Abstract

Breast cancer is the most common cancer occurring in women after cervix cancer in Indonesia. Tumor metastasis is the major cause of mortality in breast cancer. For a tumor cell to metastasize effectively, it must induce angiogenesis. 17 β-estradiol has been shown to stimulate the proliferation and angiogenesis of breast cancer cells which express estrogen receptor (ER), T47D (human breast cancer cell line). In the present study Pentagamavunon-1 or PGV-1 [2,5-bis-(4’-hydroxy-3’,5’-dimethylbenzylidene)-cyclopentanone], an analogue of curcumin [1,7-bis-(4’-hydroxy-3’-methoxyphenyl)-1,6-heptadiena-3,5-dion], were tested on their cytotoxicity and suppression effect on angiogenic factors (i.e. VEGF and COX-2) on the breast cancer cell lines (T47D) induced by 17 β-estradiol 10-8 M. The results showed that PGV-1 performed cytotoxicity effect againts T47D cells with IC50 values 3,16 μM. This was more potent than curcumin (IC50 = 19,05 μM). PGV-1 5 μM and curcumin 20 μM decrease VEGF and COX-2 expression. These results suggest both compounds possessed antiangio-genic potensial.Key words : PGV-1, curcumin, 17 β-estradiol, angiogenesis

Abstract

The 2,5-dibenzilidine cyclopentanone is a Pentagamavunon-0 analogues (PGV-0) that showed an antiproliferative activity on raji, myeloma and Hella cells. This compound can be synthesized by condensation reaction of a keton and an aldehide by using acid or base as catalyst. The synthesis was carried by reacting cyclopentanone (10 mmol), benzaldehide (20 mmol) and potassium hydroxide 30% (20 mmol) at 5oC, followed by neutralize the reaction product with hydrochloride acid and the compound was obtained by crystalization on CCl4. It showed that rendemen in organic solven: [methanol (96.3 %), ethanol (73.2 %) and isopropanol (67.7%)]. The purity of the product was determined by using melting point and TLC methods . Structural elucidation was carried out by spectroscopic method ( UV-VIS, IR, H1-NMR and MS spectra). It could be concluded that the compound is 2,5-dibenzilidincyclopentanone.Key words: 2,5-dibenzilidincyclopentanone, catalyst, solven.

Abstract

Seledri (Apium graveolens) and urang aring (Eclipta prostata (L.)L.) have been used traditionally for hair grow. In this study, we want to test wether the both substances have an activity against Pityrosporum ovale as fungi causing dandruff. Anti Pityrosporum ovale activity of ethanol extracts of seledri (Apium graveolens) herbs and urang aring (Eclipta prostata (L.)L.) leaves had been studied using agar diffusion and dilution methods. Anti Pityrosporum ovale activity was shown by both of the two extracts, but seledri herbs extract showed stronger effect with inhibition diameter of as large as 16.33 ± 2.08 at the concentration of 5% w/v whereas that of urang aring leaves showed the inhibition diameter of 12.67 ±1.15 at the same concentration using agar difusion method. With agar dilution method, both of the two extracts still showed the inhibitory effects at the concentration of as low as 0.11 mg/mL.Key words : antifungi, Pityrosporum ovale, seledri, urang aring

Abstract

Gamavuton-0 (GVT-0) is a curcumin-like compound, which is more stable than curcumin and still has an antioxidant property. The synthesis of diacetyl gamavuton-0 (diAcGVT-0) yielding an acetylation product that is more lipophile than GVT-0. The synthesis of diAcGVT-0 has been done by reacting GVT-0 and acetyl chloride as an acylating agent and tertiary amines as primary bases. DiAcGVT-0 was formed as the major product of synthesis but the product was not pure. Structure elucidation using UV-Vis, IR, 1H NMR and MS spectra showed that the main product was diAcGVT-0.Key words : diacetyl gamavuton-0, acetyl chloride, acylating agent, lipophile

Abstract

Antiangiogenesis (inhibition of new blood vessels formation) has become a strategy to inhibit cancer development lack of nutrition and oxygen supply. The aim of the present research is to investigate antiangiogenesis effect of ethanolic extract of Gynura procumbens (Lour.) Merr. Leaves in situ using chick embryo chorioallantoic membrane (CAM). Eight to 9 days old fertilized chicken eggs were treated with b-FGF (angiogenesis inductor) and extracts. Eggs were then incubated for 3 days in order to observe its angiogenesis response (new blood vessels converged toward the implant).The results showed that the ethanolic extract of G.procumbens could inhibit angiogenesis in a dose-dependent manner. Doses 10, 20, 40, 80 ug gave angiogenesis response of (in percent) 82.32 ± 6.33; 68.38 ± 6.24; 56.48 ± 11.61; 41.43 ± 7.46 (p<0.05), respectively. These results indicate a potential antiangiogenic effect of the extract.Key words: antiangiogenic, CAM, G.procumbens.

Abstract

Curcuma zedoria Rosc. has been used empirically for cancer medicine and usually used for long periods. The aim of this study is to assess the effects of dried extract Curcuma zedoria. Rosc. orally on kidney function of male rat for 12 weeks exposure. Eighty male rat (180-200g) were divided into four group, group 1 as a control group, group 2, 3 and 4 were given 9mg/200g body weight, 18 mg/200g bw and 27mg/200g bw respectively. Dried extract of Curcuma zedoria Rosc. was given daily for 12 weeks periods, serum, urine, and kidney samples were collected by killing of 5 rats in each group on week 4, 8, 12 and 16 respectively. Serum, and urine samples were analyze for creatinine,pH, specific gravity and morphological and histological of the kidney were also observed respectivelyThe results indicated that serum, and creatinine content, pH and specific gravity of urine were not significantly different between control and treatment group of dosage and time of exposure (P>0.05). From the results can be concluded that dried Curcuma zedoria Rosc. given orally at the dosage up to 27mg/200g bw for long periods was safe for male rats and safely to use as a traditional medicine given at the long periods of time.Key words: Curcuma zedoria. Abstract

The present study was carried out to investigate the effect concentration level of pregelatinized and pregelatinized marantha starch phosphate as matrix polymer on the drug release profile. Pregelatinization starch was made by heating suspension of marantha starch with drum dryer. Phosphorylation of pregelatinized maranta starch was prepared by reacting pregelatinized marantha starch with Na2HPO4 and NaHPO4 (2:3). The modified starch product was used as matrices for tablet controlled release separately by direction compression process. Theophyllin was used as a model drug hydrophobic. The dissolution test was carried out separately in 0,1 N HCl (pH 1,8) and in phosphate buffer (pH 7,2), both for 8 hours by using paddle method.The result showed that there was no significant difference (P< 0,05) among the drug release profile from different level concentration of polymeric matrices. The drug release rate was found to be governed by the type and concentration level of polymer in matrix system, polymeric concentration (25%) in matrix, the slower release rate of the drug. Release profile showed a tendency to follow zero-order kinetics from all matrix tablets, and the drug release may be controlled by combination of diffusion and erosion delivery system.Key words: pregelatinized maranta starch phosphate, controlled release, polymeric matrices.

Abstract

It has been evaluated the influence of fermented horse milk into Balb/c mice on immunity against hepatitis A vaccine. The study employed a complete randomized design using thirty mice which were divided into 3 groups. Each group orally consumed fermented horse milk at dose 0.4 mL/20g body weight, levamisole (as positive control) at dose 0.4 mL/0.45 mg/20g body weight and distilled water (as negative control) at dose 0.4 mL/20g body weight respectively on a daily basis in addition to the standard diet for 46 days. The mice were immunized intraperitoneally (3 times) at d 7, d 28, and d 42, with 0.65 μg/20g body weight of hepatitis A vaccine. Blood samples were collected from plexus retroorbitalis using heparinized capillary one week after first (d 14), second (d 35) and 4 days (d 46) after third immunizations, and then mice were sacrificed to isolate the lymphocytes. The IgM, IgG and IgA titers in the sera and the culture media were measured by ELISA.The IgM (except d 46), IgG (subtype IgG1 (except d 14), IgG2a, IgG2b, IgG3 (except d 35), and IgA titers in the sera significantly higher (p&lt;0.05) than controls. This difference was associated with enhanced in vitro antibodies secretion by cultured lymphocytes isolated from the spleen for IgA. The result suggested that orally fermented horse milk supplementation inhances immunity against hepatitis A vaccine in Balb/c mice. The SDS-PAGE showed that 30-kDa proteins were present in fermented horse milk as well as fermented cow milk, but 7.4-kDa protein present only in fermented horse milk.Key words : fermented horse milk, immunity, hepatitis A vaccine, Balb/c mice.

Abstract

The aim of the study were to determine the peroxide value and anti DPPH free radical scavenger activity of Knema laurina (Myristicaceae) methanol extract as indicator og antioxidant properties. Peroxide value wasdetermined by iodometri-titration method while anti DPPH free radical was determined by spectrophotometry. Based on phytochemical screening, the chemical compounds of K. laurina were essential oil, sterol, triterpen, tannin, peroxide sugar, alkaloid and saponin. Peroxide value (POV) of K. laurina methanol extract was 158.07 peroxide/1kg sample, while POV of α- tocopherol was 363.96 peroxide/1kg sample. It mean that the inhibition of oxidation process of K. laurina methanol extract was better that that of α-tocopherol so the peroxide formed was lower. The IC50 of K. laurina methanol extract was 39.72 ppm, while IC50 of vitamin C was 12.20 ppm. The result showed that methanol extract of K. laurina at the concentration of 39.72 ppm inhibit 50% of free radical DPPH activity. Based on the peroxide value and IC50 of K. laurina methanol extract, it can be concluded that K. laurina methanol extract had potential activity as anti-DPPH free radical and could be acted as reductor in oxidation process.Key words : Knema laurina, peroxide value (POV), free radical DPPH
