Abstract

This research was conducted to determine the growth of earthworm ( Peryonix sp) in two types of media, that is mixture of palm sheath waste with chicken manures and mixture of vegetables waste with chicken manures. This research was conducted at Biological Garden Department of Biology Math and Science Faculty Riau University. The research used completely randomized design with five treatments and three replications i.e treatments M1 (100% palm sheath waste); M2 (75% palm sheath waste + 25% chicken manure); M3 (50% palm sheath waste + 50% chicken manure); M4 (25% palm sheath waste + 75% chicken manure) and M5 (100% chicken manure), S1 (100% vegetables waste); S2 (75% vegetables waste + 25% chicken manure); S3 (50% vegetables waste + 50% chicken manure); S4 (25% vegetables waste + 75% chicken manure) and S5 (100% chicken manure). Analysis of variance showed that type of media had significantly affected body weight and amount of coccon produced. The biggest body weight and amount coccon were at media M2 and S3 in the day of 20.

Abstract

One of wastes that has high N-total is tofu liquid waste. Biologically waste disposal can be done by phytoremediation using Pistia stratiotes L. The purposes of this research were to test the growth ability of P. stratiotes in tofu liquid waste and to test its ability in accumulating N-total. The research used Random Device Complete with variation of tofu liquid waste concentrations i.e 20, 40, 60, 80, 100 %, and 0 % for control. The observed parameters consisted of fresh weight and the growth progress of P. stratiotes, and the analysis of N-total. The data was analyzed using ANOVA and followed by the DNMRT test at the level of 5 % . The result showed the decrease of fresh weight and growth rate significantly, in the concentration of 20 % ( 20,90 g and 7,07 g/ day). The analysis of N-total in final waste was significantly different in each treatment, while the content of N-total in plant was not different in each treatment, but only different with control. The decrease of plant growth and the increase of N-total content in final waste indicated that P. stratiotes was not effective to overcome the environmental contamination caused by industrial tofu liquid waste.

Abstract

Citrus nobilis Lour. is a citrus plant that is highly demanded by many people in Riau. Propagation of C. nobilis Lour. has some problems due to various diseases that decrease its production. The conventional propagation of C. nobilis. requires a lot of parent plants and takes a long time, therefore the in vitro propagation is necessary. The purpose of this study was to determine the concentration of BAP and explant source as well as the interaction between both treatments in inducing C. nobilis bud. This research used a factorial randomized block design (RBD) with two factors i.e the concentration of BAP (0, 1, 2 and 3 mg / l) and the source of explants (shoot apex and 6 week in vitro nodes). All combined treatments showed no significant difference to the emerging bud, the number of shoots and length of shoot. The results showed that the addition of 2 mg/l BAP at the apex shoot and node explants were able to induce bud. Better explant sources in inducing bud was the node explants. The node explants at a concentration of 1 mg/l BAP gave better result in inducing bud.

Abstract

Bengkalis has various soil types, one of them is acid soil. The major problem in acid soil is aluminium toxicity that can harm rice plant roots. The objective of this study was to identify sticky chromosomes in some local rice genotypes from Desa Bantan Air, Bengkalis, Riau Province grown in Al stress medium. Plant materials examined were four local rice genotypes from the Desa Bantan Air, i.e. Amat Candu, Sadani, Solok, and Yamin. All of the rice genotypes were exposed to 12 ppm Al for 72 hours in minimum nutrient culture, pH 4. Each treatment was repeated three times. Randomized Block Design was used in this research. The results showed that sticky choromosomes were only detected at Yamin genotype treated by 12 ppm Al. It concluded that the sticky chromosomes found in the local rice genotypes from Bengkalis was Yamin genotype

Abstract

Mangosteen (Garcinia mangostana L.) from Bengkalis island has many superiorities such as growing in the marshes and tolerant to acid soil (peat soil). In vitro culture is an alternative way to get large number and uniform mangosteen seedling. The purpose of this research was to determine the best concentration of BAP (Benzylaminopurine) in inducing mangosteen’s shoots from Bengkalis. This research used Randomized Complete block Design (RAK) with 5 concentrations of BAP (0; 1; 3; 5; 7 mg/l) with 5 replications. The result of this research showed the fastest shoot forming time and highest number of shoots present in a concentration of 5 mg / l BAP at 13 DAP and 2 shoots per explant.

Abstract

Kryptopterus apogon is one of freshwater fish species that was only found in floodplain river in Riau Province. Many species of lais fish are found in Riau Province rivers and morphologically has the same shape therefore it is necessary to find alternative markers to distinguish the species. The important step in genetic analysis is DNA isolation. This study aimed to isolate the total DNA from Kryptopterus apogon. The fish samples were taken from the Kampar Kiri and Tapung Hilir rivers. Isolation of total DNA was done by taking the muscle tissue from the fish tail (1 mm x 1 mm in size). Isolation of total DNA was done using DNA isolation kit (QIAGEN DNeasy Blood & Tissue). After that, the total DNA was checked on 1% agarose gel to estimate the quantity and quality of total DNA. Total isolated DNA of Kryptopterus apogon from two rivers had various quality and quantity. Tha total quantity of DNA molecules ranged from to 100-200 ng/μl, while the total quality was good enough or not fragmented. The DNA was clear, thick, and no smear.

Abstract

Aquilaria microcarpa Baill. can produce agarwood products that can be used for various purposes, therefore it can caus a high demand for the product. Nowadays, these species are being hunted in their natural habitats. It is worried that this plant will be endangered, hence the conservation efforts for Aquilaria is necessary. The method of in vitro propagation of plants is a solution to large seed production in short time. This study aimed to obtain the optimal concentration in inducing in vitro shoots. Induction of shoots from axillar bud explants that cultured in MS medium containing BAP with concentrations of 0.0; 0.1; 0.2; 0.3; 0.4; 0.5 mg/l. The design used was completely randomized design (CRD). The observed parameters included bud appearance time, shoot number and percentage of explants that formed shoots. Data was descriptively analyzed. The results showed that the fastest emerging shoots was obtained at 0.4 mg/l BAP treatment in the third week. The optimal BAP concentration that could trigger the highest number of bud formation 0.4 mg/l BAP that produced three buds with 33.33% of bud formation.

Abstract

Durian (Durio zibethinus Murr.) is one of tropical fruit belongs to Bombacaceae family that has high economic value. Kuantan Singingi District one of durian distribution areas in Riau Province which has high diversity of durian. The purpose of this study was to identify the diversity of durian in Kuantan Singingi District. This study had been conducted from August 2013 to February 2014. Sampling was done using purposive random sampling and observation on morphological characters. Morphological data was analyzed by SIMQUAL and UPGMA procedures using the program NTSYS-pc. 2:02 and followed by correlation analysis, variability and principal components using Minitab 16:13. From a total of 32 individuals of durian, the similarity coefficient was ranged from 23% to 64%. The dendrogram formed two main groups that clustered based on the similarity of morphological characters. Pearson correlation analysis obtained 14 characters of 61 characters which were positively correlated with each other with the level of confidence was >98%. Result of principal component analysis formed two groups with 30% of the diversity value.

Abstract

Sakai is a local ethnic group in Desa Petani, Duri, Riau Province. The people of this ethnic group still strongly keep their ancestor faith, such as in using medicinal plants. This study aimed to make an inventory of the medicinal plants of Sakai ethnic group in Desa Petani, Duri based on the processing of medicinal plants in disease treatment. This study had been conducted from July 2013 to February 2014. The method used in this study was survey method. The interview of three traditional medicine practitioners were carried out based on a questionnaire list. The results of this research showed that 98 species from 48 families were used as medicinal plants by the Sakai People. The plant part that was mainly used was leaves. The medicinal plants were used by Sakai people in different ways such as being eaten, drunk, lubricated, scrubed and bathed.

Abstract

Gleichenia linearis is a wide-leaf weed that contains chemical compounds such as flavonoid, allelochemical, triterpenoid, saponin, tannin, alkaloid and steroid that can inhibit the growth of other plants so it can be used as bioherbicide. The aims of this study were to determine the potential of residues and the concentration of G. linearis leaf extract on germination and growth of seedling weed Mikania micrantha. This studywas conducted using completely randomized design with 5 replicates with three concentrations i.e. 10%, 20% and 30%. Data was observed and analyzed using Analysis of Variance (ANOVA) and tested using Duncan's Multiple Range Test (DMRT) if it showed significant effect. This study showed the decrease of growth and germination and the increase of the percentage of the death of M. micrantha weed after treatment of G. linearis leaf extract. The highest decrease was found in 30% extract concentration with the percentage of germination, fresh weight, the percentage of mortality were 19%, 0.436 g and 10%, respectively.
