Abstract

Twenty five microbes could be isolated from industrial effluents.Seven isolates of those examined microbes were able to grow on benzamide as sources of carbon,energy,and nitrogen..The highest growth on benzamide was shown by bacterial isolate D1.Besides on benzamide, isolate D1 could grow on acetamide,acrylamide, benzamide,nicotinamide and propionamide, respectively.. On carboxylic acids, however isolate D1 could grow only on acetic acid, propionic acid, and benzoic acid as carbon and energy sources.When isolate D1 grew on 40 mM benzamide, the doubling time(\j was 6 h 40 minutes, the specific growth rate (\J) was 0,046 h'\ the attained maximum cell biomass was 4.96 g cell dry weigtAiter medium, and the yield coefficient (Y) was 124 g cell dry weight/mole benzamide.

Abstract

The study of genomic diversity in molecular level on two sub-species i.e indica andjavanica rice types were represented by cv.Pelita 1/1 and Rojolele respectively.Javanica called padi bulu due to long aunt on the grain. Polymerase Chain Reaction (PCR)with 17 primers have been used for detecting genomic variation between these two sub-species.Results of experiment indicated that 12 out of 17 primers showed polymorphic DNA.This study would assist plant breeder to decide,whether to improve rice quality through crossing between javanica and indica is necessary.

Abstract

Study on the differentiation of several explants by in vitro culture were conducted on Philodendron goeldii for propagationpurposes.Activated charcoal added to the initiating medium did not improve the development of the cultures.In general all type of explants were successfully differentiated into plantlets in all media tested. The most potential explants for propagation of this species are the shoot tip and the stem.Addition of 0.3 ppm NAA to the basal medium with different cytokinins induced roots development on shoot tips,petioles and young leaves cultures.The ability of culture to produce multiple shoots such as in medium with NAA + 2-iP or NAA + BA would give a better opportunity for in-vitro propagation of P. goeldii.

Abstract

Mangosteen seeds were irradiated with gamma rays from Co dosages 2.5 Gy, 5 Gy, 7.5 Gy, 10 Gy, 20 Gy, 40 Gy and 60 Gy. The germination rates of irradiated seeds were reduced by an average of 20 % at 20 Gy, 67 % at 40 Gy and no seed germination at 60 Gy.The effects of the treatment on the survival of irradiated seeds were observed.Gamma rays at low dosage (2.5 Gy) was stimulating to seedling growth. The best dosages for inducing variation were around 10 - 20 Gy;variation induced by the treatment included total leaves number and seedling height.

Abstract

Rainfall and runoff harvesting study at the Kali Garang basin, Semarang under the cooperation program between Centre for Soil and Agroclimate,Agency of Agriculture Research and Development (CIRAD) France and Government of Central Jawa for improving soil productivity,controlling flood and drought are presented in this paper.The results show that through water harvesting it is possible to increase diversity, improving added value and decreasing agriculture risks of agriculture commodity. Moreover rehabilitation of degraded land and soil conservation practices can be promoted.

Abstract

One field experiment had been conducted at Institute for Research and Agricultural Technology Evaluation at Taman Bogo FieldSta,to screen some Bradyhyzobium inoculants on soybean mutant lines developed for acid soil.Inoculants evaluated were single strain B-22,and its mixed inoculants (B-22+B37), (B-22 + G49), and (B-22+TAL 102) on two soybean mutant lines No.07 and 58,and cv.Wilis as check variety. Plant growth and N yield were determined at pod development (R4) and grain maturity (R8) stages.The effect of inoculation significantly influenced nodul-ation and N yield and tended to increase plant dry weight 25%.Mixed inoculant (B-22+B-37) showed good symbiotic compatibility with the three soybean genotypes tested,whereas inoculant(B- 22+G49) only showed good compatibility with mutant line No.58.Cultivar Wilis had broader symbiotic spectrum as compared to mutant line No. 07 and 58.The effect of plant genotypes,and its interaction with inoculants were significant on grain and N yield.It was observed that at pod development stage,the plant growth of mutant line No. 07 were slower than the other genotypes but it had better nutrient translocation rate to grain which resulted grain yield of cv. Wilis. The results showed that this mutant line could be developed for this area.

Abstract

A study was done to evaluate the effect of bee propolis and royal jelly on Staphylococcus aureus growth and duration of abscess healing.In vitro and in vivo methods were used on the study. In vitro method conducted by dilution method. In vivo 3 study treatments were divided in four groups.Each group was infected by 1.66.10 cell/ml of S.aureus fo get abscess.The group consists of treatment by abscess surgical and given bee propolis (A),abscess surgical and given royal jelly(B),abscess surgical and not given medication,and group control, no abscess surgical nor given medication.In vitro result showed minimalbactericidal concentration value of bee propolis and royal jelly were the same at concentration 6.25%.In vivo result showed duration of abscess healing were significantly different (p < 0.01).

Abstract

Mass production by using micro propagation technique has an important role for Lilium longiflorum regarding that it can produce seedlings in relatively short time, disease free, and regularly time. Several factors affected the successful micro shoots and bulblets formation, i.e.medium, explant, and temperature. This research aimed to know the effect of direction and width of scale section on the multiplication of kerk lily in vitro.Research has been conducted in the Tissue Culture Laboratory of Indonesian Coffee and Cocoa Research Institute, Jember.The research was arranged in factorial completely randomised design, i.e.direction of scale section and width of scale section. Two levels of direction of scale section namely longitudinal and transversal and five levels of width of scale section namely 1, 4, 6, 8, and 10 mm.The results showed that scales are able to produce micro shoot only, bulblet only, or both micro shoot and bulblet.The number of micro shoots produced from longitudinal section are higher than those of produced from transversal section. Width of scale section affected micro shoots and bulblets production, which their correlations are quadratic.The trend of micro shoots production was contrary with bulblet production.

Abstract

Raistonia solanacearum,the bacterial wilt pathogen, has a wide host range and genetic variability.Rapid and sensitive molecular techniques need to be developed for eariy detection and strain differentiation of the pathogen.Molecular techniques such as PCR and DNA hybridization have been succesfully used to detect and identify bacterial plant pathogens including R.solanacearum.These techniques were adopted under Indonesian condition, using purified and crude DNA from infected plant samples.An R.solanacearum specific DNA primer (OH/Y2) was used in the PCR test,and a DNA probe 5a67 were used in the non-radioactive hybridization test.The PCR techniqe could be used to detect R.solanacearum from infected plant samples in less than 5 hours.The DNA hybridization technique was applicable to differentiate strains ofR.solanacearum into three groups based on their DNA profiles.
