Abstract

Vanda tricolor Lindl. var. suavis is an Indonesian wild orchid that has been rare in nature,  so it needs attention to take care and conserve them.  The objective of the research was to investigate the effect of coconut water and / or tomato juice on the growth of protocorm of V. tricolor Lindl. var suavis from Bali grown in vitro. The experiment was laid out in the factorial design, with two factors (coconut water/CW and tomato juice/TJ), each contained three concentration (CW: 0, 100, and 200 cc L-1; TJ: 0, 100 and 200 g L-1) resulted in nine combination of treatments and replicated four times.  The results showed that tomato juice with concentration of 100 gL-1 or 200 gL-1 promotes growth of protocorms of Vanda tricolor Lindl. var. suavis from Bali regardless the presence of  coconut water. Keywords: coconut water, in-vitro, protocorms, tomato juice, Vanda tricolor

Abstract

Vanda tricolor  Lindl. var. suavis  is an Indonesian wild orchid which is now extremely rare in nature due to its habitat  destruction.  Development  of  an  appropriate  method  for  improving   Vanda  orchid  through  genetic modification could be valuable for horticulture and, indirectly, also for conservation. In this research, a method of  Agrobacterium-mediated transformation of two  Vanda  tricolor  obtained  from  Salak Mount, West Java and merapi Mount, Yogyakarta in Indonesia protocorms was improved using acetosyringone (AS). Concentrations of 0 and 25 ppm AS were used in transformation of pG35S binary vector containing kanamycin resistance gene into V.  tricolor  protocorms.  The  result  showed  that  25  ppm  AS  was  required  on  innoculation  with Agrobacterium  solution, without AS on cocultivation. Five week s after  treatment  on the 300  ppm kanamicyncontaining medium, green protocorms were obtained, that  was  11.01% for  V.  tricolor  from Salak Mount with pre-culture treatment prior to innoculation, 9.39% for  V.  tricolor  from Merapi  Mount with pre-culture treatment prior to  innoculation,  and  1.37%  for  V.  tricolor  from  Merapi  Mount  without  pre-culture  treatment  prior  to innoculation. The  best  condition  to  set  high  efficiency  of  transformation  is  pre -culture  protocorms  prior inoculation, soaking protocorm on 25  ppm  AS for 30 minutes, then cocultivate its on AS-free  callus induction medium Key words: Vanda tricolor, Agrobacterium, orchid protocorms, acetosyringone

Abstract

This research was conducted at Tissue Culture Laboratory of Agricultural Faculty, Udayana University, from May to October 2009. The aim of this Study to acquired the best in vitro medium that was to added plant growth regulator. The results showed that plant growth regulator kinetin significantly affected to germination and protallus formation of ata fern. The M1 nedium treatment the fastest of germination formation was 19,33 day after showing (das) while the protallus formation the fastest to M3 medium was 36,17 das. In description the fastest sporofit formed to founded in M3 too (122 das).

Abstract

The aim of the current research was to find out a method in producing calli of Vanda tricolor orchid for target on Agrobacterium-mediated transformation.  Six to seven months (after sowing) of orchid seedlings were used as source of planting material.  Seedlings were sliced into small parts i.e. leaves, shoots and roots.  Roots were discharged, while leaves and shoots were used as explants. Explants then were planted on the Murashige and Skoog Medium added with 2ppm 2,4-D for calli induction.  Two kinds of sugar concentration i.e. 0% sucrose and 3% sucrose were used as treatment and each was added on those calli induction medium.  The results showed that the appropriate method for producing calli on V. tricolor orchid was using shoots as source of explants and planting them on Murashige and Skoog Medium added with 2ppm 2,4-D and with no sucrose.  Keywords: calli, V. tricolor orchid, shoots, leaves, sucrose

Abstract

ABSTRACTSThe aim of the research was to determine seed germination and growth of protocorm of  V.   tricolorBali of 5 month- age and 7 month- age pods in their response to tomato extract. Five kind of concentrations of tomato extract were trialed, i.e. 0, 50,   100, 150, 200, 250 g L - 1.  Observation was done by counting the number of protocorms for each stage of growth  at 4 weeks after seed sowing.  The study concluded that seeds of V. tricolor  Lindl. var. suavis forma Bali from both of 5 month- age and 7 month-age pods required tomato extract for their germination and  for the growth of protocorms.  Concentration of 150 g  L- 1tomato extract was the most suitable for seed germination and growth of protocorms for seeds from 7 month- age pods, however, growth of protocorms was still linear up to 250 g L-1tomato extract for seeds from 5 monthage pods.Key words : age of pod  , germination, protocorm, tomato extract,  V. tricolorABSTRAKTujuan  dari  penelitian  ini  adalah  untuk mengetahui  respon  perkecambahan biji  dan  pertumbuhan protokorm anggrek V.  tricolor  Bali dari umur buah yang berbeda terhadap pemberian ekstrak tomat. Bahan tanaman yang digunakan adalah buah anggrek  V. tricolor  forma  Bali umur 5  bulan  dan 7 bulan setelah polinasi.  Perlakuan terdiri dari  lima   konsentrasi ekstrak tomat yakni 0, 50, 100, 150, 200, 250 gram L -1, diulang 3 kali untuk masing- masing umur buah. Observasi dilakukan pada 4 minggu setelah semai dengan menghitung jumlah protokorm untuk masing-masing fase. Hasil penelitian menyimpulkan bahwa biji anggrek  V.  tricolor  Lindl.  var.  suavis  forma  Bali,  baik  dari buah  umur  5  bulan  maupun  7  bulanmembutuhkan  ekstrak  tomat  untuk perkecambahan  biji  dan  pertumbuhan  protokorm.  Fase  protokorm mencapai nilai  tertinggi  pada  konsentrasi  ekstrak  tomat  150  g  L-1untuk  buah  umur 7  bulan,  namun pertumbuhan protokorm masih linier sampai konsentrasi ekstrak tomat 250 g L-1untuk buah umur 5 bulan.Kata kunci : ekstrak tomat, embrio, umur buah, V.  tricolor

Abstract

Vanda tricolor Lindl. var. suavis adalah spesies anggrek alam asli Indonesia yang di habitat asalnyasudah mulai langka karena kerusakan hutan. Pengembangan metode transfer gen untuk tujuan modifikasigenetik diperlukan baik untuk perbaikan sifat-sifat tanaman maupun konservasi. Metode transfer genmelalui Agrobacterium tumefaciens untuk anggrek V. tricolor belum ada protokolnya, sehingga perludikembangkan. Penelitian ini menggunakan protokorm Vanda tricolor Lindl. forma Bali yang berumur8 minggu setelah semai sebagai target transformasi. T-DNA membawa gen KNAT1 yang dikontrololeh promoter CaMV dari Cauliflower Mosaic Virus serta membawa gen NPTII, yakni gen ketahananterhadap antibiotik kanamisin untuk seleksi transforman. T-DNA ini dikonstruksi dalam vektor binerPgreen dan ditransfer ke genom tanaman melalui A. tumefaciens stranin LBA4404. Deteksi transgendilakukan dengan PCR untuk mengamplifikasi fragmen gen KNAT1 sepanjang 1200bp dengan primerspesifik KNAT1. Tujuan dari penelitian ini adalah: 1) mengetahui peran vitamin C pada transformasianggrek V. tricolor dalam mengatasi browning (pencoklatan); 2) menentukan pada tahapan mana(inokulasi dan atau kokultivasi) acetosyringone perlu ditambahkan dalam proses transformasi; dan 3)menentukan perlakuan yang menghasilkan persentase kandidat transforman dan frekuensi transformasitertinggi. Hasil penelitian mendapatkan bahwa aplikasi vitamin C mencegah terjadinya browning padaprotokorm anggrek V. tricolor yang ditransformasi meningkatkan persentase protokorm hijau setelahseleksi transforman. Persentase kandidat transforman dan frekuensi transformasi tertinggi diperoleh jikaacetosyringone diaplikasikan pada tahap inokulasi dan kokultivasi, kemudian diberi perlakuan vitaminC setelah kokultivasi.Kata kunci: Aggrek V. tricolor, Agrobacterium, Acetosyringone, vitamin C

Abstract

This study aimed to determine the genetic diversity and fruits quality of the "Jeruk Bali" cultivars grown in Bali. This research was conducted in all regencies and city in Bali, during 2016. Furthermore, several cultivars of “Jeruk Bali” were genetically analyzed based on RAPD markers using 10 primers. Analysis of the quality of fruit is based on physical properties and chemical content. Eighteen cultivars of "Jeruk Bali" obtained have a fruit morphological character with round, short round, and piriform fruit shapes, which are red, pink, cream and white flesh color. RAPD analysis results at 53% similarity level are grouped into 5 groups. The first group was only one cultivar, the second group consisted of 13 cultivars, the third and fourth groups were only one cultivar, while the fifth group consisted of two cultivars. The analysis of the diversity between cultivars based on the combination of physical and chemical properties of the fruit with hierarchy method on similarity level about 85% in a group is obtained by 4 (four) groups. Groupings by combination of physical and chemical properties of the fruit are not synchronized in their entirety with dendograms based on their genetic diversity. This illustrates the physico-chemical properties of “Jeruk Bali” fruit in general is not fully genetical expressed, but also influenced by conditions of environmental growth.

Abstract

A research regarding “Runner-tip culture of strawberry (Fragaria x ananassa Duch) Grown on Several Shoot-induction Medium” has been investigated. The objective of the research was to find out the most suitable medium for shoot production from runner-tip culture of strawberry at establishment step of micropropagation. The research was laid out in a Completely Randomized Design, 4 treatments of medium type for shoot induction and 10 replication, each was represented by one (1) bottle with 6-8 explants. The treatments were summarized as follows: T1 = MS ; T2 = MS + 2 ppm BAP + 0.01 ppm NAA; T3 = MS + 1 ppm of TDZ; T4 = WPM + 2 ppm BAP + 0.01 ppm NAA. The parameters observed were days of the bud emergence, the average number of shoots per explant, and the average number of leaves per explant. It can be concluded that among medium used in the current research, the medium of MS added with 1 ppm thidiazuron (TDZ) was the most suitable medium for shoot production of strawberry from the explant of runner-tips. The treatment was resulted in the earliest time of bud emergence, and producing the highest number of shoots and leaves.

Abstract

From explorations in several parts of the forest of Bali, 24 species of Dendrobium orchids were identified. Relationship among these species needed to be mapped for further development of orchids. Characteristics of leaves anatomy have been used to determine relations among plants. Characteristics of anatomy that were observed in this study from the paradermal and transversal cut of the leaves were shape and number of epidermis cells, location of stomata, the direction of stomata opening, shape and number of stomata, length of the epidermis and stomata, density of stomata, index of stomata, and thickness of mesophyll. Analysis of relations among species was displayed as a dendogram, using the software Minitab 17 Vis. Analysis of relationship among 24 species of Dendrobium Bali based on the leaves anatomy used grouped average method in squared Euclidean distance with the scale of 0.349 to 0.972. At Euclidean distance of 0.349, two clusters were formed where D. aloifolium and D. subulatum were separated from the other clusters. At 80 % similarity coefficient, seven clusters were formed using similar method, with the 3rd and 5th clusters had the shortest distance (169.35). If species that were clustered together were cross-bred, the success rate was higher; meanwhile, the further away the relation between species, the smaller the success rate of cross breeding. The possibility of getting high-quality genotypes was higher when the cross-breeding was successful.
