Abstract

ABSTRACTA tissue culture and plant generation for the ceplukan, Physalis angulata, L has beendeveloped. Callus were efficiently induced on Murashige and Skoog (MS) medium supplementedwith 0,1 ppm indole-3-acetic acid (IAA) and variation of 1, 2, 3, 4, and 5 ppm benzyladenine (BA).The regenerable callus differentiated into shoots. Phytochemical containing of the shoots weredetected and compared with the origin plant using Thin Layer Chromatography (TLC). Flavonoidwas showed by bright blue spot under UV 366 nm and became yellow after smoked by NH3 withHRf 43 and 69. Saponin was showed by yellow spot with HRf 18 and purple spot with HRf 45 aftersprayed by anisaldehyde-sulfuric acid. HRf of the shoots from culture and origin plant were thesame, showed that shoots contains the same phytochemical as the origin plant.Keyword: shoots, tissue culture, restoration, flavonoid, saponin

Abstract

A B S T R A C TThe motivation for taking self medication increased as well as the growth of drugsproduction in Indonesia. Self medication will taking a risk, so that it must be followed by the enoughsociety knowledge about medicine. The research about an Influence of Education Level to theSociety Knowledge Level about Self Medication in Sleman Yogyakarta had been done. Thisresearch tried to shoot the society knowledge level about the aspects corelated with self medicationand an influence between the education level increased to the society knowledge level about selfmedication. From this research, can be hopen that the problems about irrasional drug using in selfmedication will be decreased. A sample that had been used is the society in Sleman Yogyakarta.The sample had been taken by stratified random sampling. This research was explorative and thedata had been gotten by quisioner and interview. The analysis of data had been done by thedescriptive statistic analysis and interferention analysis. The result showed that there was asignifican difference between education level with medium and high education level aboautknowledge of self medication. Mean value were 63,35 ± 7,27 ; 68,56 ± 6,29 ; dan 68,69 ± 7,72 .The difference specially about drugs classification, side effect of drugs and characteristic of drug.The result showed that if the eduacation level increased, the society knowledge level about selfmedication will be increased.Key word : drug, self medication, knowledge, education level

Abstract

ABSTRACTMedicinal plant has formed the basis of sophisticated traditional medicine systems that havebeen existed for thousands of years. Many advances in the chemistry of novel bioactive naturalproducts are reported. This article looks at potential chemotherapeutic and chemopreventive ofsome natural products such as curcumin, polyphenol, and dietary phytochemicals. Abstract

ABSTRACTObesity is considered as a health problem that can promote the risks of getting disease, beingnot active and death. In some developing countries, many traditional therapies are applied for somediseases, including obesity. Kemuning (Murraya paniculata (L.) Jack) has been commonly knownas slimming potion. In present, kemuning leaf is used as ingredient of traditional medicine formula,indicated as slimming potion. Former researcher mentioned that 30% ethanol-bemusing extract hadbeen able to reduce trigliserid content in the blood reduce cholesterol content in blood serum ofmale Wistar furrow white rat (Rattus norvegicus). This research is proposed to examine the effect ofwater soluble fraction of 50% ethanol-kemuning extract to female Wistar furrow white rat bodyweight which given high fat diet and to compare the effect with anorextica (mazindol). Testedanimal are divided into 6 groups, each group contain 5 rats. Group I as negative control is given0,5% CMC Na solution. Group II as positive control is given 0,27 mg/kgBody Weight mazindolsuspension. Group III is given 50% ethanol-kemuning extract suspension with 475 mg/kg BWdosage. Group IV is given watersoluble fraction suspension of kemuning extract dosage I that is145 mg/kgBW. Group V is given watersoluble fraction suspension of kemuning extract dosage IIthat is 295 mg/kgBW. Group VI is given dose III that is 585 mg/kgBW. The suspension is givenorally with repeated dosage. Data collected are the increase of body weight, woof consumption anddrinking consumption. Body weight and woof consumption increases are analyzed by one-wayAnava with 90% significancy level. Research result shows that water soluble fraction of 50%ethanol-kemuning extract with 585 mg/kgBW dosage and positive control (mazindol) are able toobstruct rat body weight increase significancy (p < 0,10) compared to negative control group.Keys word : Murraya paniculata (L.) Abstract

ABSTRACTRecently, the discovery of new drugs uses the new concept by modern techniques instead ofthe convenstional techniques. In the development of scientific knowledge, the role of molecularbiology and the modern techniques in the investigations and discovery new drug becomes theimportant things. Many methods and modern techniques use in the discovery of new drugs, i.e,genetic enginering, DNA recombinant, radioligand binding assay technique, HTS techniques (HighThroughput Screening), and mass ligand screening. In this review article, its focus in the role ofmolecular biology and HTS techniques (High Throughput Screening) which accelerated thediscovery of new drugs. The role of molecular biology will be the important things on thecharacteristic of structure of protein target as well as receptor that interact with the drug molecule orligand. By the modelling binding receptor technique, its will be known the conformation of bindingreceptor and ligand in the three dimension. In addition, by the quantum mechanics, molecularmechanics, and molecular dynamics will be identified the active sites, the consuming energy, andthe conformation of the structure of molecule drug as well as the receptor when its interactbiochemicaly. In other hand, the investigation of active compounds from natural resources stillconducted. The investigation of active compound from natural resources, at present use the HTStechniques (High Throughput Screening) could screen thousand extracts combine with HumanGenom Project data in this technique. By the HTS techniques (High Throughput Screening) will beperformed screening extract from the natural resources rapidly. Active compounds resulted fromthis technique, subsequently identified and characterized the structure of molecules and synthesisinstead of natural resources. Finally the compounds could be synthesized in the large scale.Themolecular biology and HTS technique will play the big roles on the discovery of new drugs. Themolecular bology techniques related the protein target and receptor have been changed the newparadigm in the design of new molecule drugs by rationally, wheares the HTS technique willaccelerated sceening active compounds from natural resources.Key words: molecular biology, HTS (High Throughput Screening), new drugs

Abstract

ABSTRACTGlutathione S-transferase is amount of enzymes catalyze conjugation reac-tion of glutathione(GSH) with electrofilic endogen compound or electrofilic xenobiotic. At the particular cancer there isincreasing GST, especially on mu and phi class that it cause therapy on cancer cell by citostaticdrug (commonly as electrofilic compound) not effectively. Pentagamavunon-0 (PGV-0) is antiinflammationcompound. Base on in vivo research denoted this compound has five timesstrengthened than curcumin to inhibit GST enzyme activity. But using PGV-0 by in vivo to GSTactivity never report or publish yet.This research was done by measuring isolated GST enzyme activity from rat liver was given invivo PGV-0, with nine groups treatment. Each group consist of control group without treatment,control group CMC-Na 0.5 % per oral and intraperitoneal DMSO, group with doses treatment ofPGV-0 20, 40, 80 and 160 mg/Kg BB per oral, rat group was given benzo(a)pirene (BP) 1 mg/KgBB i.p. and group was given BP 1 mg/Kg BB i. p. and PGV-0 80 mg/Kg BB per oral. GST enzymeactivity is measured from reaction of conjugate GSH and DCNB that it’s absorption/minute. Base onrate (absorption/minute) GST enzyme activity can be accounted. Then inhibition percentage can beaccounted from each rat treatment compared with control without treatment and solvent control.Analyze use statistic test (Kruskal-Wallis then continued by Mann-Whitney test), to know specificdifferent of GST class mu enzyme activity in rat liver at each treatments group.Result research denoted that PGV-0 give inhibition effect to GST class mu activity of rat liver byin vivo. Doses level did not give proportional result to GST class mu activity of rat liver. Doses levelof PGV-0 give optimum result to GST class mu activity of rat liver inhibition at 80 mg/Kg BB withinhibition value 19,99 % compared with CMC-Na as solvent.Key word: pentagamavunon-0, in vivo, glutathione S-transferase, inhibition

Abstract

ABSTRACTAim of this research is to know factor influenced to antioxidative activity of chloroform extract oftea leaf ( Camellia sinensis) with -tocopherol in peanut oil system. Research was done by takedata of peroxide value, tiocyanic acid value dan barbituric acid value of the system addedantioxidant at various concentration. Variance analysis was investigated to the data by interpretingfactors consist of time, extract concentration and -tocopherol concentration followed bydetermination of regression model. Result showed that time, extract concentration and -tocopherolconcentration influence to the antioxidative activity and there was interaction effect of the factors.Optimum condition gained at time of 60 minutes, -tocopherol concentration of 110 ppm andextract concentration of 100 ppm. The model resulted from this analysis can be used to predictsynergistic effect between chloroform extract of tea leaf with -tocopherol.Key word: antioxidant, synergistic effect, extraction
