Abstract

Performance of ELISA Toxoplasmosis Diagnostic Kits on Sheep and Human Sera Toxoplasma seropositivity in Indonesia have a high prevalence, both in human and animals. Unfortunately, the availability of diagnostic tools to support dynamic surveillance are limited. Recently, the diagnostic tools for toxoplasmosis, namely ELISA BM were developed. The technology was based on ELISA technique using soluble tachyzoite antigen from tachyzoites of Toxoplasma gondii. Kit performance is one of the important issue for acceptance of diagnostic tools prior to wide application. The purpose of the studies was to asses the quality of diagnostic toolsperformances. The assesment comprises of four stages. First stage was to evaluate the performance of ELISA BM compared to Latex Agglutination Test (LAT) on sheep sera. Secondly, to evaluate the performance of ELISA BM to descriminate true seropositive andseronegative toxoplasmosis on human sera. The last stage were comparing ELISA BM, ELISA TL (commercial kit) and LAT on predetermined and unknown human sera. The results showthat the accuracy of ELISA BM is slightly better than ELISA TL. Agreement of ELISA BM with LAT was better againts ELISA TL with LAT. However, all performance as determined using Cohen’s ? and Gwet’s AC1 of ELISA BM, ELISA TL and LAT were good up to very good agreement.Keywords: Toxoplasmosis, ELISA, Latex Agglutination, inter reliability agreement.

Abstract

ABSTRAKPemilihan Tumbuhan Hutan Sebagai Sumber Pakan dan Tempat Bersarang Kuskus kerdil(Strigocuscus celebensis) dan Analisa Nutrien di Pulau Wawoni, Sulawesi Tenggara. Astudy on feed plants selection and nesting site of dwarf cuscus (Strigocuscus celebensis) wasconducted in Wawonii Island, South-East Sulawesi. The study was done in the lowland forestwith the altitude of 1 - 200 m a.s.l. The results showed 45 species of plants consisting of 23families was selected by dwarf cuscus as their feed resources and 11 species of plants as theirnesting site. Parts of the feed plants being consumed were fruit, young leaves, flower, andtopmost of trunk.Keywords: Forest plant, feed resources, nesting site, Strigocuscus celebensis Wawonii Island

Abstract

Isolation of antibacterial compounds from chloroform extract of belawan putih (Tristaniopsis whiteana) leaves hasbeen done. The bioautography analysis of the extract against Escherichia coli and Staphylococcus epidermidis,subsequently followed by isolation and purification of active compounds led us to obtain two pure compounds,compound 1 and 2. Futhermore, the compound 2 was identified as betulinic acid based on their spectroscopic dataincluding ultraviolet spectrum, infrared spectrum, 1D- and 2D-NMR spectra. In addition, this is the first report ofthe occurrence of this compound in the plant of Tristaniopsis whiteana and the genus of Tristaniopsis.Keywords: antibacterial, active compound, leaves, Tristaniopsis whiteana

Abstract

ABSTRACTInfectious Bronchitis Virus (IBV) is an acute viral and highly infectious disease which attacks the respiratory system of chicken. The impact of economic losses is very large since it decreases the egg production and itcauses the abnormal of egg shapes, decreased of body weight and death. In Indonesia, the IBV disease is still a serious problem in poultry regarding to many variants which arise from mutations of the IB virus. The decreasing possibility of the vaccination effectiveness due to incompatibility of strains used towards the virus circulated in the field. The variants of IB virus have not been well-documented in Indonesia since the lack of characterization of this virus. In this study, we characterize the IB virus isolates obtained from our local outbreaks in the field in order to determine the variant of IB virus in Indonesia and provide recommendations for the improvements of IB vaccine seed viruses which is adapted to IB virus circulating in the field. The IB virus characterized in this study shows close genetic relationship with the IB virus from Taiwan and China. However, it did not correlate with the local IB virus that has been found previously.Keywords: Identification, Character, IBV, Indonesia

Abstract

AbstrakEmisi Gas Metana pada beberapa Varitas Padi di Lahan Sawah Tadah Hujan.Keragaman emisi gas metana dipengaruhi oleh karakteristik pada varitas tanaman padiyang berbeda umur, sifat, dan aktivitas sistim perakarannya. Penelitian lapang dilaksanakandi lahan sawah tadah hujan pada musim penghujan 200312004 dengan tujuan untukmengetahui emisi gas metana dari beberapavaritas padi unggul yang dibudidayakan secaragogorancah dan untukmengetahui varitas tertentu yangmampu mengemisi metana rendahsekaligus memberikan hasil gabah tinggi. Percobaan disnsun menggunakan rancanganacak kelompok dengan 3 ulangan pada 7 varitas padi, yaitu IR64, Cisantana, Sintanur,,Way Apoburu, Dodokan, Silugonggo, Mentik (varitas lokal). Pengambilan contoh gas dilapangan dilakukan dengan sungkup dan contoh gas diinjeksikan ke alat kromatografigas dengan detektor FID untuk menetapkan fluks metana. Hasil penelitian menunjukkanbahwa emisi gas CH, dari tanaman padi varitas Way Apobum < Dodokan < Silugonggo< Cisantana < Sintanur < lokd Mentik < IR64. Potensi daya hail varitas lokal Mentiklebih rendah daripada IR 64 dan IR 64 lebiirendah daripada varitas Silugonggo, Cisantana,Way Apoburu, Dodokan, dan Sintanur. Varitas Way Apoburu yang dibudidayakan dengansistem gogorancah mengemisi gas metana paling sediit dan memberikan hasil gabahpaling tinggi.Kata kunci : Emisi metana, varitas padi, sawah tadah hujan

Abstract

ABSTRACTThe growth and regeneration of langkap in natural habitat is very fast and has invasive tendencies. The aim of study was to analyze the population and population structure of langkap in Leuweung Sancang Nature Reserve, and build spatial models of habitat suitability langkap in Leuweung Sancang Nature Reserve. Data were collected using a sampling method with a number of plots along the transect line. Langkap has become the dominant species in Leuweung Sancang Nature Reserve on saplings and pole with population structure shows an normally structure population. Analysis of spatial modelling on habitat suitability of langkap through binary logistic regression with independent variables in the form of the Forest Canopy Density (FCD), slope, distance from the river and Normalized Difference Moisture Index (NDMI) has been able to predict habitat suitability of langkap in Leuweung Sancang Nature Reserve, i.e. 61,10% of the Leuweung Sancang Nature Reserve suitable as habitat langkap and 38,90% of the Leuweung Sancang Nature Reserve is not suitable as habitat langkap. Keywords: Langkap, Arenga obtusifolia, habitat suitability, spatial modeling, Leuweung Sancang Nature Reserve.

Abstract

ABSTRACTErythrocyt and Haemoglobin on Cave Bat at Gombong Karst Area, Kebumen Regency, CentralJawa. The purpose of this study was observe physiological adaptation of the cave batsconducted from September 2009 to March 2010 in twelve caves within the karst of Gombong,Kebumen Regency, Central Java. In each caves where the bats roosting, temperature, alsohumidity, oxygen percentage in the air, and ammonia content were measured. Three individualsamples were caught from every bat roost during the day when the bats were staying in theirroosts. Then the amount of erythrocyte was counted by hemocytometer and hemoglobincontent was measured using Sahlis method. The data were analyzed using RDA and multipleregressions. It was concluded that humidity, temperature, oxygen and ammonia correlatedsignificantly with erythrocyte and hemoglobin content. The amount of erythrocyte increasedby increasing humvel.Keywords: Bat, cave, erythrocyte, haemoglobin.

Abstract

ABSTRACTDiscrepancy of urease and phosphomonoesterase activities and its role in establishing Nand P nutrition in soil collected from Wamena Biological Research Station. Microbialactivities in soil lead to know for establishing soil nutrient status. Accordingly, soil collectedfrom Biological Research Station in Wamena then sent to the laboratory and determined on itsenzymatic activities and the physicochemical, as well. In this work, the enzymatic activities ofurease and phosphomonoesterase were examined in relation with soil microbial respiration, inorder to understand the mineralization of nitrogenous and phosphorus compound in soil. Soilrespiration rate (2.43-3.21 mg C02 g-dm12hour) designated variation in each sample, as wellas urease (8.6-23.5 unit/g soil) and phosphomonoesterase (5.5-7.9 unit/g soil) activities.Phosphomonoesterase activity showed strong correlation with respiration rate within soil; andreveal to the configuration of the bioactivities and physicochemical soil figures concluded thatthe B sample has the poor fertility. The phenomenon of data fulfill that bioactivities hadcorrelation with the physicochemical compound in the soil.Keywords: respiration, urease, phosphomonoesterase, Wamena Biological Research Station.

Abstract

"Cibinong Science Center LIPI or CSC’s LIPI as an artificial habitat to the bird species conservation, especiallywith their ecological parks. This research was to study the distribution patterns and dynamics of birdcommunities in the CSC’s LIPI areas during 2005 to 2009 and on 2014. Transect Line’s method was used inthis study. The results were 39 species of birds to live in the eight sites observation on 0.27 km2 areas.However, there are no significant effects, especially relationships between Margalef’s index value to area,cotton area closest distance to the river and their habitat situation. According to “multiple linear regression”analysis show that ?= 3.626-0.69 area -1.31 cotton area closest distance to the river + 0.94 habitat situation,with determinant coefficient value (R2) = 0.161. The densities of nine species were found high. Those specieswere Lonchura punctulata (D=49.37 individuals/ha), Zosterops palpebrosus (D=22.2 individuals/ha),Collocalia linchi (D=15.00 individuals/ha), Orthotomus sutorius (D=12.70 individuals/ha), Lonchura maja(D=12.48 individuals/ha), Passer montanus (D=12.19 individuals/ha), Pycnonotus aurigaster (D=7.37individuals/ha), Dicaeum trochileum (D=6.07 individuals/ha) and Streptopelia chinensis (D=5.85 individuals/ha). The diversity indices value (H’) 1.52 to 2.51 and the evenness indices value (E) 0.52 to 0.86. Thesimilarities indices value (SI) of birds in the CSC’s LIPI will be compare to another places, i.e. the IPBcampus at Darmaga, Bogor or Yogyakarta State University’s campus in Yogyakarta showed no different,relatively. The IS value were 58.82 to 61.54%. Birds in the CSC parks were high dominant to occure in thelower strata (27.78–43.18%) and upper strata (50.43-70.83%), while in the middle strata (1.39-9.15%) tend tobe slightly. Most activities were carried by birds , especially foraging and calls. Although, the birds in the CSCparks also breeding, take a rest etc . The CSC’s LIPI parks at Cibinong could be used as a reference to thebird’s artificial habitat model to others areas. That’s an important if the areas are industries complex or theCenter office of some government buildings. But, the composition and structure of the vegetation will bedeveloped in there should be to produce some natural food resources and safety well for breeding of birds.Keywords: bird species, ecological parks, artificial habitat, distribution patterns, dynamics communities
