Abstract

ABSTRACTSoil Actinomycetes Population, Enzymes Activity, and its Relation with Carbon and NitrogenContent, in Bukit Bangkirai, East Kalimantan. Bukit Bangkirai is one of the tropical forest inIndonesia have been exposed with intense forest fire. The affected forest is subjectivelydivided into three level of damages, heavily damage forest (HD), low damage forest (LD) andcontrol (K). The objective of this research was to observe the abundance of Actinomyceteswhich have important role in ecological process. Through decompotition of organic materialsand nutriens cycle. Actinomycetes were isolated and enumerated by SDS-YE method. CFU/ gsoil (x 104) in K, HD and LD are 41,86 ± 25,52, 16,09 ± 5,70 and 18,96 ± 4,19 respectively. Amylaseand cellulase were determined by DNS method. Carbon and Nitrogen total were determinatedby CN analyzer. The different of amylase, cellulase activities and abundace of Actinomycetesbetween HD, LD, and Control plot were not significant. However, carbon and nitrogen totalare different. LD plot has the highest carbon and nitrogen total, followed by control and HDplot. There has no significant different among plot observed may indicate microbial communitiesof soil in Bukit Bangkirai have been recovered.Key words : actinomycetes, fire forest, amylase, cellulase, nitrogen total, carbon total

Abstract

Ovarian cancer cell metastasis is induced by signaling pathway activated by the binding of type IV collagen to ?1 integrinreceptor on the surface of cancer cells and estrogen binding to estrogen receptor. However, the role of estradiol-17? andtype IV collagen on the development of ovarian cancer have not been clearly understood. Therefore, this research wasconducted to observe the differential gene expression in SKOV-3 ovarian cancer cells cultured on type IV collagen andtreated with estradiol-17?, and incubated for 12, 24 and 72 hours. Differential display RT-PCR was used to express thedifferential expression gene after treatment. cDNA fragment that expressed differentially was isolated and sequenced.Sequencing result on one of the cDNA fragment showed that PI3K is one of the gene expressed in SKOV-3 ovarian cancercell. To verify this result, cDNA was amplified using PIK3CA specific primer. The increasing level of PIK3CA is inducedby three kinds of receptor activities, those are c-erbB2 receptor bound to estradiol-17?, homodimer receptor of c-erbB2, andthe activity of integrin receptor bound to type IV collagen. The increasing level and activity of PIK3CA can also increasethe expression of c-erbB2 gene. In SKOV-3 cells cultured on type IV collagen for 72 hours, the increasing of PIK3CA andc-erbB2 expression level is very low. The conclusion is that estradiol-17? gives a more significant effect than type IVcollagen to induce the increasing expression of PIK3CA and c-erbB2 genes.Keywords: ovarian cancer, SKOV-3, type IV collagen, estradiol-17?, PIK3CA, c-erbB2

Abstract

ABSTRACTThis study aimed to quantify the population of Birgus latro in the Batudaka di Togean islands, Central Sulawesi. The research on robber crab was conducted in Batudaka Island, Togean, Tomini Bay, Central Sulawesi. In the study site, 21 plots measuring of 50x50 m2 were created bounded by raffia. Feed in the form of shredded coconut is placed in each plot in the afternoon. At night was performed observations and catchs. In the "base camp" every crab crab carapace caught measured in carapace length and weight. During the study, 277 crabs were caught, consisted of 173 males (62.45%) and 104 (37.55%) females. Based on the formula calculation of Schiller (1992) population figures obtained 821 803 ± 195 030 crabs in Batudaka Island. By regression analysis between carapace length with weight, it was found that the growth of B. latro is negative allometric, i.e., weight gain is faster than the increase length of carapace. The weight gain of female is slightly higher than that of the male. Whether male crab population or female equally composed of 9 age groups. This study showed that 66.7% of male crab and 29.1% of female crab has entered the market size.Keywords: Birgus, crab, carapace

Abstract

ABSTRACTThe Habitat Characteristic of the Critically Endangered Turtle the Sulawesi ForestSulawesi Turtle Leucocephalon yuwonoi in Ganonggol and Bangkir Rivers in CentralSulawesi. The Sulawesi Forest Turtle (Leucocephalon ymonoi) is an endemic, criticallyendangered and listed in the appendix I1 of CITES. Captive breeding is needed toreintroduce the species into the original habitat. Captive breeding success includes acomprehensive knowledge on the biology and ecology of the species. The objective ofthis research is to investigate the habitat character of the turtle respectively vegetation ofthe riverbank, physical and chemical conditions ofthe water. Field surveys were conductedon the Moutong area from 1 l until 19 October 2004 and on the Bangkir area from 20until 26 October 2004. Vegetation data were obtained by developing four quadrate at theturtle habitat in each locality and were analyzed using Curtis method. The results shownthat the habitat of Leucocephalon yuwonoi is transparent streaming water, neutral, nottoo high dissolved oxygen, water height not more than 50 cm, substrat varies from bigstones, small stones, clay loam and sandy loam. The area was covered by vegetation thatcan provide food during all year round, such as Colocasia esculenta, Limnocharisflava,and Ipomoea aquatics.Key Word: Leucocephalon yuwonoi, habitat, vegetation, water conditions.

Abstract

Spodoptera exigua nucleopolyhedrovirus (SeNPV) is an entomophathogenic virus of S. exigua larvae commonly usedas bioinsecticide. This research was aimed to 1) study the symptoms of SeNPV infection on the S. exigua larvae inthe laboratory, 2) examine the virulence of SeNPV on the 3rd instar of S. exigua larvae and (3) find out the optimalconcentration of polyhedra and harvesting time. The infection of SeNPV on the S. exigua larvae was inhibitedmolting process and disturbing larval growth. The color of infected larvae gradually changed become more dark,and at the end of infection, larvae died with fragile and broken integument. Infected larvae showed reduction infeeding activities. The LC50 of SeNPV on 3rd instar larvae in the laboratory was estimated 6.65 x 105 POBs/ml.The polyhedra concentration used for virus propagation was 5.88 x 106 POBs/ml. The optimal harvesting time was5 days after inoculation, where most of the infected larvae had died but the body still intact.Keywords: Pathology, symptom, virulence, Spodoptera exigua, SeNPV

Abstract

ABSTRAKProduktivitas Fosfatase Asam pada Bacillus sp. yang Diisolasi dari Tanah Hutan TamanNasional Gunung Salak. Pada pengamatan ini dilakukan karakterisrik bakteri pelarut fosfatyang diisolasi dari tanah hutan Taman Nasional Gunung Salak. Sebanyak 21 koloni hasil isolasidiuji terhadap produktivitas enzim fosfatase berdasar pelarutan media mengandung fosfat.Isolat yang terkuat melarutkan fosfat diidentifikasi sebagai Bacillus sp. Pada pengamatanlanjutan terhadap strain teruji dilakukan penumbuhan pada media cair selama 90 jam inkubasi,dan hasilnya ternyata mampu melarutkan fosfat inorganik (Pi) dari sumber trikalsium fosfat(Ca-Pi) dan alumunium fosfat (Al-Pi) masing-masing pada kisaran 1,2 sampai 152 dan 0.8 sampai25 mg.L-1; dan menunjukkan aktifitas enzim fosfomonoesterase antara 0.2 sampai 1.01 unitpada media yang mengandung larutan para-nitrophenylphosphate sebagai media fosfatorganik (Po) artifisial. Konsumsi glukosa pada media yang diukur selama pertumbuhan sejalanpula dengan produk ortofosfat sebagai akibat adanya aktifitas enzim fosfatase. Peningkatanfosfatase juga sejalan dengan bertambahnya biomassa sel bakteri dan penambahan produkasam glukonat. Penurunan pH dari 7 menjadi 5 diakibatkan peningkatan produk asam glukonatdi dalam media tumbuh. Bakteri pelarut fosfat yang berasal dari tanah hutan Taman NasionalGunung Salak dapat memproduksi fosfatase asam untuk memineralisasi sumber-sumber fosfatmenjadi sumber nutrisi yang siap digunakan oleh akar tumbuhan, dan itu merupakan prediksikuat untuk menjadikan isolat bakteri pelarut fosfat sebagai sumber bahan pupuk hayati.Kata kunci: Bacillus sp., tanah hutan, fosfatase asam, Ca-Pi, Al-Pi.

Abstract

ABSTRACTThe rich of Indonesian biodiversity becomes an attraction for local and foreign researchers to conduct research in this country. One of scientific productivity indicators that mostly used is number of publication in scientific journal. The previous studies had never analyzed scientific productivity of Indonesian biodiversity researchers, both institutionally and individually. This paper analyzes 2,432 articles concerning Indonesian biodiversity research affiliated with institution from Indonesia (published in 1990-2015) from Scopus database, including 8,544 authors from 1,638 institutions. Data pre-processing resulted publication year, number of author, and author’s institution location. Then, they were analyzed using scientometrics analysis. Scientometrics analysis is a quantitative method related to measurement of the science communication. The result shows that internation-al collaborations produced most of scientific publications on Indonesian biodiversity. The number of scientific publication on Indonesian biodiversity was significantly increase and showed exponential growth trend. Indo-nesian author mostly wrote articles from Animalia kingdom. Multi and mega authors wrote the most of scien-tific publications on Indonesian biodiversity.Keywords : Indonesia, biodiversity, scientometrics, productivity, scientific publication, Scopus

Abstract

ABSTRACTResearch aimed to determine effect of genotype x environment interactions (GEI) on agronomic characters and yield stability of soybean genotypes were conducted at eight locations, namely Ngawi, Probolinggo, Banyuwangi, Jember, Malang, Sleman-Yogyakarta, Rembiga and Sesela-West Lombok. Materials used in the study were nine soybean genotypes consisting of six promising lines and three improved cultivars (Malabar, Wilis, and Ringgit). A randomized complete block design was used in each location, repeated four times. Observations were made on flowering days, maturity days, plant height, number of filled pods, seed weight per plant, 100 seed weight and seed yield. Results showed that the effect of GEI was significant on all of the observed agronomic characters. Maturity days, filled pods number, seed weight per plant, and seed yield were predominantly influenced by environmental factors, plant height and 100 seed weight were predominantly influenced by genetic factors, whereas flowering days was influenced by GEI. Genotype I/M-824-19 was stable with yield average lower compared to that of all genotypes and Agl/I-1114-8 was stable with yield average higher than the yield average of all genotypes. Wilis and M/IB-895-2 were classified as unstable, but gave yield average higher than the yield average of all genotypes.Keywords: adaptation, promising lines, soybean, stable

Abstract

Produksi biomasa gugur serasah dan variasi musimannya dimonitor selama 5 tahun untuk mengamati polamusiman dari gugur serasah dan kontribusi gugur serasah terhadap ekosistem hutan. Gugur serasah dikumpulkansetiap bulan dengan menggunakan 20 perangkap serasah (luas masing-masing perangkap serasah adalah 1 x 1 m2)yang berada pada petak permanen. Seluruh sampel di oven pada suhu 70 ºC. Total gugur serasah adalah 8,36 ±0,39 t ha-1 tahun-1, dengan konstribusi terbesar pada daun (6.55 ± 0.44 t ha-1tahun-1), diikuti oleh komponen yangtidak teridentifikasi (0.69 ±0.2), batang kecil (0.76 ± 0.1), bagian reproduksi (0.16 ± 0.06), and batang besar (0.21± 0.04 t ha-1 tahun-1). Pola musiman menunjukkan bahwa gugur serasah meningkat selama musim hujan. Gugurserasah daun tertinggi berada pada bulan Juli. Jenis-jenis dominan menggugurkan daun utamanya pada pertengahanmusim kering. Total gugur serasah daun dari yang tertinggi ke yang terendah diamati Nauclea lanceolata (0.36 ±0.16), Maesopsis eminii (0.25 ± 0.11), Schima wallichii (0.09 ± 0.02), Pternandra azurea (0.02± 0.01), dan Dyxoxylumdensiflorum (0.01 ± 0.01 t ha-1 tahun-1).Kata kunci: Biomassa, Gugur serasah, Hutan dataran rendah, TN. Abstract

ABSTRACTAntagonistic Interaction between Lactobacillus 5NS and Candida tropicalis. Centrifuged supernatant of medium where Lactobacillus 5NS grown inhibited growth of three strains of Candida tropicalis. The Inhibition activity was detected using paper disc method. When the supernatant was induced with living cells of C. tropicalis Y393 strain, it inhibited the tested yeast strains. Inhibition activities of this supernatant against C. tropicalis Y390, Y391 and Y393 were equivalent to that of phenol at the concentration of 20.56 , 25.02 and 25.04 mg/ml, respectively. However when the supernatant was induced with heated or living cells of Candida tropicalis Y390 and Y391, it did not show any inhibition activity on all yeast strain tested. Thus the data indicates that there may be a specific interaction between Lactobacillus 5NS and C. tropicalis Y393 in which under the induction of specific certain yeast such as C. tropicalis Y393, Lactobacillus 5NS produces a toxin-like that is not spesific for the yeast target. Such interaction between normal flora and patogenic yeast may be happen in the digestive tractus.Key words: Lactobacillus, yeast, Candida tropicalis, antagonist
