Abstract

Detection of Ocurrence Citrus Vein Phloem Degeneration (CVPD) Disease on Citrus Plant with Comprehensive Symptoms using Polymerase Chain Reaction (PCR) Technique Citrus Vein Phloem Degeneration (CVPD) is one of very serious disease of citrus plant. The disease is caused by Liberobacter asiaticum bacteria and transmitted by Diaphorina citri Kuw insect. The outspread of disease can also caused by the use of citrus seedlings which have been infected by CVPD. Preliminary visual observations CVPD disease symptoms, there are two types of plants showing symptoms of partial and systemic. Partial symptoms are not all the leaves of plants showing symptoms CVPD, while the systemic symptom is on the whole leaves of plants showing symptoms CVPD. This research aimed to determine the outspread of L. asiaticum bacteria in every parts of the plant that made detection by PCR. The research has been conducted from in Batukaang, Bangli and Plaga, Badung then continued at Laboratory of Genetic Resources and Biology Molecular, Udayana University. Results of PCR visualization using 1% agarose gel showed that there were a DNA band at 1160 bp on the leaves, twigs, branches and trunks. Because the size of 1160 bp DNA bands is expression by L. asiaticum, then the citrus leaf samples were detected positive for the L. asiaticum and it can be ascertained that L. asiaticum haas been distributed to all parts of the plant.

Abstract

Land Suitability Evaluation of Horticultureand Plantation Commodities in the Kerta’s Village Payangan Subdistrict Gianyar District Judging from their natural conditions Kerta’s Village Payangan Subdistrictpotential in developing horticultural crops (pineapple, banana, orange, and bark,) and plantations (robusta coffee). The purpose of this study are (1) determine the suitability of land several commodities (pineapple, banana, orange, and bark) and plantations (robusta coffee), (2) determine the limiting factor horticultural commodities (pineapple, banana, oranges, and bark) and plantations ( robusta coffee), (3) create a map of the land suitability of horticultural commodities (pineapple, bananas, oranges, and bark) and plantations (robusta coffee). This study uses a survey of the land suitability evaluation by Djaenudin and others (2003). Based on the results of delineation, there are 6 units of homogeneous land. Actual Land Suitability horticultural crops (pineapple, banana, orange, and bark,) ranged from S2, S3 and N while the suitability of potential land to plant pineapples, oranges, and the bark is on land units IVHPKC, VHPKC, IVHPTG, VHPTG is S1 and to plant banana on land is S2. On land units VIHPKC and VIHPTG for banana plants belonging to N by a factor limiting the slopes. For Robusta coffee classified as S1 on all land units except on land VIHPKC and VIHPTG.

Abstract

Effect of Thickness Plastic Low Density Polyethylene to Shelf Live of union Left (Allium fistulosum L.) The research is to determine effect treatment of thickness polyethylene plastic and to determine level of thickness polyethylene plastic which are suitable against shelf life of left union. The Research conducted in Laboratory of Post-Harvest Technology Faculty of Agricultural Technology Udayana University in March 2016. The research used a completely randomized design (CRD) with five treatments, that are (P0) treatment control, (P1) the treatment use plastic thickness 0.02 mm, (P2) the treatment use plastic thickness 0.03 mm, (P3) the treatment use plastic thickness 0.04 mm and (P4) the treatment use plastic thickness 0.05 mm. The observation done for 5 days. The results showed that plastic thicknes of the polyethylene most real significant effect against change of the CO2 concentration, weight loss, leaf chlorophyll and shelf life left union during storage at a temperature of 160C. Treatment of polyethylene plastic packaging 0.04 mm (P3) shown to maintain the quality and extend the shelf life until 5 days with the characteristics of the stem and the leaves begin to wilt, yellow spots more and more 30%-40% on leaves and disability of 40%, compared with leek without treatment (P0) were only able to survive until 3 days with the characteristics increasingly withered stalks and leaves, the more yellow spots, the leaves yellow spots > 50% and dry leaves, defects of more than 50%.

Abstract

Effect of Plant Utilization Limiting Fringe Against Population Pests and Natural Enemies On Planting Cabbage (Brassica oleracea l.)   The purpose of the study is  to determine the effect of plant utilization Limiting Fringe barrier against pest species diversity, species diversity of natural enemies, pest species abundance, and abundance of natural enemy species on cabbage crop. The research was conducted in two places items, namely in the field and in the laboratory. Sampling is done diagonally plant. Pest species diversity on both compartments consisting of six species of Aphis brassicae, Crocidolomia pavonanaFab,Plutella xylostella L,Spodoptera litura Fab, Helicoverpa armigera Hubner, and Chrysodeixis orichalcea. There are three species that have a more dominant population abundance namely A. brassicae, P. xylostella and C. pavonana. Species diversity of natural enemies of equal treatment, both in the parasitoid diversity consists of a single species Diadegma semiclausum.e. whereas in the diversity of the species there are six predator Menochilus sexmaculatus, Ischiodon scutellaris, Paederus fuscipes Curt, Dolichoderus bituberculatus, Oecophylla smaragdinaand Sycanus sp. The results Showed an Increase in the percentage of the natural enemies of the moment map of plant cabbages planted with crops in the suburban area of the Cabbage Cropping.

Abstract

Chrysal Use to Extend Vaselife Cut Flowers (Rosa hybrida L.) RosesThe demand for cut roses increasing but the problems that often occur in old rose cut flowers is the vaselife of flowers very short. One way that can be done to prolong the vaselife of flowers adaah by giving a solution of the marinade. The main function of a marinade solution is to maintain the vaselife of cut flowers as long as possible. Chrysal. Chrysal a marinade solution containing sucrose, dextrose and citric acid that can extend the life of flowers (vaselife) Another important factor in maintaining the vaselife of flowers is the availability of water. Water will go faster if the wider cross-section, for example by cutting the plant stem oblique. Objective To know the concentration of the shredded material Chrysal optimized to produce a maximum vaselife of flowers. Knowing the difference in the vaselife of roses stalks in pieces tilt and horizontal concentration Chrysal know the interaction between the treatment of cuts rose to the vaselife of roses. This research was conducted at the Laboratory of Post Harvest Technology Faculty of Agriculture, University of Udayana. This study uses a completely randomized factorial design with two factors. Based on this research, interaction between concentration Chrysal (L) by cutting the flower stalk (A) does not affect significantly (p ? 0.05) of all observed variables. In the single factor cutting flower stalk treatment (A) significant (p> 0.05) to variable interest weight loss, dantotal solution is absorbed, whereas the concentration of Chrysal (L) a very significant effect on the flowers begin to bloom, longer vaselife, and the total solution is absorbed.

Abstract

Detection of CVPD (Citrus Vein Phloem Degeneration) Disease Using PCR Technique of Citrus from Untalan Sub Village, Jungutan Village, Bebandem District, Karangasem Regency This research aims to know about presentation of CVPD disease on citrus plants and detect CVPD disease using PCR technique with specific primer called 16S rDNA. This research was conducted at Laboratory of Genetics Resource and Biology Moleculer, Udayana University. Result for visual monitoring of presentation in CVPD symptom in four locations is 25%, 37%, 27%, 19%, and the average is 27%. The result for average the presentation of CVPD symptom in the shoot of citrus in each locations is 7.9%, 15.73%, 8.9%, and 7.07%. The isolation of leaves citrus DNA were appeared on 1% agarose in electrophoresis showed DNA band. The result of DNA amplified that were appeared on 1% agarose in electrophoresis showed DNA bands 1160 bp. 3 samples from 4 with specific symptom of CVPD gave positive PCR reaction. DNA Bands 1160 bp is expression by Liberobacter asiaticum, than the sample from Untalan Sub Village were detected positive for the Liberobacter asiaticum and the samples were infected by CVPD disease.

Abstract

Mikropropagation Plant Strawberries (Fragaria sp.) Through Induction of Organogenesis Problems for farmers strawberry in Bali are the difficulty of get healthy strawberries  seeds a with prices are relatively cheap. Micropropagation method is ways to multiplay seeds for solving the problem. Research aims to understand the influence of concentration ZPT NAA + BAP to organogenesis on explant leaves strawberries in vitro. Experiment arranged using a Randomized Block Design (RBD). Medium the base used is MS medium with adding of 2 gL- 1 of activated charcoal, and 0.1 ppm NAA. As the treatment is BAP with concentration of 5 ppm, 10 ppm and 15 ppm with 5 replications. The results showed that the use of 0,1 ppm NAA + 15 ppm BAP most effective in deflection (3,0 DAP) with the percentage of 90,00%. The use of the NAA 0,1 ppm + 15 ppm BAP also most effective in a time of swelling (7,6 DAP) with the percentage of 82,95 % , while only the use of the NAA 0,1 ppm + 5 ppm BAP that will be capable of eliciting shoots at 23 DAP time.

Abstract

Detection Of The Existence of Liberobacter Asiaticum in Citrus Stricken with Sympthoms of Partial CVPD Citrus Vein Phloem Degeneration (CVPD) is a major disease of citrus caused by Gram-negative bacteria Liberobacter asiaticum. The symptoms of the disease were varied due to many factors such as type of pathogenic races of the pathogen, types of citrus trees, and the presence or spread of plant pathogens in the plant. This study aims to detect the existence or distribution of pathogens of CVPD in citrus plant with partial symptoms. Samples were taken from the bone leaves, bark twigs, bark of branches and bark of stem whether symptomatic or non symptomatic. Sampling was carried out in Plaga village, Badung Regency, Bali and analysis of polymerase chain reaction (PCR) was conducted in UPT Lab Genetics Resources and Molecular Biology, Udayana University. The results of this study indicate that the amplified DNA of 1160 bp was found in the first plant that was  symptomatic leaf, twig bark of leaf symptomatic and non symptomatic leaves. While on the second sample of citrus plants, DNA fragments were found on the leaves and symptomatic leaf and non symptomatic, and they were not found in other parts of plant, namely twig bark, branch bark, bark of stem. These results indicate that in citrus with partial symptoms the distribution of pathogens L. asiaticumwere only partial in plant.

Abstract

Detection of the Presence of Citrus Vein Phloem Degeneration (CVPD) Disease Causing in Molecular on Citrus (Citrus nobilis Lour var. microcarpa Hassk)which Based on Variation of Chlorosis Symptoms This research was aimed to identify the presence of Liberobacter asiaticum which caused of Citrus Vein Phloem Degeneration disease on citrus leave with different chlorosis symptoms. This research was conducted at Laboratory of Genetics Resources and Biology Molecular, Udayana University. The leaves sample were measured the chlorophyll content using a chlorophyll meter and then were identified in molecular using Polymerase Chain Reaction (PCR) technique, using primer O11 and O12 which will amplified the specific fragmen of 16S rDNA. The result for chlorophyll content indicated that the leaf samples were containing 26.80, 36.52, 36.62, 40.74, 44.56, 47,06, 58.44 and 65.16 SPAD.DNA amplification showed that the leaves which contained chlorophyll at 58.44 and 65.16 SPAD didn’t show DNA bands with size 1160 bp, and leaves which contained chlorophyll at26.80, 36.52, 36.62, 40.74, 44.56 and 47.06 SPAD showed DNA bands 1160 bp. Therefor the DNA bands 1160 bp is expression by L. asiaticum, then the citrus leaf samples were detected positive for the L. asiaticum and the samples were positive infected by CVPD disease, so that the chlorophyll content of citrus leaves at ?47.04 SPAD can be used as basis to diagnosis CVPD disease on citrus plants.

Abstract

Performance Analysis of Watershed Indicators Based On Land Use and Discharge of Water in DAS UndaWatershed (DAS) is an integral ecosystem elements consist primarily of natural resources, water and vegetation and human resources as users of those resources. Unda watershed allegedly decreased function, such as the decline in forest area, increasing critical lands and known to potentially flood. This research was conducted in order to deternine Unda watershed response to changes of use of land in the years of 2006-2013 and the water discharge conditions. DAS performance indicators used are the Precentage of Vegetation Cover (PVC) and Coefficient of Flow Regime (CFR). Method of research is descriptive comparative view of the two parameters, that is the use of land and water discharge. The results showed the prencentage of vegetation cover in the medium category ranges from 55,07% and deceased by 1,87 in 2013. Indicator coefficient of flow regimes gained 67,66 value before it is published Plan of Integrated Watershed Management (2006-2009) included in the medium category and scores 18,8 after being issued Plan of Integrated Watershed Management (2010-2013) with both categories. CFR suspected impairment is not only influenced by the value of the percentage of vegetation cover, but is also influenced by othe factors, such as climate, soil type, vegetation type, the growth phase, vegetation density, how management and others.
