Abstract

ABSTRACTPollution in the marine environment caused by industrial activity, marine transportation, offshore mining and oil refineries, one of which comes from petroleum contamination. One of  efforts to prevent and to manage oil pollutionwas by the use of oil degrading bacteria. The objective of this research was to examine degradation ability of marine bacteria isolates and identify the bacterial isolate based on 16S rRNA sequence. The research was conducted from March to May 2018. The oil degradation analysis was conducted at Laboratory of Marine Microbiology,Faculty of Fishery and Marine Science, while the molecularidentification of bacteria was done in Genetics Laboratory, Faculty of Mathematics and Natural Sciences, University of Riau. The results showed that five isolates of oil degradation bacteria, selected(BM 1d, BM 7b, BM 7c, BM 14a and BM 14b) were able to degrade crude oil at different concentrations. The highest degradation at 1% concentration is indicated by BM 14b isolate, at 2% concentration was indicatedby BM 1d isolate and at 3% concentration was indicated BM 14b isolate. From the degradation analysis results, three isolates (BM 1d, BM 14a, BM 14b) have the highest degradation ability to determine the type of oil degrading bacteria. The molecular identification results by the 16S rRNA sequencing and BLAST analysis showed that BM 1d isolate were similar to Bacillus sp. (95%), BM 14a isolate were similar to Bacillus flexus (92%) and BM 14b isolate were similar to Bacillus cereus (98%). Keyword : Oil Degradation Bacteria , Crude Oil, 16S rRNA, Bacillus

Abstract

ABSTRACTSoft coral extract of Sinularia sp. can inhibit pathogenic bacteria Escherichia coli, Staphylococcus aureus, and Pseduomonas aeruginosa with strong inhibitory power up to very strong inhibitory power. The purpose of this study was to determine the inhibition of Sinularia sp. extract against E. coli, S. aureus, and P. aeruginosa bacteria. This research was conducted in March - April 2019. Sampling was carried out in the waters of Tamiang Island, West Pasaman Regency, West Sumatra. The method of this research is the experimental method and the method used to test antibacterial activity is the agar diffusion method.Soft coral extract of Sinularia sp. very potential to inhibit pathogenic bacteria. The highest clear zone in E. coli bacteria with a concentration of 100% is 25.67 mm, the highest yield of S. aureus bacteria at a concentration of 100% with a yield of 22,05 mm,and the highest yield of P. aeruginosa bacteria at a concentration of 25% with a result of 24.62 mm. Clear zone increases in proportion to the increase in concentration, but in P. aeruginosa bacteria is inversely proportional, because the highest yield is obtained at a concentration of 25%. Keywords:Antibacterial Activity, Pathogenic Bacteria, Sinularia sp. Abstract

ABSTRAKThe research was conducted from November 2018 until May 2019. The purpose of this study was to determine the effect of T. hemprichii seagrass extract on the sensitivity of V. alginolyticus and A. hydrophila bacteria. The research used experimental method. Sensitivity test was carried out in the Marine Microbiology Laboratory of the Faculty of Fisheries and Marine, University of Riau. Based on the, sensitivity test positive control (K+) indicated hishest inhibitory zone diameter against V. alginolyticus  andA. hydrophila39,57 mm and 42,11 mm, respretively. While there was not clear zone found in the negative control (K-). Extract of T. hemprichii had the hishest inhibitory zone against V. alginolyticus  andA. hydrophila at a concentration of 100%, these were 2,45 mm and 2,58 mm, respectively. The lowest 10% indicated the lowest inhibitory zone against V. alginolyticus (0,65 mm) and A. hydrophila (0,38 mm).Key words: Extract, T. hemprichii,Sensitivity, V.alginolyticus and A. hydrophila,

Abstract

A research on the production, decomposition rate and identification of bacteria on Avicennia alba litter in the coastal zone Kuala Indragiri, Riau Province. This research was aimed to observe the contribution of mangrove forests in the productivity for surrounding environment and conducted from December 2014-March 2015. The litter production was calculated by using litter-trap. Sample was collected once in every 15 days during 45 days of observation. The litterfall was oven-dried and weighed. The result showed that avarage production of litterfall of A. alba in all station was 297.696 g/m2/45days (fresh weight) and 189.763 g/m2/45days (dry weight). The average total shrinking dry weight of litterfall leaf /day was at 0.13225 g/day or 1.3225 %/day. The average reduction dry weight of A. alba litterfall in every station was 7.848 g during 45 days of observation with a percentage value of the decomposed litterfall weight of 78.480% or 0,523 g/day, with final result of litterfall dry weight is 2,152 g. Seven types of bacterial colonies were identified on each station and consisted of Bacillus sp (2), Micrococcus sp, Xanthomonas sp, Pseudomonas sp, Vibrio sp and Escherichia sp.Keywords: Avicennia alba, litter, production, decomposition rate, bacteria, Kuala Indragiri

Abstract

ABSTRACT Sungai Guntung has a potential for crabs because it still has fairly good mangrove forests in several areas. One of the problems found in the crab catchwas high mortality rate caused by disease and ectoparasites. This research aimed to determine the types of parasites that attack crabs and to determine the prevalence and intensity of ectoparasite attacks on crabs precisely and systematically. The field survey was conducted from April-May 2019 in Sungai Guntung waters, Kateman District, Indragiri Hilir Regency with sampling techniques from several fishermen to inventory ectoparasites on crabs.The observations found that ectoparasites include phylum protozoa namely Zoothamnium sp, and from arthropod phylum is Octolasmis sp.The prevalence value of ectoparasites infested Scylla serratawas Zoothamnium sp. 2.27%, Octolasmis sp. 80%, a mixture of Zoothamnium sp and Octolasmis sp. 6.67%. The intensity value of ectoparasites infested Scylla serrata was Zoothamnium sp. 9.5 ind/crab and Octolasmis sp. 149.23 ind/crab.Kata kunci :Ectoparasites, Crab, Protozoa, Prevalence, Intensity

Abstract

Probiotic is an alternative subtance to prevent fish diseases which is environmentally friendly. The research purpose was to isolate potential probiotic to control bacteria pathogen in barramundi (Lates calcarifer) to control the bacteria pathogen. The probiotic bacteria candidates were isolated from intestine of barramundi by using the spread and streak methods on MRS and TSA media. The antibacterial activity was examinined by using disc diffusion agar method on Zobell 2216E medium. The results showed thirty eight isolates were found which consisted of coccus, white, orange and yellow colonies. Twelve isolates had inhibition against Vibrio alginolyticus,two isolates showed highest inhibition zone and antivibrio activities. The highest inhibition zone (10.04 ± 1,48 mm) and (9,50 ± 2,12 mm) and the antivibrio activities (4.04 mm) and (3.50 mm) were indicated by KPBU2-5 and KPMU2-6 isolates respectively.Keywords: Probiotics, Isolation, Barramundi, Antagonistic.

Abstract

Heterotrophic bacteria in marine ecosystems have important as decomposer of organic material into the elements essential. This group of bacteria can be found in a variety of habitats including marine sediment. This research aims to isolate the heterotrophic bacteria and the activity against pathogenic (Vibrio alginolyticus, Aeromonas hydrophila and Pseudomonas sp). The research was conducted from November until December 2016. Samples were obtained from the waters of Tanjung Medang, District of North Rupat, Riau Province. Isolation and antagonistic test were conducted at the Laboratory of Marine Microbiology and Biotechnology Department of Marine Sciences Faculty of Fisheries and Marine. In this research was a survey method where heterotrophic bacteria from the sediment were isolated on marine agar medium by pour plate and streak quadran techniques. Activity of the heterotrophic isolate against bacterial pathogens was conducted using diffusion agar on Mueller Hinton Agar (MHA). The number of heterotrophic bacteria ranged from 4,4 × 106 up to 13,5 × 106 CFU/g.Twelve isolates of the sediment bacteria were selected for the antagonism test. However, only nine isolates (Isolate A, C, E, F, O, S, V, W, Z) were able to inhibit the growth of pathogenic bacteria. One isolate (E) performed the highest ability to inhibit the growth of patogens (V. alginolyticus, A . hydrophila and Pseudomonas sp).Keywords: Isolation, Heterotrophic, Sediment, Antagonism, Pathogenic Bacteria.

Abstract

This research was conducted in November-December 2016 in the coastal waters of Dumai, Riau Province. This research aims to determine the growth and survival rate of seeds of mangrove shells P. expansa in natural habitats with different densities. This research uses experimental method. This research consists of one treatment that is with stocking density of three levels of 20, 25 and 30 individuals / container. The container used is a pond made of net (2 mesh size) with a size of 50x50 cm. The results showed that the best growth was found in the treatment with the density of 20 individuals / containers. Also, the best survival rate was found in the treatment of 20 individuals / containers of 98.6% size.Keywords : Polymesoda expansa,seeds Growth and Graduation rate, Stocking Density, Dumai.

Abstract

Oil degradation bacteria can be found in various habitats in the marine environment including in the waters around the harbor. This study aims to isolate the oil degrading bacteria from waters around the port of Bengkalis and test its ability to degrade crude oil. Isolation of the degrading bacteria has been done from February to April 2017 at the Marine Microbiology Laboratory, Department of Marine Science, Faculty of Fish-eries and Marine University of Riau. The amount of oil degrading bacteria ranges from 0.2 x 105 to 8.9 x 105 CFU/ml. The isolation result was obtained by 10 isolates of oil degradation bacteria, ie BM1a, BM1b, BM1c, BM1d, BM7a, BM7b, BM7c, BM7d, BM14a and BM14b. Identification based on morphological, physiological and bio-chemical characteristics of each isolate, all isolates were round and elevated, six were white in color, three were yellow, one cream-colored isolate, eight isolates with slip-pery edges and two isolates with periphery waves, seven isolates were indol positive, two isolates produced H2S, four isolates were methyl red positive, all isolates were motile, capable of fermentation of glucose and sucrose in sugar test, able to ferment citrate, positive catalase and Gram negative bacteria. For degradation test only five bacterial isolates were selected of BM1a, BM1b, BM1c, BM7a and BM7d. The highest degradation at 1% concentration was indicated by isolate BM1a, the highest degrada-tion at 2% and 3% concentrations was indicated by BM1c isolate. Therefor, bacteria isolate capable of degrading the highest crude oil was BM1c.

Abstract

Grouper fish has a fairly high economic value, especially in Asiancountries, namely Hong Kong, Singapore, Taiwan and China. High marketdemand. On aquaculture, is one important component of fish feed that plays animportant role in the cultivation. Application of probiotics in aquaculture fish feedhas not been done yet in the Fisheries sector. The research was conducted inOctober 2008, Grouper is used as the intestines and stomach samples were takenas a source of probiotics, whereas pathogenic bacteria obtained from thecollection of marine microbiology laboratory of Faculty of Fisheries and MarineRiau University. From the isolation of bacterial strains found 11 strains that havethe potential as probiotics, then proceed to the molecular test by using rDNA16 s.Which is obtained two strains of probiotic bacteria Bacillus cereus and the speciesof bacteria Strenotrophomonas maltophilia. The test results found that theantagonist Bacillus cereus isolates to inhibit the growth of pathogenic bacteriaAeromonas sp and two species of Vibrio sp. While the bacteriaStrenotrophomonas maltophilia showed no antagonism against pathogens.
