Abstract

ABSTRACTTomatoes (Lycopersicum esculentum Mill.) horticultural which are very good for consumption as well as industrial materials.  However, the cultivation of tomatoes still faces many obstacles, one of them is the seed.  The quality of old seeds decreases with age of seeds, so that it will affect a crop production.  This study aims to determine whether the magnetic field strength can improve tomato plant vigor.  The study was conducted using a completely randomized design (CRD) of one factor, the induction of a magnetic field consisting of 3 levels, namely 0.1 mT (M0.1), 0.2 mT (M0.2), 0.3 mT (M0,3) for 7 minutes 48 seconds. This study uses two controls;  positive control the new seed (Sn) and negative control was the old seed (So) from not being given a magnetic field treatment each experiment unit is repeated 5 times.  The parameters measured were plant height, chlorophyll content, and carbohydrate content.  The data obtained were analyzed.  If there was a difference between treatments, it was continued with the smallest difference between treatments using the Tukey's test at the 5% level.  The results of the analysis prove that the magnetic field induction of the old seed can increase seed vigor, causing plant height, chlorophyll and carbohydrate content to be the same as plants from new seeds.    Keywords: tomatoes, old seeds, strong magnetic field

Abstract

ABSTRACT The gold mining wastewater effluent is potential source for isolating cyanides-degrading bacteria, since cyanide is commonly used in gold extraction process in the mining industry. An indigenous bacterial strain LP3, capable of growing on and utilizing of a high concentration of cyanomethane (up to 1.0 Molar), could be isolated from Cikotok gold mine effluent. Based on 16S rDNA sequence, the strain was identified as Rhodococcus pyridinivorans. During the growth on cyanomethane (CH3CN), ethanamide (CH3ONH2) and ethanoic acid (CH3COOH) were detected in the growth media, indicating that nitrile hydratase and amidase involved in the metabolism of the substrate. The involvement of both enzymes on the conversion of cyanomethane was also proved by our study on cyanomethane biodegradation using whole cells of R. Pyridinivorans LP3. Besides cyanomethane, the R. pyridinivorans LP3 could also utilize various aliphatic, aromatic, heterocyclic nitriles and amides as growth substrates. Base on these results, R. pyridinivorans LP3 is expected to be used as a potential candidate for biological treatment for cyanide-containing wastes, although further research is still needed, before being applied on a field scale.  Keywords: biocatalyst, cyanide degrading bacteria, gold mining, Rhodococcus pyridinivorans LP3

Abstract

ABSTRACTSimeuleu island is located in the western part of Sumatera belonging to Aceh Province. Floristic and phytososiological information of vegetation in Simeuleu island is limited and not much study has been conducted, especialy trees species group (diameter ?10 cm). Two  study plots were used: one hectare at Kuala Makmur forest and one ha at Alafan forest (100 m × 100 m). Density analisys at Kuala Makmur recorded 329 individuals/ha while at Alafan consisted of 377 individuals/ha (average of 353 individuals/ha).  Total basal area at Kuala Makmur 29.48 m2/ha while at Alafan  was 44.28 m2/ha (with an average of 36.88 m2/ha). The Shannon-Wiener diversity index (H?) at Kuala Makmur was 2.94 and at Alafan was 2.73 (whit an average H?of 2.84). The total canopy heights of the two study sites as were relatively similar consisting of four strata. Dominant species occurred at Kula Makmur was Shorea atrinervosa (Importance Value, IV=83.55%) and at Alafan was Dipterocarpus grandiflorus (IV=128.06%). Dominant familly at Kuala Makmur was Dipterocarpaceae and at Alafan was Ebenaceae. The distribution of dominant tree species at Kuala Makmur is less prevalent than at Alafan.  Keywords: Simeuleu, Aceh, trees structure, species richness, lowland forest.

Abstract

ABSTRACTOn CoP 18 which will be held in Sri Lanka in 2019, house gecko was proposed to be uplisted up listed from the non-appendix status of CITES to be included the list of CITES appendix II on the reasons that the hunting of the reptile was very large and uncontrolled. In order to anticipate the up listing of house gecko status, LIPI as a Scientific Authority in Indonesia recommended quota for house gecko in 2019 to be 1,800,000 individuals. To see that such a quota does not have a negative impact on the population of house gecko in the wild, a survey was conducted by using the close questioner system method by visiting housing in a village; where in this survey the villages were located at the foot hill of Gunung Karang, Pandeglang Regency, Banten Province. The survey was conducted on March 18 to April 1, 2019. The total number of villages visited was 8 villages which were divided into three sub-districts. The total area surveyed in 8 villages was around 94.5 hectares.  The total number of houses visited was 740 houses; the total number of houses that contained the house gecko was 211 houses. The total number of house gecko by interviewed was 260 individuals, while the total number of house geckos by observed was 88 individuals. The survey results obtained an average population density of house gecko by interviewing eight villages was 3.66 individuals/hectares, while the observation results were 1.15 individuals/hectares. The approach to the average population density of houses gecko by interviewing with the results of 3.66 individuals/hectares was assumed to be the density that was close to the logical number.  Extrapolating the density of house geckos in Pandeglang Regency which covers an area of 229,071 hectares excluded paddy field area was considered to be inhabited by house gecko, so the extrapolation of the number of house geckos in Pandeglang Regency is around 838,399.86 individuals.  Keywords: Gekko gecko, house gecko, Java, population

Abstract

ABSTRACTBoth Citrus macroptera Montrouz. and Citrus micrantha Wester were cited as different species from C. hystrix by several authors. Morphological characters that can be used to differentiate those species are leaf shape and fruit. C. hystrix is cultivated at Bogor Botanic Gardens originated from Central Java Province and East Sumba Island, East Nusa Tenggara Province. In previous study, C. hystrix, both from Central Java and East Nusa Tenggara, did not produce flowers as well fruit yet. The only useful characters for differentiation of the species were leaf morphology and its  thorns. Therefore, further study is needed. The research was carried out from January 2013 to December 2018 at Bogor Botanic Gardens and the Anatomy and Cytology Laboratory, Botany Department, Research Center of Biology LIPI. Flower and fruit morphology have been observed on C. hystrix originated from both Central Java and East Nusa Tenggara as well as from other localities which is commonly used. To support this reasearch then leaf and petiole anatomy have been done. The result showed that morphological characters of leaf shape and fruit of C. hystrix where originated from Central Java, differed from  C. hystrix originated from East Nusa Tenggara and those are commonly used by local people. However, based on leaf and petiole anatomy observations of all specimens used on this research are quantitavely similar, so that these results are not supported the morphological characters observation. Therefore, it is assumed that both C. hystrix, originated from Central Java and East Nusa Tenggara, as variety of  C. hystrix that commonly used by local people.

Abstract

ABSTRACTGenetic improvement through tetraploid induction of Stevia rebaudiana is important in order to increase the sweetener content, steviol glycoside. Tetraploid plants of several species after induction with colchicine and oryzalin have higher growth and secondary metabolite contents compared to the diploid plants. This study was aimed to evaluate growth as well as their stevioside and Reb-A content of S. rebaudiana tetraploid and diploid (control) plants cultured in vitro and grown in the field after acclimation process. This study used 3 tetraploid clones, namely B60.3H8, P1T22, P3T5, and 1 diploid clone as control. Shoot tips were cultured on MS medium without addition of plant growth regulators for 6 weeks, then they were acclimated in a greenhouse, followed by planting them in the field. Growth of shoot culture, plantlets in the greenhouse and plants in the field were observed. Analysis of stevioside and Reb-A was done by HPLC. The results showed that plantlets of diploid clone had higher in vitro growth and survival rate in the greenhouse than that of tetraploids. Tetraploid clone P1T22 had similar growth as diploid plants, but higher than the growth of tetraploid B60.3H8 and P3T5. Fresh and dry weights of B60.3H8 was similar with diploid plants, but higher than P1T22 and P3T5 tetraploid clones. The highest level of stevioside and Rebaudiosida-A was found in tetraploid B60.3H8 clone, the lowest was found in the diploid plants. The highest ratio of stevioside : Reb-A was found at B60.3H8 tetraploid clone.  Keywords: Stevia rebaudiana, in vitro, field, growth, Stevioside, Rebaudioside-A, tetraploid

Abstract

ABSTRACTOur previous works revealed that Trichoderma harzianum InaCC (Indonesian Culture Collection) produced volatile organic compounds that inhibited the growth of Fusarium oxysporum. The objective of this study was to evaluate the ability of T. harzianum InaCC to solubilize insoluble phosphate and optimize the carbon sources condition. Screening was conducted to 10 isolates of T. harzianum on Pikovskaya?s broth in 100-mL conical flasks. The cultures were incubated at 30 ± 1°C for  7 days on a rotary shaker at 80 r/min. All  T. harzianum isolates were observed for the solubility of insoluble tricalcium phosphate with the highest ability was performed by T. harzianum InaCC F88. Using this isolate, the optimization of insoluble phosphate solubilization was conducted on modifed Pivoskaya medium in 500-mL conical flasks in similar screening incubation condition with applying of different combination of phopshate sources (tricalcium phosphate and rock phosphate) and carbon sources (dextrose, lactose, and sucrose). When tricalcium phosphate was substituted by rock phosphate, less soluble phosphate was produced. Glucose was the best carbon source used for solubilization of both tricalcium phosphate and rock phosphate, then followed by lactose and sucrose. Acidification of medium was not the major mechanism of phosphate solubilization by T. harzianum. Based on the ability of high phosphate solubilization, T. harzianum InaCC F88 is the most potential strain as a plant growth promoting fungus among the 10  isolates of T. harzianum Ina CC.  Key words:  biocontrol, Pikovskaya medium,  phosphate solubilization, Trichoderma harzianum

Abstract

ABSTRACTThe study on population dynamic and  roosting trees damage by flying fox was conducted for 16 months (March 2008-June 2009) in Bogor Botanical Garden.  Recording of individual flying fox was carried out by use direct observation in the roosting trees habitat at 9.00-15.00, with one replication every week. The result indicated that in 2008 the total flying fox were 420 individuals and in 2009 were 563 individuals (160 juveniles, 218 adult females and 185 adult males). The natality and mortality rate in this location is 0.0214 and 0.0099 respectively. The flying fox population growth can be predicted using exponential equation is N(t+1)=563(1.8395)t.  The study also indicated that in the KRB there are 9 individual trees which are the main perch, 2 individual trees of Pterodotus alata, Pterocarpus indicus, and Shorea leprosula, and each 1 individual tree of the species  Dipterocarpus cornutus, Ficus drupacea, and Syzygium syzygioides. The extent of damage to the tree where the perch is very dependent on the diameter of the canopy, bat population size in one tree, and time periods per roosting season.  Keywords: Population Dynamic, Flying Fox (Pteropus vampyrus), Bogor Botanical Garden

Abstract

ABSTRACTThe administration of Platelet Rich Plasma (PRP) is expected to be a supplement for treatment of diabetic wounds and hyperglycemia by increasing growth factors. The purpose of this study was to examine the effect of Platelet Rich Plasma (PRP) on the proliferation and viability of human dermal fibroblast (HDF) in high glucose conditions, as a model for healing diabetic wounds in vitro. HDF cells are grown in DMEM medium containing high glucose which are then with PRP. To measure the effect of PRP on the HDF cell proliferation, CCK-8 kit was used and evaluated by using a microplate reader. To evaluate the viability of HDF, an automated cell counter. The results of the research showed that PRP stimulate the HDF cells proliferation. The optimal dose of PRP to increase HDF cell proliferation is at dose of PRP 10%.  Supplementation of PRP is significantly increased cell viability and HDF cell counts within 48 hours. The results showed PDGF growth factor secreted by PRP is increased significantly. The conclusion is PRP stimulated HDF cell proliferation and viability in a high glucose condition. Abstract

ABSTRACTFoxtail millet [Setaria italica (L.) Beauv.] is one of potential cereal plants that can be developed as a source of carbohydrates in marginal and dry lands. The purpose of this study was to examine the tolerance to drought of foxtail millet accession of Buru Merah as a result of gamma radiation treatment. In this study, the seeds of the Buru Merah accession were treated with gamma ray at doses of 0, 25, 50, 100 and 200 Gy. The established growing plants were then treated with different intervals of watering, which were every day (as control), 2 days, 4 days, and 6 days which were arranged in factorial randomized block design with three replications. The observed variables were increase plant height, increase of leaf number, leaf chlorophyll content, plant water potential and  yields. The results revealed that the radiation treatment did not show any significant difference in the production of plant biomass. With radiation treatments of 0, 25, 50 and 100 Gy. and a two-day watering interval the plants can still maintain its water potential above -3 MPa and showing optimum growth. However radiation treatment resulted a significant difference in panicle production, especially between the radiation dose of 200 Gy. and others. The combination of 0-100 Gy. radiation treatment and 2-day watering interval produced plants with optimum panicle production.Keywords: drought, growth, millet, production, radiation, tolerant
