Abstract

ABSTRACTMillet [Setaria italica (L.) P. Beauv.] is one of the potential cereal crops to be developed in supporting food diversification program. The purpose of this study was to investigate the response of millet to low light (shade) condition. The experiment was conducted in the field with nested design that consisted of two factors that are, 1) Accession from Buru Island which are yellow seed and red seed; and 2) Shading nets that consisted of 0%, 40%, and 70% shade. Shade factor was designed as main plot, while accession factor was designed as sub plot. Growth (growing percentage, plant height, leaf number, leaf area, and plant dry weight), yield (panicle length and panicle weight), photosynthetic pigments (chlorophyll a, chlorophyll b, and carotenoid), and stomata density of the millet were observed. The result showed that the growth and yield of both accession were decreased by the increase of shading. However, yellow seed millet tended to be more tolerant than red seed millet with optimizing photosynthetic capacity through enhancing leaf area, leaf longevity, and enhancing antenna pigments such as chlorophyll b and carotenoid.Keywords: Growth, millet, photosynthetic pigments, shading, yield

Abstract

ABSTRACTAmylase is one of the most important industrial enzymes which can be used in a number of industrial processes including food industry, textile, paper industry, renewable energy, and pharmaceutical. This study reports the environmental conditions and nutrient media for amylase production from marine bacterium Arthrobacter arilaitensis. Various parameters such as substrate concentration, pH medium, temperature of fermentation, co-substrate, and nitrogen source was determined to obtain the optimum conditions. Maximum amylase enzyme production was obtained at starch concentration 1%, pH 7, temperature fermentation 30°C, co-substrate maltose with activity 2.7 U/mL. While the addition of several nitrogen sources was given decreased amylase activity, such as addition casein was decreased the activity into 2.3 U/mL. Keywords: Arthrobacter arilaitensis, amylase activity, optimization, co-substrate, pH, temperature, substrate, nitrogen source

Abstract

ABSTRACTStevia rebaudiana is plant species producing natural sweetener with has low calories. The species propagation could be done by tissue culture tecnique to obtain propagules with high quality and sustainability. Modification of media composition and in vitro environment will increase growth and vigority of explants so that they have high survival rate during acclimatization. The aim of this research was to increase growth of stevia shoot culture by increasing the level of vitamins in combination with different type of test tube covers. Stevia shoot tips were cultured on MS medium containing normal concentration of its vitamins (control treatment; Myo-inositol 100 mg/l; Nicotinic acid 0.5 mg/l; Pyridoxine-HCl 0.5 mg/l; Thiamine-HCl 0.5 mg/l and Glycine 2 mg/l), twice and 4 folds of vitamin levels, they were grown on culture tubes with Al-foil and ventilated-plastic with filter (2 cm diameter and pore size at 0,22 micron). Height of shoots, number of nodes, number of leaves, number of roots were observed every week till 8 weeks of culture. Biomass (fresh and dry weights) and chlorophyll level and acclimatization were done 8 weeks of culture. The results showed that type of culture tube covers affected significantly to all growth parameters, biomass as well as level of chlorophyll, meanwhile level of vitamins only affected number of nodes, shoots and roots. Interaction between vitamin level and covers types only occured for height of shoots and number of roots. Plantlets grown on medium containing 4 fold of vitamin level (Myo-inositol 400 mg/l; Nicotinic acid 2 mg/l; Pyridoxine-HCl 2 mg/l; Thiamine-HCl 2 mg/l and Glycine 8 mg/l) with ventilated-plastic cover had larger leaves compared to other treatments. All plantlets survived in a greenhouse.Keywords: Stevia rebaudiana, in vitro growth, increase in vitamin concentration.

Abstract

Advertisement calls of Microhyla palmipes and Oreophryne monticola have never been described before; howevercharacteristics of M. orientalisâ??s calls has been described just to support new species publication. The advertisement calls oftwo individual males of M. orientalis and M. palmipes which originated from Wongaya Gede, Bali were recorded in July2010 at air temperatures of 26.0°C. Three individuals males of O. monticola were recorded in Eka Karya Botanical Garden,Bali at air temperature of 25.0°C. Call components were obtained from 65 calls of M. orientalis, consisting of 95 pulses; 10calls of M. palmipes, consisting of 113 pulses; 3 calls of O. monticola, consisting of 127 pulses, which were then analyzed toobtain the characteristics of sound waves by using software of Adobe Audition 3.0. Sound waves of M. orientalis mainlyconsists of two dominant frequencies raging from 1873-2062 Hz and 3375-3562 Hz; however sound waves of M. palmipesand O. monticola mainly consist of only one dominant frequency that ranges of the frequency is the same, i.e. 3000-3937Hz. Although M. palmipes and O. monticola have the same range on dominant frequency, but the two species have significantlydifferent on number of period per pulse, pulse duration, pulse interval, pulse period, call duration and call interval.Key words: frog call, Microhylidae, Bali Island

Abstract

ABSTRACTThis study was aimed to determine the structure and genetic characteristics of the Javan Parakeets (Psittacula alexandri alexandri) in some populations in Java Island based on ND2 gene sequences. This study used 26 birds composed of 9 birds from Bogor (West Java), 8 birds from Cepu/Blora (Central Java), 5 birds from Yogyakarta (DIY), and 4 birds from Ngawi (East Java). Blood samples were taken from each bird and DNA was extracted, and then a single fragment of ND2 gene was PCR amplified. A totaly of 1020-bp of DNA sequence of all individual birds were analyzed and produced low genetic distance between bird populations: 0.0031 ± 0.0010 (0.0024 ± 0.0012 to 0.0058 ± 0.0022). The highest genetic distances among individual bird were found in the Yogya?s population followed and were lowest in Cepu/Blora?s population. There were 16 haplotype sequences (Hpa Hpa1-16) found in 26 birds. There were some identical haplotypes in each population, but each population has different haplotypes from each other. The highest haplotype diversity and nucleotida diversity were found in Yogya?s and Bogor populations. Abstract

ABSTRAKParameter Kinetika Endokitinase dari Bacillus licheniformis MB-2. Bacilluslicheniformis MB-2 yang diisolasi dari air panas di Tompaso (Sulawesi Utara)menghasilkan 5 enzim kitinase ekstraselular. Salah satu enzim, Chi-67, telah dimumikandan diarekterisasi. Chi-67 dapat menghidrolisis substrat kitin buatan maupun alami.Analisis kinetika aktivitas enzim terhadap berbagai substrat kitin telah dilakukan. Enzimdapat memotong 4-methylumbelliferyl N', N'-diacetylchitobioside {MUF(GINAc),}, 4-methylumbellifelyl N:N' N'-triacetylchitotrioside {MUF(GINAc),), glikol kitin, dankoloidal kitin, tetapi tidak dapat melepaskan senyawa MUF dari 4-methylumbelliferylN-acetyl-R-D-glucosamine {MUF(GINAc)}. Ini menunjukkan Chi-67 memiliki tipepernotongan endo. K u ~kaec epatan reaksi substrat menunjukkan pola profil Michaelis-Menten. Kecepatan reaksi maksimum (V,,) terhadap MUF(GlNAc), identik denganVmatxe rhadap MUF(GINAC)~ya itu 0.02 pM.mitil . VmaeXn zim terhadap koloidal kitinadalah 0.01 pM.h-' atau lebih rendah 3 kali dari nilai VmaXpa da substrat glikol kitin (0.03M h ) Nilai Kcu, untuk koloidal kitin sebesar 8.01 h-' atau hampir 2 kali lebih tinggidari glikol kitin (4.32 h-I) sementara Kc, untuk MUF(GlNAc),adalah 3.65 min-' atau1,5 kali lebih tinggi dari Kc%,M UF(GINAc), yang hanya 2.90 min-' . Afinitas Chi-67terhadap koloidal kitin, glikol kitin, MUF(GINAc),, dan MUF(GINAc), yang ditunjukkanoleh nilai konstanta Michaelis (K,) masing-masing 3.08 rng.ml-I, 0.32 mg.ml-I, 0.26 mMdan 0.1 mM.Kata kunci: Bakteri, endokitinase, enzim, dan stabilitas panas

Abstract

Thirteen plant species of main vegetation alliances were detected upon their preferences on various abiotic factors,among them, six species had preferences on either soil or topographical factors, while four species only had preferenceson soil factors, and one species only on topographical ones. Abstract

Apis koschevnikovi: Distibusi di Kalimantan Selatan dan Variasinya pada Mitokondria Cytochrome b. Variasi mitokondria berdasarkan Cytochrome b (Cyt b) pada Apis koschevnikovi di lakukan di tujuh lokasi di Kalimantan Selatan Indonesia. Di koleksi 29 koloni A. koschevnikovi yang diperoleh di kawasan hutan primer dan sebagian koloni tidak dijumpai dihutan bekas penebangan. Abstract

ABSTRACTNitrite Oxidation by Heterotrophic Bacteria Under Aerobic Condition. The nitrite transformingactivities of heterotrophic bacteria from isolates from agriculture soil, Lampung were studiedunder aerobic conditions. Among the 9 bacterial isolates tested, almost all are reported haveability to consume of nitrite, but none of the bacterial isolates formed significant nitrate in themedium. NOB H1 (Bacillus licheniformis), is denitrification-negative, consumed 16. 4 mg/L ofnitrites with the accumulation of 4.45 mg/L nitrates. While, NOB H8 (Pseudomonas sp.) isdenitrification-positive, consumed 49. 64 mg/L of nitrite with the accumulation of 3.34 mg/Lnitrates. Nitrite oxidations of both isolates NOB H1 and NOB H8 took place during stationeryphase to the dead phase. Growth pattern of both isolates NOB H1 and NOB H8 were sigmoidwith generation time of 1.69 and 2.19 hour, respectivelyKey words: heterotrophicbacteria; nitrite oxidation; denitrification
