Abstract

The aims of the study were to: Investigate polymorphisms in the ND-5 region of bovine mitochondrial DNA in the composite and purebred Hereford herds from the Wokalup selection experiment, sequencing and compare the sequences between haplotypes and published sequence from Genebank. A total of 194 Hereford and 235 composite breed cattle from Wokalup Research Station were used in this study. The mitochondrial DNA was extracted using Wizard genomic DNA purification system from Promega. ND-5 fragment of mitochondrial DNA was amplified using PCR and continued with RFLP. Each haplotypes were sequenced. PCR products of each haplotype were cloned into pCR II, transformed, colonies selection, plasmid DNA extraction continued with cycle sequencing. Polymorphisms were found in both breeds of cattle in ND-5 region of mitochondrial DNA by PCR-RFLP analysis. Abstract

“Wader” fish is one of the local Indonesian wild fish that not cultured yet. This fish have amount of species, included Genus Rasbora. The aim of this research was to known the “wader” fish diversity of Genus Rasbora that potentially to aquaculture. The research conducted in 10 weeks to three species, namely R. lateristriata, R. aprotaenia, and R. argyrotaenia. The research was designed in two treatments, namely giving pellet and non-giving pellet. The growth data was measured every week. The result indicated that pellet could increase size and growth of the fish. R. argyrotaenia had bigger size and faster growth than R. lateristriata and R. aprotaenia. The optimum growth of R. argyrotaenia occured at third to sixth weeks, and it got maximum size at seventh week. Erniwati, Zuhud E, Santosa Y, Anas I. 2016. The value of secondary forest patches for bird conservation in palm oil landscapes of Riau, Sumatra. Biodiversitas 17: 791-798. Land use change due to palm oil expansion is considered to be one of the key drivers of biodiversity loss in the tropics, particularly in Indonesia, which is the biggest producer of palm oil in the world. In the last three decades, large scale plantations and smallholdings of palm oil have come to dominate the agricultural landscape, leaving small secondary forest patches surrounded by plantations. We currently have only limited current knowledge about the value of secondary forest patches for bird conservation in the palm oil landscape. The aim of this study was to contribute to our understanding of the value of remnant forest patches, smallholdings, and large scale plantations for bird conservation in the palm oil plantation landscape. We also examined the influence of the age of the palm oil plantations on bird diversity. We conducted the survey from March to April 2016. We surveyed 40 line transects in palm oil landscape in Riau Province, eight transects in secondary forest patches, 16 transects in smallholdings and 16 in a large scale plantation. Seventy three bird species, 41families and 1579 individuals were recorded; 16 species being protected in all sites. Our result showed that secondary forest has higher bird diversity than the palm oil plantations; large scale plantation support higher bird species abundance than smallholdings, while old age stands (>19 year) have higher species abundance within large scalepalm oil plantation. An important management implication arising out of our results is that preserving natural forest patches in a landscape dominated by palm oil plantations is one of the strategies to conserve avifauna diversity.

Abstract

The aims of this research were to identify moulds in candied fruit within three varieties of salak (i.e. sleman, gading and pondoh), and to know the effect of sugar concentration added, the time of storage, and additional of preservative chemical substance (benzoic acid) for the diversity of moulds in candied salak. The isolation method of moulds was used direct plating. In order to determine the kind of moulds, which tolerance in sugar solution (osmotic pressure), the samples were put on the surface of glucose 25% peptone yeast-extract agar (GPYA) medium, and then incubated at 30oC for seven days. After that the colony was transferred on potato dextrose agar (PDA) and czapeks dox agar (CDA) identification media. The results indicated that there were 10 different kind of moulds can be found in all samples, namely Aspergillus flavus, A, niger, A. versicolor, A. fumigatus, Aspergillus sp., Monilia sp., Mucor sp., Penicillium sp., Rhizopus sp. and Wallemia sp. In order to examine the influence of sugar concentration on the growth of moulds, the candied salaks were treated in different concentration. Candied salak with or without additional benzoicacid were treated with sugar concentration of 200 g/l, 250 g/l and 300 g/l. The highest concentration of sugar showed to lowest diversity of moulds for varieties of sleman and gading, conversely for variety of salak pondoh, the additionalof high sugar concentration showed increase in their diversity. The diversity of moulds in day of seventh was smallerthan the diversity of moulds in day of null. Abstract

The importance of medicinal plants for the preparation of “jamu" has been recognized in Indonesia and especially Java since ancient times. Jamu usually consists of blends of several kinds of plants or other natural materials. Jamu remains popular and there is an increasing demand for jamu in such diverse fields as health and beauty care, medical treatment and the preparation of fresh beverages. The problems result from the fact of most materials needed for preparation of jamu being natural resources. These materials are collected by the people in the forest and sold to the producers without thought for the conservation of these plants in nature with the consequence that some of the medicinal plants concerned have become difficult to find in the wild. Studies in the field in West, Central and East Java and at jamu enterprises and raw material sales centre show that several species of jamu raw material are very scarce. They should be classified as endangered. In this paper the significance of the endangered species as jamu in traditional Indonesian medicine is assessed and a strategy for conserving them "in situ and ex situ " is discussed.© 2002 Jurusan Biologi FMIPA UNS SurakartaKey words: conservation, medicinal plant, jamu, Java.

Abstract

The aims of this research are firstly, to study the advantages of electrophoretic techniques. Secondly, to look at the usefulness of a few mediums support of electrophoretic proteins especially the acrylamide gel. Thirdly, to examine thenumber of plant organs which could be used as the sources of plant proteins, and how these plants protein should be applied in the medium support that has been selected. Besides, the staining and detection procedures would be described, while the application of electrophoretic approach in higher plant taxonomy will also be evaluated. In this study we recorded that a number of taxonomic problems usually caused by morphological complexity within species can be solved using this experimental approach of electrophoresis. Populations of leopards continue to decrease over time. This decline is caused by many factors, such as decreasing animal prey and habitat loss. Due to a lack of animal prey, leopards frequently enter villages to find food, including livestock. Therefore, some conflicts between human-leopard have frequently occurred, and in many cases the leopard has been hunted by the villager. Consequently, the abundance of leopard in some areas of West Java have decreased. The aim of this research is to investigate: (i) local knowledge of the Girimukti Village on morphological variation of leopard; (ii) conflict between leopard and the people of Girimukti Village based on local knowledge; (iii) local knowledge on the hunting of leopard; and (iv) utilization of leopard resulting from human-leopard conflict in Girimukti Village, Sukabumi, West Java, Indonesia. Mixed methods andfield observation were applied in this study. The result of this study shows that the village people of Girimukti recognize variations of leopard and their behavior; conflict between humans and leopard has increased; hunting leopard is been undertaken by both traps and shotgun; and leopard are used for various purposes, such as trading skin and other body parts, food, traditional medicines, and as amulets. Based on this study, it can be inferred that many drivers of environmental changes that impact faunal and floral communities are social in origin and strongly related with peoples’ activities. As a result, in addition to biological properties, the social, economic and political systems must be considered and integrated into the conservation program of Javan leopards. Abstract

Rivers have essential role in human cultures. They are sanctuary for amount of biodiversity, but threatened seriouslynow. The objective of this research is to know Surakarta (Solo) rivers quality based on plankton diversity. This townhas amount of kampongs and industrial estates that discard wastes to rivers directly. Plankton community is one of the river qualities indicators, because pollutant and other organisms can influence their population. The research wasconducted at four rivers in Surakarta, namely Pepe River, Premulung River, Anyar River and Jenes River. Data wascollected in triple before and after rivers through the town. Abstract

Mangrove vegetation is one of the most richness ecosystems in tropical forest. It has high value economically and ecologically. Mangrove product can be used directly as timber, firewood, charcoal, tannin, dyes, food, medicine, raw material of industries, etc. It also can be used indirectly as fisheries, wastes processing, seashore protection, ecoturisms, educations, etc. In the past time, river estuaries in south coast of Java was mangrove habitat. However, anthropogenic activities had been reduced mangrove vegetation into relix habitat. The aim of the research was to know (1) sites of mangrove vegetation in river estuaries in south coast of Java, (2) diversity of mangrove vegetation, (3) density of Sonneratia alba J.E. Smith, and (4) physical and chemical properties of these sites. The research was conducted in March-April 2002, at 20 river estuaries from Pacitan until Cilacap, south coast of Java. The results indicated that mangrove remnant could be met in 10 river estuaries, namely Grindulu, Teleng, Bogowonto, Cakrayasan, Lukulo, Cincingguling, Ijo, Bengawan, Serayu, and Jeruk Legi-Donan. There were 29 mangrove species in estuaries, consist of major components (9 sp.), minor components (2 sp.), and mangrove associated (18 sp.). The density of Sonneratia alba J.E. Smith varied from 0 till > 250 individual per hectare. The soil sediment could be grouped into sand, silt, and clay, where silt and clay could support mangrove growth finely. The average of environmental parameters as follows: temperature of water and sediment respectively were 32.0oC and 31.4oC, pH of water and sediment respectively were 7.29 and 6.96, total dissolved solid of water was ~ 2000 ppm, dissolved oxygen of water was 9.29 ppm, and water salinity was 16 ppt.© 2002 Jurusan Biologi FMIPA UNS SurakartaKey words: mangrove remnant, Sonneratia alba, diversity, density

Abstract

A total of 150 cattle consisting of 3 breeds (Bali, Madura and Ongole) of Indonesian native cattle were used in this study. The aims of the study were to identify and characterize polymorphisms in the growth hormone loci of those breeds of cattle. The genomic DNA was extracted using Wizard genomic DNA purification system from Promega. Two fragments of growth hormone gene were amplified using PCR and continued with RFLP using restriction enzymes of AluI and MspI.
