Abstract

ABSTRACTThe growth character and nitrification activity of nitrifying cultures (N-Sw). Theculture of nitrifiers (N-Sw) was obtained from acclimated sludge of sawit palm oil industrywastewater. The growth and nitrification activity of those cultures were investigated.The result shows that the growth and nitrification activity attained optimum at pH 7-8,and temperature of 30 0C. The culture of nitrifiers was still growing at pH 5, but thenitrification activity was not detected. The growth of nitrifiers and the nitrification activitywas inhibited at 40 0C. The ammonium conversion rate reached 0,088-0,090 mg NNH4+/L/hour/g biomass. The rate of ammonium conversion in the bioreactor increasedto 0.630 mg N-NH4+/L/hour/g biomass as the pH maintained at 7.5-8 and dissolvedoxygen at 3-4 mg/l O2.Key words: nitrifying culture, ammonium, ammonium convertion rate

Abstract

The Effect of Anorganic- Iron Supplementation on The Blood Cell of Humpback Grouper Fish (Cromileptes altivelis ) Infected by Kbrio parahaemolyticus This experiment was conducted to find the immunity response of humpback grouper (Chromileptes altivelis) correlated to the anorganic-iron supplement in feed. The blood cell was analyzed as the infection response of F5brioparahaemolyticus. The fish weight 6.9 5 0.22 g/ind. was cultured in 60x40~50cm aquarium and stocked at density 10 ind. / aquarium. After the fish fed by inorganic-iron supplementation at 0,25,50 and 100 mg/ kg for 68 days, then infected by J! parahaemolyticus. The blood cell showed the 50 and 100 mg Felkg inorganic-iron supplement enhanced erythrocytes, hematocrite, hemoglobin, lymphocytes and neutrophil after the 24hand the 72ndh ours infection. Total leucocytes backed to normal after the 7Pd infection and the index of phagocytic was higher with the 100 mg Fe/kg of iron supplementation. 1 Key words: iron (Fe), humpback grouper, feed, bacteria and blood cell.

Abstract

ABSTRAKAktivitas Induksi Superoksida Dismutase dan Resistensi Etanol oleh Sodium Kloridadan Plumbum Asetat pada Khamir Candida sp. Y390. Untuk mengetahui sifat khamirCandida sp. Y390 dalam melindungi diri dari cekaman oksidatif, telah dilakukan risetkajian toleransi sel terhadap etanol dan hubungannya dengan enzim superoksida dismutase(SOD) pada khamir tersebut. Viabilitas sel yang ditumbuhkan pada media gliserol dandiinkubasi pada etanol 17,s % selama 1 jam adalah 0.30 f 0.09 % , pada media gliseroldan 1 1,7 % natrium khlorida adalah 1.65 f 0.5 % sedang pada media giserol dan 1 ppmplumbum asetat adalah 1.16 f 0.7 %. Aktivitas CuZnSOD pada sel yang diinduksi olehnatrium khlorida atau plumbum asetat turun sedikit, yakni masing-masing 1,7 dan 1,9kali. Aktivitas MnSOD pada sel yang diperlakukan dengan natrium khlorida naik sedikit(sekitar 1,03 kali), namun pada sel yang diperlakukan dengan plumbum asetat naik secaramenyolok (sekitar 3,6 kali). Data ini menunjukkan bahwa metabolisme etanol dapatmenyebabkan Candida sp. Y390 mengalami cekaman oksidatif. MnSOD khamir mungkinberperan dalam melindungi sel dari kerusakan akibat cekaman oksidatif.Keywords: Candida sp. Y390, yeast, ethanol metabolism, oxidative stress, SOD.

Abstract

ABSTRACTScreening of Nitrile and Protein-Degrading Marine Bacteria Isolated from Sponge in TernateSea Water. Thirty three marine bacteria have been isolated from marine sponge in Ternate byenrichment culture. Screening bacteria-degrading nitrile was done by microtitter plate methodbased on growth ability tested by Iodonitrotetazolium chloride. Product of nitrile degradationwas determined by Gas Chromatography (GC) and the potential bacteria-degrading proteinwas also screened by using selected media which contained casein. The results showed thattwenty one isolates were able to show the clearing zone in selected media. Five isolatescapable of utilizing acetamide as the sole source of carbon and nitrogen. Abstract

ABSTRACTSome commercial varieties of Guava seedless are triploid and rarely found naturally. A triploid variety with less seeds and better yield potential can be achieved by crossing tetraploid to diploid variety. Guava tetraploid plants can be synthetically induced by using oryzalin or colchicine to double the chromosome from its diploid. This research was aimed to obtain tetraploid lines by studying the effects of oryzalin in germination and in vitro growth of Guava. Seed from Red Guava were cultured in liquid medium MS + 2 mgL-1 BA with adding oryzalin according to the treatment, then seed planted in MS solid medium. Seed explants were exposed to Oryzalin 0 (controls), 15, 30 dan 60 ?M with exposure time are 23, 36 and 49 days or 3, 5, and 7 weeks. Results from polyploid induction were 8 tetraploid (5.48%) and 9 (6.16%) mixoploid shoots in vitro. High est tetraploid shoots were obtained from treatments by exposing the seed explant to 15 – 30 ?M oryzalin for 23 – 36 days. Oryzalin treatments inhibit germination and in vitro growth of Guava until 1st subculture. After second subculture, tetraploid or mixoploid shoots quantitatively showed no difference respons on in vitro growth with its diploid.Keywords: guava, tetraploid, seed explant, in vitro germination, polyploid

Abstract

ABSTRACTDibenzothiophene is one of polycyclic aromatic hydrocarbon (PAH) compound containing sulfur element. This compound has toxicity, mutagenic and quiet persistent in environment. From sreening test, it was known that isolate LBF-1-0061 was potential to degrade dibenzothiophene. The objectives of this study are to study dibenzotiophene degrading capability by marine bacteria isolate LBF-1-0061 using screening test; analysis of dibenzothiophene residue by GC/MS and identifiy the isolate by molecular identification. The result of this research shown that LBF-1-0061 isolate could grow up to 100 ppm of dibenzotiophene. This isolate also presented degrading capability approximately 37.5% of dibenzotiophene in 14 days incubation. Based on partial 16S rRNA gene analysis, LBF-1-0061 was identified 99% as Novosphingobium mathurense strain SM117.Keywords: sea bacteria, biodegradation, dibenzotiofen, hydrocarbon aromatic polisiclic

Abstract

ABSTRACTBiodiversity of Phenanthrene by Alcanivorak borkumensis M5 Isolated from Teluk Jakarta.Phenantrene is one of the most persistent organic substances in environment. AlcanivoraxBorkumensis M5 was isolated from sea water, and able to degrade phenantrene after 5 hours.About 75 % of phenantrene was degraded after 12 hours. This isolate grew optimum at 300C,pH 7.8 with a doubling time of 14.5 hours and specific growth rate of 0.0476/hour.Polyhydroxybutirate (PHB) was produced during culture growth, but the synthesis wasinversely correlated with the cell growth. Abstract

Research has been carried out through the utilization of organic fertilizer to improve the productivity of local maizecultivar originated from South Sulawesi. The study was conducted at the research station of Research Center for Biology,Cibinong Science Center. Corn seed were planted in polybag containing mixture of soil and compost, 6 and 2kg, respectively. The research were carried out by Randomized Complete Block Design arranged in factorial experimentwith four replications. The first factors are three types of local maize namely A: rice corn (pulut beras, BataraKoasa), B: waxy corn (pulut biji, Batara Kamu), C: pulut hibrida (hybrid cultivar). The second factors are 14 combinationtreatment as a mixture of LIPI organic fertilizer (Beyonic-StarTmik, Bio121, EM-121 and MegaRhizo) andanorganic NPK fertilizer. The variables observed were plant growth and production. The results showed that thehybrid cultivar had higher total corn yield productivity compared to the local one, although hybrid cultivar hassmaller plant biomass. Waxy corn (B) accession showed the weight, length and diameter of cob, and also the weightof 100 grains larger than rice corn (A). Fertilization treatments significantly affect the corn-cob productivity which isincludes the weight, length, and diameter units. Fertilization treatments such as EM-121 + (½ dose of NPK), BIO-121 + (½ dose of NPK), and Beyonic + (¼ dose of NPK) showed a good effect on plant, and it was not significantlydifferent with 100% NPK (full doses of NPK). The three corn cultivar showed different responses to fertilizationtreatments.Keywords: local corn, biofertilizer, growth, production

Abstract

The efficacy of entomopthogenic fungus for Tussock Moth Caterpillars Control. The occurrence of Tussock Moth Caterpillar (TMC) out break reported in some areas in East Java at Probolinggo, Malang, Batu and other districts were worried. The high caterpillar population not only attacking mangos, cherry, and wild watercress, but also migrating to living house before pupation. The available control of these caterpillar is chemical pesticide. However, thechemical will kill the natural enemies and other beneficial insects. A laboratory experiment to study the efficacy of five entomopathogenic fungus against TMC and pupae was conducted at the Entomology Laboratory of Indonesian Legumes and Tuber Crops Research Institute (ILETRI) in April 2011. Our experiment was arranged using completely randomized design (CRD) with three replicates using five entomopthogenic fungus. The efficacy was determined by the mortality of caterpillar/larva and pupae infected by the fungus. Result showed that these five fungus had not effectively kill the caterpillar, as the dense setae in integument acted as a mechanical barrier to fungus infection. The pupal stage was more susceptible to fungus infection. It was found that Paecilomyces fumosoroseus and Lecanicillium lecanii fungus were more effective than Metarhizium anisopliae, Beauveria bassiana and Nomuraea rileyi repectively. These two fungus could kill 90% of pupae and also were easier to be cultured in a wide range of natural media. Therefore P. fumosoroseus and L. lecanii are potentially used as a biological control agent against TMC out break.Keywords: Entomopathogenic fungus, hairy caterpillar, incubation period, colonization
