Abstract

The research goals to determine the species of bacteria that can live in soil containing metal as a source of biomaterial biosorben. The research was conducted through the stages of isolation and characterization by testing the morphology, growth and biochemistry. Two isolates which have characteristics similar to the Pseudomonas genus and species of Pseudomonas sp. Pseudomonas sp has the ability to grow in extreme conditions.

Abstract

Fenol is a hazardous substance that can pollute the environment. Traditional herb Industry is one of industries produces liquid waste with high pollution level, the fenol content more than 9 mg/L.  Biological treatment  with anaerobic activated sludge method is one effective way of processing waste. This research was aimed to study the effect of MLSS variable and fenol load, the efficiency of fenol decrease in anaerobic waste processing system. The experiment is conducted by synthetic herbs waste with COD content of ± 3610 mg/L, retention time of 6 hours at a temperature of 30oC, atmospheric pressure in anaerobic baffle reactor continuously. The reactor volume is 60 L, the MLSS content of 3000; 4000; 5000 mg/L,  fenol load at 0, 3, 6; 9; 12 mg/L. Result of research show that active sludge of mixture breeding  from cage manure effective for the processing of waste with the low rate fenol (less than 6 mg/L) with the efficiency of degradation fenol 89,7-99,87% but less be effective at high burden fenol (more than 6 mg/L) with the efficiency of degradation fenol equal to 18-52%. The results obtained compared well with other reduction process.

Abstract

Adhesive polyblend polychloroprene-phenolic resin grafted with methyl methacrylate, best grafted composition due to tensile strength result will be tested the endurance of tensile strength in acid condition. The goal of this research is for knowing endurance of tensile strength after sunk in acetic acid solution with variation of sink time. Tensile strength analysis results showed that adhesive degradation was started after 56 hours. FT-IR spectogram showed C=C bond of polychloroprene which is the back bone structure of graft copolymer not founded in ~1600 cm-1 that indicated adhesive polyblend degradated caused by polyblend and acetic acid reaction.

Abstract

Apis mellifera is a favourite honey bee for the beekeepers throughout many countries. This species comprise of 24 subspecies. Based on phylogeography and morphometric evidences, these subspecies have been grouped into four lineage; namely the African (A), Western and Northern Europe (M), Southeastern Europe (C), and Near Eastern (O). Apis mellifera have been imported to Indonesia since 1972, and mostly from Australia. However, until recently there are no data about the A. mellifera subspecies and the origin. Therefore the objective of this research is to determine the lineage of A. mellifera in Indonesia based on mtDNA intergenic region between cox1/cox2 genes. In this region there are two DNA fragments, P and Q fragnant, that can be used to determine the A. mellifera lineage. The methodology used consist of samples collection, DNA isolation, DNA amplification, DNA restriction using DraI enzyme, DNA sequencing, and DNA alignment using Clustal X and MEGA spftwares. DNA fragment amplified by using E2 and H1 primer revealed a 863 bp. Digestion of the region with the DraI restriction enzyme revealed one haplotype, which consist of five DNA fragments. Based on DNA sequences and DNA alignment, A. mellifera in Indonesia was homologue with the C lineage. Its subspecies is A. m. ligustica that lived natively in Italy, they were imported to Indonesia from Australia

Abstract

Ketepeng cina (Cassia alata Linn.) was known as vegetal fungicides because it contains toxic materials to fungi. This study aimed to determined the concentration of ketepeng cina (Cassia alata Linn.) leaves extract that effective in inhibiting the growth of Cercospora personatum and to know the fungicide activity of ketepeng cina (Cassia alata Linn.) leaves extract on the growth of Cercospora personatum. This research was conducted through stage that were manufactured PDA media, isolation of Cercospora sp. and extracting ketepeng cina leaves. Fungicidal activity test was done using poisoning food method with 6 concentration level: 0%, 1%, 2%, 3%, 4%, and 5%. Experiment using Completely Randomized Design with 6 treatments and 3 replications, effect of variance between treatment were analyzed and followed by Duncans test. The results showed that the concentration of 3% effective in inhibiting the growth of Cercospora personatum. Fungicidal activity of leaves extract ketepeng cina against fungi Cercospora personatum categorized as strong in concentrations ≥ 3% and nature fungistatic.
