Abstract

Isolation and identification of arbuscular mycorrhizal fungi (AMF) in cashew plants was carried out in Datah Village, Abang District of Karangasem Regency. This study aims to determine the diversity of mycorrhizal spores in the cashew rhizosphere and AMF infections in the roots of cashew plants. The result in microscopic observations found 5 genera of AMF spores such as Glomus, Racocetra, Entrophospora, Septoglomus, and Acaulospora and the population of the genus Glomus were found most dominant in the rhizosphere of cashew plants. The result of staining root with trypan blue obtained the internal structure of AMF in the form of vesicles, arbuscules and hyphae found in root tissue of cashew plants. The result of AMF DNA amplification with primer NS31 and AML2 showed 560 bp bands in electrophoresis with 1% agarose gel determined AMF infection in root of cashew plants.

Abstract

The research was coducted at Laboratory of Biotechnology Faculty of Agriculture Udayana University. The purpose of this research were to investigate moleculer detection of CVPD pathogen in its vector, other phytophagous insects on citrus cv, Siam and kemuning (Murraya paniculata L. Jack.). The result of this research showed that adults and nymphs of Diaphorina citri Kuw. could transmit CVPD disease, but the pathogen of CVPD could not be transovarially transmited. As a host of CVPD disease , kemuning could be source of infection. The adult of Toxoptera citricidus Kirkaldy and Maleuterpes dentipes Hell. could not potensial transmited CVPD disease.

Abstract

Bulung Sangu (Gracilaria spp) as many other macroalgae, is a source of beneficial phytochemical for health. Phytochemicals in macroalgae are widely used as active ingredients in cosmetic preparations. The study was aimed to determine the antioxidant properties of Bulung Sangu formulated in topical preparation. Bulung Sangu was macerated in ethanol 96% following by extract formulation in cream preparation in two concentrations: 5% and 10%. Antioxidant activity was determined against DPPH radical, compared to ascorbic acid. Percentage of reducing power in five concentration series (10, 20, 30, 40, and 50 µm/ml) statistical analyzes using one-way ANOVA with 95% of confidence level, following by IC50 measurement. Absorbance was read in wavelength of 517 nm. Concentration of 5% and 10% cream extract exhibited IC50 of 13,091 µg/ml and 11,867 µg/ml, respectively. These values are lower than IC50 of ascorbic acid (18,505 µg/ml), indicating the strong antioxidant properties of Bulung Sangu extract cream which is more potential than ascorbic acid. Bulung Sangu exhibit a potential source for being developed as medicine and cosmetics in topical preparation.

Abstract

Antioxidant is a chemical compound that could give one or more electrons to inhibit the free radical reaction. One of the plants that potentially produce antioxidants is purnajiwa (Euchresta horsfieldii (Lesch.) Benn.). The research was conducted using the fruit of purnajiwa. The purpose of this study is to know antioxidant activity from purnajiwa fruit using the DPPH method and measured by UV-Vis spectrophotometry. The extract of purnajiwa fruit gained by the maceration method using 96% ethanol solvent. The results of phytochemistry screening found out that the extract contains flavonoid, alkaloid, saponin, tannin, and terpenoid. According to flavonoid testing using quercetin, the results obtained from purnajiwa extract contained flavonoid of 1,651 mg QE/g. The activity test of antioxidants quantitatively by DPPH using UV-Vis spectrophotometry found out that purnajiwa extract has IC50 value of 57,28 ppm so that purnajiwa extract has strong antioxidant activity that could be used as natural antioxidant resources.

Abstract

The study focused on improving the fruit quality supported by handling of the cultivation, harvest and post-??harve st handling such as fruit packing, transportation, storage, and others, as well as marketing strategiesand regulations that are ne eded locally, so local Bali fruits can compete better in the Bali tourism market. The purposes of this study were to find some problems face by the local fruits to e nter the tourism marke t inBali. The local fruits studied were citrus, salacca, mangoes, mangosteen, banana, and papaya. This study wasthrough multiple surveys such as, inte rviews with farme rs, whole salers, fruit suppliers, and the manage rsof the hote ls and restaurants. The results of this study found some local fruits have e nte red the tourismmarket in Bali, in addition, local people in Bali te nd to consume the imported fruits for some culturalceremony, events and others. The results of this study showthat the main problems in the local fruits suchas citrus and banana were harvesting, packaging, storage, and transportation. The laboratory studies foundthat some microbes and inse cts infe cted local fruits due to the lackof handling such as packaging, storage, and transportation. The findings of this study was local fruits such as orange s and bananas face manyobstacles ranging from harvest, post-??harvest management, transportation, and the marketing strategies. Many problem has found on bananas such as damaged on storage, as well as citrus fruits. Improvements tothe appearance of the fruit has been tried to do by the application of emulsion of he rbal oils, oleic acid, wax, twee n, and the results showe d the fruits still in a fre sh condition afte r four weeks of treatme nt while theuntre ated fruits were rotten due tothe infe ction of microbes. The blackspots of insects ormicrobes infectionson the treated fruits can be eliminated as well. Some inse cts and microbial attacks were found that leads tofruit rot which can essentially be removed by tre atment with antimicrobial tre atment. This research tries tochart the effects on the competitiveness of local fruits, such as continuity of production, standardization andregulation which could be developed at the local level in the village community.
