Abstract

Indonesia kaya akan umbi lokal yang semakin hari semakin dilupakan oleh generasi sekarang sebagai sumber bahan pangan yang sehat. Seiring dengan makin meningkatnya penyakit degeneratif terkait tingginya asupan glukosa, perlu digali kembali kekayaan lokal umbi-umbian dan pembuktian kandungan karbohidrat sehat di dalamnya yang dapat dimanfaatkan sebagai pangan di masa depan. Penelitian ini bertujuan untuk mengetahui indek glikemik (IG)  umbi-umbian yang diberikan secara oral pada tikus. Umbi yang diteliti antara lain, Uwi (Dioscorea alata), Gadung (Dioscorea hispida) dan Talas (Colocasia esculenta). Tikus dikelompokkan menjadi 9 kelompok masing-masing terdiri 4 ekor. Umbi diberikan dalam bentuk serbuk yang disuspensikan ke dalam Na CMC 0,1%  dengan dosis 2,5 g/kg berat badan (BB), kontrol positif berupa glukosa pemberian oral dosis 2,5 g/kg BB. Dilakukan pengambilan sampel darah hewan uji pada jam ke 0 (sebelum pemberian senyawa uji atau kontrol), 0,5; 1 dan jam ke-2 setelah pemberian senyawa uji.  Serum darah selanjutnya ditetapkan kadar glukosanya menggunakan GOD-PAP kit. Data berupa kadar glukosa darah hewan uji versus waktu dianalisis menjadi Area Under Curve (AUC) kadar glukosa versus waktu (0-2 jam). Perhitungan IG dilakukan dengan membandingkan AUC (0-2jam) senyawa uji terhadap AUC (0-2jam) glukosa. Hasil penelitian menunjukkan bahwa ketiga umbi yang diteliti memiliki nilai IG rendah (14-22). Umbi lokal dapat dikembangkan sebagai sumber karbohidrat yang sehat.

Abstract

It has been evaluated the influence of fermented horse milk into Balb/c mice on immunity against hepatitis A vaccine. The study employed a complete randomized design using thirty mice which were divided into 3 groups. Each group orally consumed fermented horse milk at dose 0.4 mL/20g body weight, levamisole (as positive control) at dose 0.4 mL/0.45 mg/20g body weight and distilled water (as negative control) at dose 0.4 mL/20g body weight respectively on a daily basis in addition to the standard diet for 46 days. The mice were immunized intraperitoneally (3 times) at d 7, d 28, and d 42, with 0.65 μg/20g body weight of hepatitis A vaccine. Blood samples were collected from plexus retroorbitalis using heparinized capillary one week after first (d 14), second (d 35) and 4 days (d 46) after third immunizations, and then mice were sacrificed to isolate the lymphocytes. The IgM, IgG and IgA titers in the sera and the culture media were measured by ELISA.The IgM (except d 46), IgG (subtype IgG1 (except d 14), IgG2a, IgG2b, IgG3 (except d 35), and IgA titers in the sera significantly higher (p<0.05) than controls. This difference was associated with enhanced in vitro antibodies secretion by cultured lymphocytes isolated from the spleen for IgA. The result suggested that orally fermented horse milk supplementation inhances immunity against hepatitis A vaccine in Balb/c mice. The SDS-PAGE showed that 30-kDa proteins were present in fermented horse milk as well as fermented cow milk, but 7.4-kDa protein present only in fermented horse milk.Key words : fermented horse milk, immunity, hepatitis A vaccine, Balb/c mice.

Abstract

Senyawa aktif Gamma Amino Butyric Acid (GABA) merupakan asam amino yang terdapat di otak mammalia dan berperan sebagai neurotransmiter.  Selain pada sistem saraf pusat, GABA juga terdapat dalam konsentrasi tinggi di sel β pankreas serta memiliki potensi sebagai antidiabetes. GABA dapat disintesis oleh hewan, bakteri asam laktat dan tumbuhan.  Penelitian dilakukan dengan tujuan mendeteksi keberadaan GABA dalam bagian tumbuhan yaitu daun Artocarpus altilis.  Deteksi dilakukan dengan Kromatografi Lapis Tipis (KLT) menggunakan fase diam plat silika, fase gerak BAW(3:1:1), dan GABA murni sebagai pembanding.  Larutan GABA dibuat dengan pelarut etanol:air (7:3).  Hasil penelitian menunjukkan GABA terdeteksi pada ekstrak daun Artocarpus altilis.

Abstract

LR-2(4-phenyl-3,4-dihydro-indeno[2’,1’]pyramidine-2(1H)- thione;  Leni  Ritmaleni  2),  which  designed  and  assumed  to  have biologically  activity  as  anticancer,  has  been  successfully synthesized  by  using  the  Biginelli  reaction.  This  research  was aimed  to  investigate  the  effect  of  benzaldehyde  excess  in  the synthesis  of  LR-2  and  to  evaluate  the  cytotoxic  activity  of  LR-2against HeLa cancer cell lines. The synthesis was done by reacting benzaldehyde, 2-indanone and together  with thiourea at one time as  said  as  one  pot  reaction  synthetic  methodology  and  the reaction was acid catalysed. The mole equivalent of benzaldehyde was  in  excess  compare  to  others.  The  effect  of  benzaldehyde  in excess is the higher the mole of benzaldehyde, the lower the yield of  LR-2.  The  cytotoxicity  of  LR-2  was  done  by  using  MTT  method and the LC50 was 268.15 μM.Key words : LR-2, benzaldehyde, cytotoxic, HeLa

Abstract

Children’s social emotional development aged 5-6 year in Permendikbud number 137 year2014 on national education early childhood standards there are level of achievement in child development (STPPA) PAUD 2013 curriculum which consists of self-awareness, a sense of responsibility for themselves and other people, and social behavior. Self-awareness covering on how to show the ability of themselves based on situation, shows prudence to the unknown people (then brought belief in a right adults), and knowing his own feelings and work them in (to exercise restraint in appropriately). A sense of responsibility of themselves and others covering on how she knows her right, obey the class rules (rules, activities), organize themselves, and responsible for her own doing for her own goodness. The act of pro-social is by playing with their peer, knowing others feeling and response it appropriately, sharing with others, appreciate others’ right/argument/and works, using acceptable norm in solving the problem (using their logical thinking in solving their problems), being cooperative with their friends, being tolerant, expressing their true feeling (happy, sad, enthusiastic etc), and acknowledge themselves with norms in their surroundings.   The Purpose of this study is to find an improvement in children’s social emotional development aged 5-6 years in kindergarten in Pertiwi kindergarten Pakis sub district Magelang regency through Geculan Bocah Dance treatment. A method of this research is experimental research with one group pretest posttest design. In this case treatment is given to children aged 5-6 years which consist of 33 children. Technique of analyzing data is using paired sample t-test. Children’s social emotional development aged 5-6 years in Pertiwi kindergarten Pakis sub district at first of 47,24 %. Then Geculan Bocah dance treatment was given. Children’s social emotional development aged 5-6 years in Pertiwi kindergarten Pakis sub district increased to 82,12 %.The size of the increase occurring of 34,88%. Conclusion that obtained in this research is: there has been increasing on children’s social emotional development aged 5-6 years in Pertiwi kindergarten Pakis sub district Magelang regency.

Abstract

Plant callus extracts are potential to be developed as ingredient in skincare products. Tomato callus is supposed to contain protein-derivatives and or other components such as secondary metabolites that play a role in skin regeneration. Therefore the production of calli is important to be studied for callus sustainable supply. This research aims to obtain optimum medium for callus induction and to analyze tomato callus development anatomically. In vitro culture response was assessed in tomato plant (Solanum lycopersicum L. ?Permata?) for optimum callus induction. Seeds were grown on ¼ MS medium for 10-15 days. Hypocotyl was excised and cultured on MS medium + 2 mg/l 2,4-D for 15 days as the explants for callus induction. Callus was transferred to MS medium with 8 variations of PGRs including the combination of BAP + NAA, and 2,4-D. Both fresh and dry weight was measured every 5 days over 60 days to establish the growth kinetics and growth efficiency of callus. Anatomic characters of calli were examined through paraffin-embedded method. The result showed of MS medium supplemented with 2.0 mg/l NAA and 0.2 mg/l BAP is optimum for tomato callus induction, based on highest number of the absolute growth rate on fresh weight (73.77% per day), dry weight (3.84% per day), and callus initiation time (5.56 days) achieved by the medium. Cells in the ground tissue of tomato hypocotyl are competent to be dedifferentiated into a callus. This research results were expected to find out suitable methods for tomato callus production in preparation for skincare uses.

Abstract

Carica papaya L. leaves are potential for cancer treatment, this is due to the presence of Ribosome-inactivating proteins (RIPs), which demonstrated a cytotoxic effect on cancer cell lines. Therefore, this research was done to find out the effect of protein fraction containing RIPs isolated from C. papaya in the induction of apoptosis on cancer cell-lines (Raji and myeloma cell-lines).The protein fraction was isolated from Carica papaya L. leaves using ammonium sulphate precipitation and analyzed the activity on supercoiled double stranded DNA cleavage, in order to identify the presence of RIP. The active fraction called ‘CP protein’, was then analyzed on the induction of apoptosis by double staining method using acrydin orange-ethidium bromide, followed by analyzing the expressions of p53 and bcl2 using immunocytochemistry.The results demonstrated that the CP protein possessed cytotoxic activities on myeloma and Raji cell-lines with the IC50 of 1.80 mg/mL and 2.49 mg/mL respectively. Late apoptosis was detected on myeloma but not on Raji cell lines which was incubated with 0.9 mg/mL and 0.6 mg/mL of CP proteins respectively. In addition, the protein was able to increase the expression of apoptotic regulatory protein, p53, on myeloma cell-line and almost undetected on Raji cell-line.Key Words : Ribosome-inactivating proteins (RIPs), Carica papaya L. , expression of p53 and Bcl-2, myeloma cell-line, Raji cell-line.

Abstract

Babi (Sus scrofa domestica) dan celeng (Sus barbatus) hewan satu spesies yang memiliki kemiripan sangat tinggi, perbedaan keduanya dilihat dari susunan basa DNA yang dimiliki. Metode analisis DNA antara babi dan celeng banyak yang belum berhasil membedakan kedua spesies tersebut seperti pada penelitian yang dilakukan Hikmah (2019) untuk membedakan babi dan celeng menggunakan primer Nk-ND1 dengan metode real time PCR namun berhasil mendapatkan perbedaan basa antara fragmen DNA babi dan celeng dari hasil sekuensing sehingga perbedaan urutan basa tersebut dijadikan sebagai primer spesifik babi untuk membedakan kedua spesies babi dan celeng dengan metode real time PCR. Lima spesies hewan yang digunakan yaitu babi, celeng, sapi, kambing dan ayam, primer babi forward 5’-GATGCCCTAAAACTATTCACC-3’ dari hasil sekuensing dan primer babi reverse yaitu 5’-TAGTGCTAGGGATAAGGCTAGG-3’ desain Hikmah (2019), SsoFast™ EvaGreen® Supermix (Bio-Rad), proteinase-K (Invitrogen), isolasi DNA menggunakan Geneaid Genomic DNA Mini Kit. Validasi metode real time PCR dengan uji spesifitas primer terhadap 5 isolat DNA jaringan segar, uji efisiensi dan sensitivitas pada 6 konsentrasi DNA (50; 5; 0,5; 0,05; 0,005 dan 0,0005 ng) dan sosis referensi campuran daging sapi:babi (100%; 50%; 25%; 10%; 5% dan 1%), uji keterulangan dilakukan sebanyak 5 kali replikasi. Hasil analisis menunjukkan sepasang primer spesifik terhadap DNA babi dan celeng. Efisiensi DNA babi yaitu 109,5% dan R2 = 0,967, efisiensi celeng 99,1% dan R2 = 0,999. Batas deteksi absolut DNA babi dan celeng yaitu 0,05 ng/µL dan 0,5 ng/µL, batas deteksi relatif sosis babi yaitu 5% dan sosis celeng 1%. Nilai RSD rata-rata pada analisis keterulangan DNA babi dan celeng yaitu 4,210% dan 3,611%. Pada sampel tidak terdteeksi adanya DNA babi dan celeng, secara kuantitatif metode ini tidak memenuhi persyaratan validasi.

Abstract

Recently, the research to find alternative sources of carbohydrates as a replacement for rice has been developed. Walur is one of the carbohydrate sources that can be explored because it can be grown in any area with out special treatment. However, walur has limitation for direct consumption, because it contains calcium oxalate. The purposes of this study were to determine the chemical character (macronutrient content), calculate glycemic index and get the proper washing techniques to elimin ate calcium oxalate of walur. Macro nutrients content studied in this research include carbohydrates, fats, protein, star chand crude fiber. Analysis of macronutrients has been chemically done, while the glycemic index was measured by in vivo using glucose as a standard. Elimination of calcium oxalate was conducted by washing the fresh walur tubers using a solution of0.01NHCl-NaOH, 5% NaCl, and solution of lemon-lime. The content of oxalate before and after washing was analyzed by permanganometry method. The results showed that walur containing 4.34 ±0.07% of reducingsugar,3.24 ± 0.06 % of not-reducing sugar, 11.27±0.40 % of crude fiber,0.03±1.05 % of starchand0.57±0.01 % of protein. Qualitative analysis of fatty acids showed that hexade canoicacid, octade cadienoicacid, and the acide icosatetranoic were detected in high concentrations. The glycemic index valueof walur was relative lylow, about of 16.9. In addition, washing technique using a solution of lemon-lime was the most excellent technique and can reduce the oxalate content up to61.82%.Fromthis research, it can be concluded that walur can be used as food substitute esrice after washing treatment using lemon-lime solution to remove the calcium oxalate content.
