Abstract

An experiment was done to test the color, size, structure, dispenser cover, surfactant, and duration of observation of the sex pheromone trap for the yellow rice stem borer (YRSB) Scirpophaga incertulas Wlk. Each trap was equipped with an attractant for the male moth of YRSB in a rubber septa dispenser containing 1 mg of synthetic female YRSB sex pheromone {3(Z9-16:Ald) + (Z11-16:Ald)} (Cork, NRI, UK). All tests were done in RCBD with replications. Difference among treatments were evaluated by DMRT. Data revealed that yellow colored trap was the most attractive for the YRSB folowed by the white ones. Cylindrical trap with the diameter of 25 and 55 cm caught more moths compared to 15 cm and 35 cm, although it was not different from the 25 x 25 cm box trap. The design of dispenser cover could be either rectangular or rounded in 20 cm size. The surfactant for the trap could be lubricating oil, vegetable oil or detergent. The number of moths were observed daily. Twice and once a week observations did not differ significantly.

Abstract

Methyl eugenol (= ME) is an insect attractant. It is known that male Bactrocera dorsalis is attracted to ME. The objective of this research is to examine the effects of ME consumption on mating behavior and its success of Bactrocera carambolae. Observations were done daily from 17.00 to 18.00 hours. Mating success was characterized by the occurrence of copulation between male and female flies while mating behavior analysed by their fight intensities and wing vibrations. The results showed that the fighting and wing vibration periods of ME consuming flies were longer than those which did not. Mating success of those flies which fed on ME was relatively higher. It was concluded that ME is a stimulant for enhancing mating success of Bactrocera carambolae.

Abstract

Forty isolates of Fusarium oxysporum were tested for their pathogenicity to roselle (Hibiscus sabdariffa L. var. sabdariffa) in a plant house. The most virulent isolate was later used in a disease complex experiment with a root-knot nematode Meloidogyne incognita. Disease severity of roselle seedlings inoculated with a combination of fungus and nematode was higher than those inoculated with either fungus or nematode individually. Seedlings that were inoculated with fungus two weeks after nematode inoculation showed the highest disease severity compared to that inoculated with nematode two weeks after fungal inoculation or that inoculated simultaneously with both pathogens. It seems that root infections by M. incognita increased the colonization of roselle by F. oxysporum and subsequently caused higher damage to the roselle seedlings. The high wilt incidence in the presence of M. incognita and F. oxysporum may be due to the synergistic relationship between these two pathogens.

Abstract

Mice monoclonal antibodies (MCAs) against Phytophthora palmivora, Butl. (the pathogen of coconut bud rot) have been produced and tested. The selected hybridoma (Ab-PM.3) secreted monoclonal antibodies of IgM subclass and the titer in vitro of 10^2 - 1O^3, was employed to develop Indirect-Enzyme Linked lmmunosorbent Assay. This assay was able to differenciate coconut isolate of P. palmivora not only from cocoa and pepper isolate of the same species, but also from other fungus isolated from coconut bud rot diseased coconut (Fusarium sp., Colletotrichum sp., and Thielaviopsis sp.). This assay was able to differenciate P. pulmivora isolate from diseased coconut with those isolated from cocoa and pepper, and other fungus isolated from diseased coconut, thereby providing specific diagnostic tool for coconut bud rot which is needed for supporting the application of the concept of integrated pest mangement on coconut.

Abstract

Studies of the effect of allelochemicals or other insect growth regulator on the development of insect is difficult using parameter currently employed. By measuring the effect growth inhibiting of seed crude extract of soursop seed, (Annona muricata); neem seed, (Azadirachta indica); and leaf crude extract of Lantana, (Lantana camara) on the Heliothis armigera larvae, growth index (GI) and relative growth index (RGI) calculation are demonstrated in this paper.

Abstract

Diapause of larvae of the white stem borers, Scirpophaga innotata (Walker), is interesting and can be terminated by exposing them in a certain low thermoperiod, so that the brain is activated to initiate the production of prothoracotropic hormone (PITH). In this case water is strongly suggested responsible for stabilizing this low thermoperiod by mechanism of buffering (capture and release) heat.

Abstract

The reaction of hypersensitive Nicotiana species or cultivar (N. glutinosa, N. tabacum cv. Xanthi nc., N. tabacum cv. H-877) to three isolates of TMV from Jember showed variable symptoms. Pattern of total protein from leaf was hard to distinguish between cultivar or species. The intercellular fluids (IF) of leaf tissues which contained pathogenesis related (PR) proteins of cigar tobacco cultivar H-877 (it derives the N-gene from N. glutinosa) has lost several protein bands than that of N. glutinosa. The H-382 cultivar which systemic reaction to TMV has a band with very low molecular weight, which was not detected in other species or cultivar infected with three TMV isolates. It seems there was an effect of longterm storage of leaf before IF-extraction to the resolution of some PR protein bands in SDS PAGE. Analysis of PR-proteins of H877 cultivar in 20-SDS PAGE showed it consist of b-4, b-5, b-6a, b-6b, b-7a, and b-8a proteins of Parent and Asselin(1984).

Abstract

The objective of the study was to evaluate the responses of the diamondback moth, Plutella xylostella (L.) larvae to Bacillus thuringiensis and Neem extract. Therefore, dosage mortality studies using bioassay method were conducted on populations of P. xylostella from Garut, Pangalengan, and Lembang. Tests with B. thuringiensis, resulted in no significant differences in susceptibility between Garut and Pangalengan populations. However, those two populations were differed significantly in susceptibility to Lembang population which had the highest value of LC50 with resistance factor of 2.63, suggesting that a significant level of resistance against B. thuringiensis already occurred. In response to neem extract treatment, the results, as expected showed that there were no significant differences in susceptibility among the three populations. This indicates that neem extract could be used to control P. xylostella that has developed resistance to B. thuringiensis.

Abstract

This experiment was conducted to study the influence of temperature, pH of media, and length of radiaton on conidial germination, sporulation and growth of Trichoderma harzianum on PDA (Potato Dextrose Agar). It was conducted in a Completely Randomized Design (CRD) with 3 replications. The results indicated that the highest conidial germination occurred at 30°C, and so the sporulation and the growth of T. harzianum. Length of radiation did not significantly affect conidial germination. However sporulation was enhanced by 12 hrs light-12 hrs dark treatment and colony diameter of T. harzianum was significantly greater at dark. Acidity influenced mainly the conidial germination and the sporulation of T. harzianum. The greatest of conidial germination occurred at pH 8, whereas the smallest occurred at pH 5. Sporulation was also enhanced by higher pH. The influence of acidity on the growth of T. harzianum occurred at pH 9 which its diameter colony was significantly smaller compared to the other treatments.
