Abstract

Research was done to elucidate the role of Ca2+ intracellular in stomatal movement induced by auxin. Two types of auxin (IAA and NAA) increased stomatal opening and the increase was concentration dependent. The addition of EGTA, Ruthenium red and Procaine as the modulator of Ca2+ concentration , inhibited stomatal opening induced by auxin. EGTA and procaine significantly inhibited somatal opening at 100 uM and 1 mM. Ruthenium red worked at lower concentrations which were 10 uM, 50 uM and 100 uM on LAA-induced stomatal opening, while on NAA-induced stomatal opening, Ruthenium red had an effect at 10 uM, 50 uM, 100 uM dan 1 mM. It is suggested that Ca2+ involves in stomatal opening induced by LAA and NAA as signaling agent.

Abstract

The purpose of this study conducted to know variety of : long of axis polar, equatorial plane diameters, and P/E index of ten Kembang Sepatu (Hibiscus rosa-sinensis L.) with differnt colour flowers. Acetolysis method and 1 % Safranin staining used to make pollens slides, micrometri were used to measured long of polar axis and diameter of pollens. The result of this research showed : pollen structure have: 90,10 ± 3,02 ^im to 117,42 ± 1,37 l^m axis polar long, 89,66 ± 3,13 |^m to 112,92 ± 0,94 (xm equatorial plane diameter, and 0,99 to 1,04 P/E index. Based on these data is indicated that the type of the pollen are prolat sferoidal (except bud type red corolla, and white flower) polypantoporat type appertura and periporat ornamentation exine .

Abstract

Alas Purwo National Park is one of representative tropical low rain forests at eastern Java. There are many natural attractive sites and historical legend, which lead to, famous for ecotourisme activities. There are also rich of flora and fauna that some of them endemic and endangered species. Medicinal plant is one important material on the forest, which still unexplored more. Ten sampling transects were established in some sector of Alas Purwo for inventory of wild medicinal plants. The result shows that at least 5 species have been categorized as an endangered plant. Meanwhile, the structure and composition of endangered medicinal plants is not better than common medicinal plants.

Abstract

Ten isolates of fungus were isolated from oil palm stem at oil palm plantation in Medan All of them were tested its abilities to degrade lignin. The results showed that one of them was able to grow on ligninase media and the fungi has the ability to degrade ligin. The fungi is identified as Melanotus sp.

Abstract

The disturbance of spermatogenesis after exposure with 2,5-HD (2,5-Hexanedione) can be observed through the increasing of death cell in apoptosis mechanism, that can be observed through the expression of Bax protein. Therefore, it had been conducted a research to study the expression of Bax protein against white mouse of Wistar strain exposured with 2,5-HD. A number of 56 white mouse were divided randomly into 8 groups, each group consists of 7 white mouse, two groups were used as pretest, three groups used as control treatment, and three groups used as treatment were exposured with 2,5-HD of 0,06 mg/WW during 2, 4, 6 week. The changes in the Bax protein expression observed in the slices of cross section of testis using imunohistochemical method. The research result shows that the longer duration of 2,5-HD exposure causes the more increase the expression of Bax protein at the sertoli cell and the germinal. The expression of Bax protein compared to the control groups increases into 132%, 112%, 108% and 123% respectively for the sertoli, spermatogonial, spermatocyt, and spermatid cells in the treatment were exposured with 2,5-HD during 6 weeks. It is needed a further research to complete information of apoptosis mechanism as the result of 2,5-HD exposure, by study the ratio of Bcl-2/Bax expression as the antiapoptosis and proapoptosis proteins.
