Abstract

Xylan is a carbon source in growth medium of extracellular xylanase producing bacteria. The purpose of this study was to get the optimum medium for the growth of Bacillus sp. in producing the xylanase. The factors consist of production time, carbon, and nitrogen source, as well as simple sugars. Addition carbon source used was delignified sugarcane bagasse, rice hulls, and corn cobs with different concentrations (0.25%; 0.5%; 0.75%; and 1% w/v) . Ammonium chloride, ammonium sulfate, and sodium nitrate with different concentrations (0.08%; 0.17%; 0.26%; and 0.35% w/v) were used as a source of nitrogen, while the simple sugar used was glucose, lactose, sucrose, and xylose. The results showed that the optimum culture media of Bacillus sp. to produce xylanase is media with 0.25% natural starch from the corn cob xylan as a carbon source, 0.26% ammonium chloride as a source of nitrogen, 0.0625 grams of sugar xylose, at pH 6, incubation temperature of 40°C, and 12 hours production time. In that media, xylanase activity was 0.2 U/mL.Keywords: agricultural waste, medium optimization, xylanase, Bacillus sp.   ABSTRAKXilan merupakan sumber karbon pada media pertumbuhan bakteri penghasil enzim ekstraseluler xilanase. Tujuan penelitian ini adalah mendapatkan media optimum untuk pertumbuhan Bacillus sp. dalam memproduksi xilanase. Perlakuan percobaan terdiri dari waktu produksi, sumber karbon, sumber nitrogen, dan penambahan gula sederhana. Sumber karbon yang digunakan adalah bagas tebu, sekam padi, dan tongkol jagung dengan variasi konsentrasi 0,25%; 0,5%; 0,75%; dan 1% (b/v) . Amonium klorida, amonium sulfat, dan natrium nitrat dengan variasi konsentrasi 0,08%; 0,17%; 0,26%; dan 0,35% (b/v) digunakan sebagai sumber nitrogen, sedangkan gula sederhana yang digunakan adalah glukosa, laktosa, sukrosa, dan xilosa masing-masing sebanyak 0,0625 b/v. Hasil percobaan menunjukkan bahwa media optimum pertumbuhan Bacillus sp. untuk produksi xilanase adalah media dengan 0,25% tepung xilan dari tongkol jagung sebagai sumber karbon, 0,26% amonium klorida sebagai sumber nitrogen, 0,0625 gram gula xilosa, pada pH media 6, suhu inkubasi 40°C, serta waktu produksi 12 jam. Dalam media tersebut, aktivitas xilanase yang dihasilkan sebesar 0,2 U/mL.Kata kunci : limbah pertanian, optimasi media, xilanase, Bacillus sp.

Abstract

ABSTRACTTomatoes (Lycopersicum esculentum Mill.) horticultural which are very good for consumption as well as industrial materials.  However, the cultivation of tomatoes still faces many obstacles, one of them is the seed.  The quality of old seeds decreases with age of seeds, so that it will affect a crop production.  This study aims to determine whether the magnetic field strength can improve tomato plant vigor.  The study was conducted using a completely randomized design (CRD) of one factor, the induction of a magnetic field consisting of 3 levels, namely 0.1 mT (M0.1), 0.2 mT (M0.2), 0.3 mT (M0,3) for 7 minutes 48 seconds. This study uses two controls;  positive control the new seed (Sn) and negative control was the old seed (So) from not being given a magnetic field treatment each experiment unit is repeated 5 times.  The parameters measured were plant height, chlorophyll content, and carbohydrate content.  The data obtained were analyzed.  If there was a difference between treatments, it was continued with the smallest difference between treatments using the Tukey's test at the 5% level.  The results of the analysis prove that the magnetic field induction of the old seed can increase seed vigor, causing plant height, chlorophyll and carbohydrate content to be the same as plants from new seeds.    Keywords: tomatoes, old seeds, strong magnetic field

Abstract

Bananas are one of the horticultural crops of the fruit group that have quite high social and economic value. One type of banana that is very popular is the pisang kepok. The main obstacle in pisang kepok  production is the availability of seedlings. The above constraints can be overcome by propagation of banana seedlings using tissue culture techniques by adding colchicine to the tissue culture media. Colchicine is often used to improve plant quality. This study aims to determine the effect of adding colchicine to the media on the growth and formation of pisang kepok abu plantlet. The study was conducted from November 2019 - February 2020 at the MTC Laboratory, PT Great Giant Pineapple PG 4, East Lampung. The concentration of colchicine added to the media was 0.1%. The growth and development of plantlets produced from media with colchicine treatment will be compared with plantlets from the control group. The tests were repeated 10 times. The results showed that the growth and formation of pisang kepok abu plantlets from the treatment of colchicine were lower than the growth and formation of the pisang kepok plantlets in the control group.

Abstract

This research was purposed to understand the effect of 0.2mT magnetic field exposure treatment for 10 minutes toward medium components to the production of protease in Bacillus sp. That magnetic field exposure treatment was given to 8 medium components namely Milk, Yeast, NaCl, KH2PO4, MgSO4, (NH4)2SO4,Agar and Aquadest. Data from Qualitative Proteolytic Activity test on Bacillus sp. indicated that in all treatment, the bacteria were able to produce the enzyme. The highest Proteolytic Index (IP) from all those treatments came from the magnetically exposed KH2PO4 which was 7.17 at the 10th incubation hour. Treatment of exposure to magnetic fields is also given to the liquid medium. Quantitative data of enzyme activity showed that the best incubation time of protease production by Bacillus sp. is the 24th incubation hours with result of 0.031 U/ml. Exposure of 0.2 mT magnetic field for 10 minutes to the NaCl component in Mendel's fluid medium yielded the highest protease activity of 0.067 U/ml.

Abstract

Lettuce plants are widely cultivated in Indonesia. Based on the 2019 Central Bureau of Statistics, the amount of lettuce plant production has decreased production, because the soil in Indonesia has decreased fertility and drought. To overcome this problem by switching the use of planting media in the form of Soilless Culture System (SCS) media containing compost on Trichoderma sp. induced bromelain. SCS media containing Trichoderma sp induced bromelain solid compost will produce media that will be rich in macro and micro nutrients so as to produce quality lettuce plants. The purpose of this study was to determine the effect of using SCS media containing compost on bromelain induced Trichoderma sp. and get the best dose of SCS media containing compost on bromelain induced Trichoderma sp. on lettuce plants. This study used a completely randomized design (CRD) with media treatments used, namely husks, vermicompost, compost, coconut husk, and bromelain solid compost with the ratio of P1 (2:1:1:2:2), P2 (2:1:1:2:3), P3 (2:1:1:2:4). The parameters observed were leaf area and levels of chlorophyll a, chlorophyll b, and total. The results obtained after being tested with ANOVA 5% and BNT 5% further test stated that the leaf area and chlorophyll a, chlorophyll b, and total levels had a significant effect and the best dose was obtained, namely treatment P3 (2:1:1:2:4).

Abstract

West Lampung Biodiversity Park is a biodiversity conservation area established in 2015. The purpose of establishing a biodiversity park is to provide a reserve area, both in-situ and/or ex-situ, for local biological resources, especially plants whose structure and composition can support the existence and sustainability of animal diversity. One group of animals that has an important role in the sustainability of flora biodiversity is seed disperser. This research was conducted to find out the current biodiversity status of animals having potential as seed dispersers. The research was carried out in September–November 2021 using interview and observation methods. The data was analyzed descriptively. The results show that in the West Lampung Biodiversity Park, 10 types of animals have the potential to spread seeds, consisting of 4 members of the aves class and 6 members of the mammal class. There are 4 types of fruit spread by Aves and 17 types of seeds spread by Mammals

Abstract

Phosphorus is a very important nutrient for plants. However, most of the phosphorus is bound by soil colloids and is insoluble, so the role of phosphate solubilizing bacteria is needed to release the bound phosphate. This study aims to isolate, characterize, and test the pathogenicity of phosphate solubilizing bacterial isolates. This research used an exploratory method including taking soil samples, bacterial isolation, calculating the density of bacterial colonies, calculating the phosphate solubility index value and selecting potential bacteria, morphological and physiological characterization, and pathogenicity test. The results showed that the colony density was 3,2 x 103 CFU/g with phosphate solubility index values ranging from 1,38-3,65. The colony with the highest phosphate solubility index value is JE1 with a value of 3,65; JE2 with a value of 3,63; JE3 with a value of 3,13; JE4 with a value of 3,08; and JE5 with a value of 2,94. Colonies have an irregular shape with undulate edges, the colony color is predominantly white with umbonate, raised and flat elevations. Most isolates have short bacillus cells that are Gram negative, do not form spores, are able to ferment glucose, produce the enzyme catalase, and are not motile. The pathogenicity test showed that only the JE1 isolate was not pathogenic to tobacco leaves.

Abstract

Salah satu upaya dalam peningkatan ketahanan pangan dan gizi keluarga yang dianjurkan pemerintah adalah pemanfaatan sumberdaya yang tersedia di lingkungan, melalui pemanfaatan lahan pekarangan. Strategi penataan lahan dan pemilihan komoditas (tanaman) yang tepat diperlukan dalam pemanfaatan lahan pekarangan yang sempit dan terbatas. Hasil survei kami, menunjukkan bahwa sebagian besar ibu-ibu di kota Bandar Lampung belum mendapat pengetahuan yang cukup tentang pemanfaatan pekarangan tersebut. Ibu-ibu belum pernah mendapat pengetahuan tentang model-model penatahan lahan pada pekarangan sempit perkotaan dan pemilihan jenis tanaman yang tepat. Oleh sebab itu, pada pengabdian ini dilakukan kegiatan penyuluhan yang bertujuan memberikan pengetahuan tentang berbagai model penataan lahan pada pekarangan sempit perkotaan dan strategi pemilihan tanaman yang tepat sebagai upaya untuk meningkatkan kesehatan dan kesejahteraan keluarga. Kegiatan penyuluhan dilakukan secara ceramah dan diskusi, serta demontrasi. Kegiatan ini diikuti oleh 23 peserta yang terdiri dari ibu-ibu anggota majlis Taklim An-Nisa lingkungan Swadaya 8 dan 9, Gunung Terang, Langkapura, Bandar Lampung. Evaluasi kegiatan dilakukan dengan pretest dan postes, serta observasi langsung pada peserta melalui lembar observasi. Hasil postes menunjukkan terdapat peningkatan rata-rata skor sebesar 15,2, meningkat 22,42% dibandingkan rata-rata pretes. Sedangkan berdasarkan observasi selama kegiatan berlangsung, menunjukkan bahwa peserta selalu memperhatikan penjelasan dan terlibat dalam diskusi.

Abstract

Household organic waste is all organic waste generated at home from daily activities. Household organic waste has been a problem because it causes unpleasant odors if it decomposes and is not treated, its presence left unattended in the home environment also triggers the growth of microorganisms and insects such as flies. Household waste comes from various sources, especially food waste, unnecessary purchases. Household waste can cause illness, air pollution and environmental contamination. Household waste treatment options include landfill, burning for energy, recycling, and composting. This community service activity aims to introduce organic household waste composting methods to the congregation of majelis taklim Annisa Swadaya-89, Gunung Terang, Bandar Lampung through the method of making eco-enzyme, making compost with the help of composting microbes, and introducing composting with maggot, so that people are familiar with composting and recycling activities. The results showed an increase in the participants' knowledge level as measured by the pre-test and post-test. The participants' knowledge increased by 32% from the average final score of the respondents. The positive results of this service activity can be used by the community as a basis for changing household waste treatment patterns, so that waste is no longer a problem that reduces the quality of household environmental health of the congregation of Majelis Taklim Annisa Swadaya-89 Gunung Terang.
