Abstract

As one of the most important crops in Indonesia, sugarcane productivity is currently still hampered by Sugarcane Streak Mosaic Virus (SCSMV), with the intensity of leaf damage reach 60% . The symptoms of this disease are the occurrence of irregular pattern between light green to pale in sugarcane leaves.  The information about SCSMV in plant anatomy and physiology was still lacking. Therefore the objective of this studies was to determine the effect of SCSMV infection to morphological, anatomical and physiological characters of sugarcane leaves. The research was performed on 30 sugarcane clones by using a randomized block design with three replications. The results showed that there was change in the morphology of leaves, but leaf color did change. The decrease in the ratio of chlorophyll a / b leaves 65% higher in the resistant clones, i.e. clones PS 06 103 and PS 06 199. The number of stomata also decreased in clones by SCSMV.

Abstract

Rubiaceae is a cosmopolitan family of flowering plants distributed at almost all over the world. This family consists of 611 genera and 13,500 species. One of the 611 genera is Guettarda that has 50 to 160 species. Guettarda speciosa L. is one of species within Guettarda which is distributed from East Africa to the Pacific Islands including Indonesia. The objective of this study, therefore, was to determine the species’ distribution in Indonesia and its potential use. The data was collected from herbarium specimens deposited in Herbarium Bogoriense, field collection data from exploration by Center for Plant Conservation, Indonesian Institute of Sciences, and NKRI Expedition in West Papua. Based on the collected data, an analysis was conducted to create distribution map of the species. Information related to the potential use of this species in Indonesia showed thatit iscommonly used as ornamental plant, although Rumphius reported that in Ambon the wood or bark of this species was usually boiled together with root ofArtocarpus as a traditional medicine for diarrhea.

Abstract

An evaluation of protein content and pasting properties of rice using molecular markers approach is still limited. This study was aimed to evaluate protein content and pasting properties of japonica rice using DNA marker in complement to conventional tools. This study was further analysis based on the data of protein content and pasting properties of 22 japonica varieties and genotyping data using 30 DNA markers. Multiple regression analysis was performed to formulate equations to estimate the protein content and pasting properties. As a result, the protein content and Rapid Visco Analyzer (RVA) pasting properties values of 22 varieties demonstrated in the range of japonica rice with high variation. Correlation between protein content and RVA pasta properties showed differential profiles. Several type of DNA markers tested in this study showed allele variation among varieties. To estimate the protein content and several parameters of pasting properties of japonica rice, six marker sets in the form of model equations were successfully developed. High resolutions of equations with signifikant R2 in range of 0.95 - 0.99 showed their potency to predict rice physicochemical properties. These developed marker sets are an initial step which needs further validation in order to be able for evaluation of thephysicochemical properties in early generation of breeding and germplasm collection of japonica rice in the future.

Berita Biologi 3 : 38 - 44. A study was conducted to evaluate some Rhizobium strains on the growth of A. chinensis and A. lebbeck and used them as the inoculants. Three experiments were carried out under the green house conditions, the 0.5 gallon plastic pots were filled with sterile sand as the medium. The experimental design was carried out Completely Randomized Design with five replicates. The first and second experiments were implemented on A chinensis which were consisted of 10 and 7 Rhizobium strains respectively, whilst the third experiment was implemented on A. lebbeck by using 18 Rhizobium strains. Each experiment was lasted in three months. The parameters of investigation were dry weight of total plants, the plant parts above the medium surface, the plant parts under the medium surface, the roots and the nodules respectively, also was counted the symbiotic capacity. The results showed that all inoculated plants produced nodules except the plants that were inoculated with Rhizobium sttains Cg and Ri. The best inoculant was strain number 34 which implemented on A chinensis and A lebbeck.

Abstract

An experiment was conducted to evaluate the performance of production traits of the synthetic population of Border Leicester x Merino crossbred. A comparison was made between the F2, F2.5, F3 generations ( synthetic population ) and Fl generation. Estimates the effects of maternal heterosis in the synthetic populations of Border Leicester x Merino crossbred were analyzed by the method of least squares for Birth Weight (BW), Weaning Weight (WW) and Yearling Weight (YW). Two kind of analyses were executed for each traits i.e. within Fl sire generation and within F2 sire generation. For each analysis, maternal heterosis was respectively calculated as twice the difference between the least squares means of F2 and F2.5 lambs and F2.5 and F3 lambs.Pool estimates of maternal heterosis effects were also calculated for each traits. Research results showed that there was no significant difference of BW between F2, F2.5 and F3 generation. Weaning Weight was significantly higher (P < 0,05) in the synthetic population as compared to the Fl generation. Yearling Weight (YW) was significantly higher in the F2 generation as compared to F2.5 and F3 generation at the level of 5% (P < 0,05 ) and the 10% level (P < 0,10 ) respectively. Within Fl sire generation, maternal heterosis effects were 12,65%, 3,4% and 1,67% for Birth Weight (BW), Weaning Weight (WW) and Yearling Weight(YW). From the analysis within F2 sire generation, maternal heterosis effects were 1% for BW, 4,49% for WW and 3,09% for YW.Pool estimates of maternal heterosis effects were 5,78%, 3,95% and 2,37% for BW, WW and YW respectively. These results indicated that the effect of maternal heterosis gradually declined from preweaning to postweaning periods. In general, the present estimates of maternal heterosis for body weights are in close agreement with previous publications. In order to gain maximum benefit from heterosis, these results suggest that in designing a crossbreeding program, especially in developing a new synthetic population, we should take into account not only individual heterosis effects but also all other different sources of heterosis such as maternal heterosis. Genetic methods presented in this paper can be used in the breeding program in Indonesia, especially in the development of the improved synthetic sheep population. However to the determine the superiority of synthetic population over the Fl generation, the overall comparison should be made on body weight, growth rate, survival rate and fertility traits.

Abstract

Hoya purpureofusca Hook.f. (Apocynaceae: Asclepiadoideae) has been used as an ornamental plant and the international trade of this species has become increasing. This species has restricted distribution on the high elevation of Java and Bali mountains. This epiphyte climber has succulent leaves and umbellate delicate flowers. Flower has star shape, succulent and waxy, purple., c.1 cm in diameter. The observation on the morphological characteristic is aimed to select the best sample for ornamental plant. The total of 17 observed samples were obtained from three populations at Gunung Gede Pangrango National Park, West Java, Indonesia. The selection was based on the node length, size of the leaves, and flower number, size and color. The result showed that P01 from Cibodas could be developed as leaf ornamental plant, because its shortest node and small leaves. SP1 from Selabintana could be developed as flower ornamental plant, it has numerous, larger and deep purple flower. SP1 and P01 have the farest relatives distance among all observed accessions, but still have a similarity of 75%.
