Abstract

Coxiella burnetii belongs to rikettsia group living obligate intracellularly and as the agent of zoonosisQ fever. Enrofloxacine is an antibiotic in quinolon group used to treat infection of C. burnetii in chicken,goat, calve, pig, dog, cat,  and horse. From ruminant practical experience, enrofloxacine if combined withBioATP  can enhance the enrofloxacine activity. Research for the effecivity of enrofloxacine and BioATP totreat C. burnetii has never been carried out. The research was conducted to explore effect of enrofloxacinewith supplementation BioATP against C. burnetii. Enrofloxacine pharmacokinetic study was carried outby using simental beef as an experimental animals. The effectivity of BioATP supplementation onenrofloxacine activity to treat C. burnetii was tested by using Vero cell tissue culture. The results showedthat combination of enrofloxacine and BioATP increased kinetic profile of enrofloxacine in term of onset,duration, pharmacology intensity, and bioavailaibility. Enrofloxacine had activity to treat C. burnetii withvalue of minimal inhibitory concentration (MIC) at 1-2 ppm and value of minimal bactericidal concentrationat 4 ppm. Supplementation of BioATP improved the effectivity of enrofloxacine in treating C. burnetii.

Abstract

The objective of this study was to determine sperm membrane stability after sexing with gradientalbumin (egg white) using andromed and CEP-2 supplemented with 10% egg yolk. The observations ofsperm membrane included membrane integrity, uncapacitated, capacitated  and acrosome reacted. Albumingradient was made by mixing egg white with andromed or CEP-2 supplemented with 10% egg yolk resultingin a concentration of egg whites 10%, 30% and 50%. Membrane integrity was observed using HOST(Hypoosmotic Swelling Test), while uncapacitated, capacitated and acrosome reacted sperm  were observedusing CTC (Chlortetracycline) staining. The results showed that both andromed and CEP-2 supplementedwith 10% egg yolk could protect membrane integrity, uncapacitated, capacitated and acrosome reactedsperm. Andromed and CEP-2 supplement with 10% egg yolk could reduce the demage of membrane integrityand uncapacitated sperm, whereas capacitated and acrosome reacted sperm could be kept low.

Abstract

Campylobacter spp cause infection in human in the form of gastroenteritis and Guillain-Barre syndrome.Campylobacter jejuni is one of  Campylobacter spp responsible for about 90% of cases of Campylobacteriosisin human with gastroenteritis.  Efforts to prevent the incidence of Campylobacteriosis in humans should bestarted with an assessment on its level of poultry. This study aims to develop a diagnostic test for C. jejunion chicken using immunochromatographic serological method. Stages of the research consist of thepreparation of test card, antigen preparation, optimization of the antigen and serum. The results showedthat immmunochromatographic card can distinguish infected serum from the uninfected.The minimumamount of antigen  was found to be 1,5 ng/µl. It needs 10 µl serum to perform the test properly.

Abstract

Koi herpesvirus (KHV)  is the cause of  a lethal disease that affects common carp (Cyprinus carpio)  andkoi (Cyprinus carpio koi). Although, it has been reported that common carps could act as carriers for KHV,their histopathologic findings, especially the gills  have not been identified up to now. In the present study,12  normal, healthy looking common carps including their gills were collected from the endemic area ofKHV in  Sleman, Yogyakarta. All fish were necropsied and the gills were collected and fixed in 10% bufferformalin. Then, the gills were processed histopathologically using routine hematoxyline-eosin stain andexamined under the microscope. Histopathologic examination of the gills exhibited an apparent infiltrationof inflammatory cells, especially lymphocytes. The large oval to polygonal basophilic cells containing largeintranuclear inclusion bodies were also identified. Gills epithelial cells show mass hyperplasia and adhesionwith necrotic changes. Thus, results of this study has led to a reasonable conclusion that KHV infection ispresent in the normal, healthy common carps. One possibility is those KHV  are live viruses (carriers forKHV) and might could act as a source of infection is being discussed.

Abstract

Lactic acid bacteria (LAB) have an ability to enhance innate responses in fish. The aim of this studywas to study the effect of three LAB isolates (Kerapu Situbondo Usus: KSBU 12C, KSBU 5Da, and KSBU9 on non-specific immune responses of tiger grouper (Epinephelus fuscoguttatus). The fish were fed dietsupplemented with each of the bacterial isolates at final concentration of 108 CFU mL-1 twice every day.The haematology parameters (haematocrit, leucocyte count, heterophils/neutrophils, monocytes,lymphocytes), phagocytic activity and total LAB were measured every seven days. After 28 days, the fishwere challenged with Vibrio alginolyticus. During the feeding period, the haematocrit values of fish fed withLAB isolates were higher than those of control group, and values ranged from 24.67±8.33 % to 40.50±7.09%. The leucocyte counts increased, mainly after being challenged with V. alginolyticus, and the valuesranged from 40.33 ±6.03 × 1000 cell mL-1 to 139.33±15.57 × 1000 cell mL-1. With the exception oflymphocytes, proportion of heterophils and monocytes, and phagocytic activity were higher in fish fed withLAB isolates than those of control. The presence of LAB in the gastrointestinal tract of fish fed with LABresulted in higher haematology parameters and phagocytic activity in comparison to fish fed control diet.In conclusion, supplementation of LAB in diet could enhance non-specific immune response of tiger grouper.

Abstract

This study aimed to determine the effect of different methods of saving the storage by using a waterjacket and a free water jacket to quality semen Limousin cattle after being stored at a temperature of 5°C.Semen used the criteria motility e” + + collected two times a week. Semen evaluated macroscopically andmicroscopically and diluted in the ratio 1: 1 using the Cauda Epididymal Plasma (CEP-2) diluent added10% egg yolk. Stored in the refrigerator temperature 5°C and semen quality  was observed immediatelyafter dilution, 1 hour, 2 hours, 3 hours, 24 hours, 48 hours and 72 hours. The results showed that themethod of free storage waterjacket better in maintaining the percentage of motility, viability, abnormalityand plasma membrane intact spermatozoa

Abstract

The objective of this study was to analyze the influence of chitosan on coronary artery wall thicknessand lumen diameter of Sprague Dawley rats induced by high fat ration. The animals for this study weretwenty adult male rats divided into four groups, i.e. group I as the control was fed with basal rationcontaining normal fat for 3 months, group II was fed ration containing high fat for three months, group IIIwas fed ration containing high fat and given chitosan 180 mg per kg body weight per day orally in 2 mLaquadest for 3 months, group IV was fed ration containing high fat for three months and after one monthgiven chitosan 180 mg per kg body weight per day orally in 2 mL aquadest for two months. Each groupconsisted of five animals. After 90 days, the rats were necropsied and the hearts were collected tohistopathological. The difference between treatments was statistical analysis by Anava, and continued byLowest Significant Difference Test to locate the difference. Normal fat ration did not influence coronaryartery wall thickness and coronary artery lumen diameter in rats. High fat ration increased coronaryartery wall thickness and chitosan could be decreased coronary artery wall thickness in rats. High fatration + chitosan that given simultaneously could be increased coronary artery lumen diameter in rats.Based on the result of this study, it was concluded that high fat ration was a major factor able to causeatherosclerosis and chitosan was able to prevent atheroma plaque formation.
