. heterocarpum, and D. fimbriatum at 98.33% and 95.00%; 91.67% and 91.67%; and 95% and 98.33%, respectively. Our findings can serve as baseline information when developing conservation strategies of Dendrobium orchids from Bali, particularly from the aspect of propagation

Abstract. Damayanti I, Nurbambang A, Soeprobowati TR. 2021. Plant diversity of Petungkriyono Forest of Dieng Plateau, Central Java, Indonesia. Biodiversitas 22: 3497-3507. Petungkriyono is one of the remaining forests considered as the key biodiversity site in Central Java, Indonesia. The study aimed to analyze the composition, structure, and diversity of flora. The research used 60 circular plots with 0.04 hectares on five sites: Tlogopakis, Kasimpar, Yosorejo, Kayupuring, and Tlogohendro. The result indicated that the forest was dominated by 54.6% mixed forests, 29.1% pine forests, 10.7% puspa forests, and 5.6% unproductive areas. A total of 979 individuals belong to 108 species, among 88 genera and 43 families. The dominant family was Moraceae (32.6%) and has 10 species of Ficus spp as a key pioneer in a dry habitat. According to the Red List, the International Union for Conservation of Nature (IUCN), there were 4 species categories of endangered species (EN), 11 species as vulnerable category (VU), 46 species as least concern (LC), and 45 species not evaluated (NE). The horizontal and vertical structure of trees was dominated by 10-20 cm diameter classes and 82% at a medium level of the Shannon-Weiner index (H’). The conclusion is that plant diversity plays a significant role to maintain ecosystem stability, therefore conservation activities have to be developed properly. Ecosystem services are produced in the social-ecological system to increase human well-being and resilience.

Abstract. Yusuf RA, Tank UM, Karnila R, Pato U. 2021. Biological and management design for sustainable wetland rice farming in Siak District, Riau, Indonesia. Biodiversitas 22: 2803-2814. This study aimed to develop a management design for sustainable wetland rice farming to promote food security in Siak District, Riau Province. This study was conducted through survey research wherein primary and secondary data were used. In the sustainability analysis, the Rapfish method was used which is based on eight attributes that are a combination of key factors in the sustainability analysis and stakeholder needs analysis. Results showed that the management design that can be implemented to improve the sustainability of rice farming involves preventing the continuous paddy field conversion and thus promote their preservation, improving the farmers’ knowledge and skills to create superior human resources, and strengthening agribusiness-based farmer institutions to improve farmers’ welfare. The multidimensional sustainability value obtained in three scenarios was within the 50–75% range, indicating a fairly sustainable status. For this reason, it is recommended to apply scenario 1 wherein sustainability can be increased at low costs and within a relatively short time

Abstract. Maharijaya A, Syukur M, Salma LN, Sanubary UL. 2021. Diversity and performance of eight new promising potato (Solanum tuberosum L.) genotypes in Garut District, West Java, Indonesia. Biodiversitas 22: 2848-2858. Potato (Solanum tuberosum L.) is one of the main crops in the world. Potato consumption in Indonesia continues to increase, but the supply of potatoes cannot be fully met by domestic production. The low productivity of potatoes is due to the low quality. Efforts to increase potato production are by developing new varieties with higher yields. This study aimed to obtain information about the diversity and performance of eight IPB new potato genotypes in Garut District, West Java. The study used a randomized complete block design with a one-factor genotype and four replications. The characters observed included 46 characters consisting of 27 quantitative characters and 19 qualitative characters.The genotypes that was used as materials are PKHT-2019-010, PKHT-2019-011, PKHT-2019-012, PKHT-2019-013, PKHT-2019-014, PKHT-2019-015, PKHT-2019-016, PKHT-2019-017 and two varieties Medians and Intas as control. The result showed that the potato genotypes have a diversity of leaves, stems, flowers, and tubers. PKHT-2019-010, PKHT-2019-010, PKHT-2019-015, and PKHT-2019-017 have higher productivity than Medians and Intan as control. PKHT-2019-012 productivity was not significantly different from Intan as control. The genotypes that had a higher or were the same productivity as the control varieties might be the expected genotypes that can be developed as new varieties. The genotypes expected to be developed are PKHT-2019-015, PKHT-2019-010, PKHT-2019-017, and PKHT-2019-012

Abstract. Nisa C, Jadid N. 2021. Exogenous acetic acid pre-treatment increases drought tolerance of two Indonesian foxtail millet (Setaria italica) accessions. Biodiversitas 22: 2117-2124. Drought is one of the external factors that affect the productivity of food crops. Some biotechnological approaches had been developed to increase plant resistance to drought stress. Moreover, an adjustment in plant cultivation system is also reported as alternative way to enhance plant resistance to water deficit conditions, especially during their initial vegetative phase. This study aimed to determine the effect of exogenous acetic acid pre-treatment on some Indonesian foxtail millet (Setaria italica) accessions under drought stress conditions. Two different foxtail millet accessions were used in this study: Gambir Manis and Polman Kuning. Morphological and physiological parameters were observed during this study, including plant height, number of leaves, root length, relative water content, total chlorophyll and carotenoid content, and panicle weight. Our results showed that all foxtail millet accessions survived drought stress condition after being pre-treated with exogenous acetic acid. Yet, some morphological parameters were significantly affected. Meanwhile, physiological parameters were not significantly affected. Overall, our data suggest that exogenous acetic acid pre-treatment could enhance drought avoidance in S. italica accessions. This might be due to acetic acid-induced jasmonic acid modulation and secondary metabolites production in S. italica during water deficit exposure.
