Abstract

Exploration of Cellulolytic Bacteria from Organic Waste in DenpasarMunicipal waste is one of the problem that must be overcome in Denpasar City. Municipal waste volume of Denpasar City 2014 was 1,247,769 m3, Year 2015 was 1,294,696 m3, and 2016 was increased to 1,296,438 m3. Total of waste that produced by Denpasar City is 70% organic waste. This type of organic waste has a major component of lignocellulose consisting of three polymers namely, cellulose, hemicellulose, and lignin. Cellulose is a component of plant cell walls. Cellulose has a low digestibility so it takes a long time to be degraded. One of the microorganisms that can produce the enzyme is the cellulolytic bacteria. The purpose of this research was to find and identify isolate of cellulolytic bacteria isolated from organic waste of Denpasar City. This research was conducted from September to 2017 until March 2018. Organic municipal waste samples were taken from each District in Denpasar City each one sample. Bacteria from the organic waste was isolated dan identified by moleculer identification in order to know the cellulolytic bacteria spesies. Based on the result of the research, two isolates of bacteria with the highest cellulolytic indeks were B-6 and U-6 isolates that were 7.3 and 3,0. The formation of clear zones around the colony showed that the isolates had qualitatively measured cellulolytic activity. The identification of both isolates was found were the B-6 isolate had the same percentage as Lactobacillus acidophilus (95%) and U-6 isolate had similarity percentage with Enterobacter cloacae (94%).

Abstract

Antifungal Activity of Keben (Barringtonia asiatica (L.) Kurz) Seed Extract against Curvularia verruculosa the Causal Agent of Curvularia Leaf Spot Disease on Rice (Oryza sativa L.)This research was conducted to determine the effectiveness of keben (Barringtonia asiatica (L.) Kurz) seed extract to control Curvularia verruculosa the causal agent of curvularia leaf spot disease on rice. The extract concentration of 0.1%, 0.2%, 0.3%, 0.4%, 0.5%, 0.6%, 0.7%, 0.8%, 0.9% and 1.0% was tested on in vitro test to determine the minimum inhibitory concentration (MIC) and inhibition percentage. Crude seed extract was partitioned and analyzed using GC-MS to determine the active antifungal compound. Concentration of 0.6%, 0.7%, 0.8%, 0.9% and 1.0% is tested on in vivo test in a greenhouse to determine the effectiveness of keben seed extract formula to control the curvularia leaf spot disease on rice. The result shows that the MIC of keben seed extract was 0.5% with inhibitory percentage that increases from 0.1% to 0.5% and cause 100% inhibitory on 0.6% concentration and above. GC-MS analysis showed antifungal compound such as Butyl Hydroxy Toluene; Dodecanoic acid, methyl ester; Methyl tetradecanoate; Benzoic acid 2, 5-bis (trimethylsiloxy)-trimethylsilyl ester; 1-Tetradecanol; Hexadecanoic acid, methyl ester; Octasiloxane, 1, 1, 3, 3, 5, 5, 7, 7, 9, 9, 11, 11, 13, 13, 15, 15-hexadecamethyl-; 13-Docosenoic acid, methyl ester (Z)- and 1, 2-Benzenedicarboxylic acid, dioctyl ester are present in keben seed. Greenhouse test showed that keben seed extract can suppress curvularia leaf spot disease from 25.16% to 2.57%.

Abstract

Citrus spp conservation. in Bali Botanical Gardens and Potential UtilizationCitrus is one of the genus on Rutaceae. Plants who derived from East Asia and Southeast Asia has many benefits because in addition to having fruits rich with vitamin C, also produces essential oils in some parts of the body. Essential oil is widely used in the field of food flavoring industry, comestics and perfumery industries. Essential oil is widely used for treatment as aromatherapy. In plant breeding efforts the existence of various wild species is important to ensure the availability of genes, so that efforts to improve the quality of the plant can still be done. Several factors such as pests and diseases can cause the presence of Citrus spp. threatened sustainability. Therefore, it is necessary to do a conservation effort to protect and preserve it. Bali Botanical Gardens as one of the ex-situ conservation agencies, is in charge of preserving various types of plants from nature. Until the Year 2017 Bali Botanical Garden has collected 12 types of Citrus among others; C. aurantifolia, C. aurantium, C. amblycarpa, C. hystrix, C. grandis, C. maxima, C. medica, C. natsudaidai, C nobilis, C nobilis var. microcarpa C. papaya and Citrus sp. which originated from several regions in Indonesia. Judging from its content, essential oil from Citrus spp. potentially exploited as anti-insect and anti-cancer.

Abstract

Distribution, Population and Damage Percentage of Citripestis sagittiferella Moore (Lepidoptera: Pyralidae) at Bali Citrus cultivation Center.This research was conducted at citrus cultivation center in Bali Province in Bangli and Gianyar regencies and in Integrated Pest and Disease Management Laboratory, from August to November 2017. The purpose of this research is to know the distribution, population and damage percentage of citrus fruit borer, Citripestis sagittiferella. This research used survey method. The study area is divided into three zones: zone I (altidute 800-900 m asl), Zone II (altitude >900-1000 m asl), and Zone III (altitude >1000-1.100 m asl). The results showed that C. sagittiferella was found to spread evenly in all zone locations, both zone I, Zone II, and Zone III, with a total population of 6,909 larvae, 4,861 larvae and 3,508 larvae, respectively. The average larval population found in damage citrus fruit is 5.24 larvae per fruit. The percentage of damage on Zone I was 30,57%, Zone II was 28,90%, and Zone III was 27,96%. The correlation test results showed a very strong relationship between the larvae population and the percentage of damage with r = 0.994.

Abstract

Utilization of Landsat 8 and Google Earth For Identification of Land in Cibiru Bandung CityThe availability of paddy field in urban areas from year to year experienced a decrease in wetland area as a result of conversion agricultural land to non-agricultural. Cibiru Sub Region City (SWK) Ujungberung is one of the priority areas of development. The goal of Bandung City government to have eternal paddy field needs to be supported with distribution of rice fields map for development and minimize land use conversion. The research method was used descriptive survey method, Landsat 8 and Google earth image interpretation results are then validated by conducting field surveys at the location points of the initial interpretation of rice fields and other land cover that is still verification from the results of studio intrepetation. The results obtained by NDVI value of Landsat 8 image for paddy field in Kecamatan Cibiru minimum 0.09296; maximum 0.23502 with an average of 0.16225. The result of identification with high resolution image google earth width of rice field in District of Cibiru identified 58.47 hectares.

Abstract

The Potential of Predator Coccinella transversalis Fabricius (Coleoptera: Coccinellidae) as A Biology Control Agent Thrips parvispinus Karny (Thysanoptera: Thripidae) in Chili Plant (Capsicum annuum L.)Thrips parvispinus Karny (Thysanoptera: Thripidae) is one of the main pests in the cultivation of chili. T. parvispinus existence can damage chili plants and decrease production. This study aims to determine the potential of predator Coccinella transversalis required predation testing which includes a value of searching time (a), handling time (Th) and functional response C. transversalis to T. parvispinus on chili plants. The prey densities presented were 2, 4, 6, 8, 10, 12, 14, 16, 18, 20 and 22 individuals. The exposure is done for twelve hours. Predator C. transversalis used is a female imago that has been fasted for 24 hours. The determination of functional response type was analyzed by regression analysis, while the predation rate is analyzed using Holling formula. The results showed that the prey density affects the predation rate and the functional response type C. Transversalis. The rate of C. transversalis predation is significantly different in each number of prey exposed. Similarly, on the basis of regression analysis, it is shown that C. transversalis has a functional response of type I to T. parvispinus prey. These results indicate that C. transversalis has potential as a biological control agent.

Abstract

Differences of Greening Planting Ages Against Soil Quality Changes in Pelaga Village, Petang District, BadungLand as an environmental component must be maintained, protected and avoided from adverse impacts and must be maintained and enhanced. This study aims to determine the effect of different age of reforestation plants on soil quality changes in Pelaga Village, Petang District, Badung Regency. The method used in this research is observation method with direct observation in field and laboratory analysis. The research was conducted in accordance with several stages, namely the planning and preparation of the initial survey of reforestation land that aims to determine the point of sampling in accordance with the age of reforestation, soil sampling, analysis of physical properties, chemistry, and biology in the laboratory and analysis of research data. The results showed that there was no change in soil quality on land with different plantation age 1 year, 2 years, 3 years, compared with control (land with grass vegetation) and decreased soil quality was observed on land with planting age 4 years and 5 years (after harvest).

Abstract

Affectivity of Trichoderma sp. that Added in Leaf Compost to Control Fusarium Wilt Diseases on Strawberry Plants (Fragaria sp.) in Pancasari Village Buleleng RegencyThis research purpose in order to find the best concentrations of Trichoderma sp. which is added into the leaf compost to control fusarium wilt disease on strawberry plant. This research was conducted on December 2017 to March 2018 in Plant Diseases Laboratory of Udayana University and Pancasari Village, Sukasada District, Buleleng Regency. The experimental design used a Randomized Block Design (RBD) with six treatments and four replications, for each treatment consists of ten plant units. Observation variables in this research are disease percentage, Trichoderma sp. population on the ground, plant height, yield, and the weight of plants. The results showed that Trichoderma sp. treatment which were added into leaf compost is more effective to control the fusarium wilt diseases on strawberry plants rather than control treatments. P5 is the best treatment as it has the lowest disease percentage for around 20%, stimulate the highest growth of the plants into 25.8 cm, and has the highest yields 187 g.

Abstract

The Effects of Rhizobacteria to Promote the Growth and Peanut Resistance (Arachis hypogaea L.) Against Leaf Spot and Rust DiseasesPeanut (Arachis hypogaea L.) is an important legume plants that produces seeds with high protein and vegetable fat content, as well as many processed into various food products and as a source of vegetable oil. The purpose of this research is to know the effect of rhizobacteria to promote the growth and resistance of the peanut against diseases. The research used a Randomized Completely Block Design with 5 treatments and 5 replicates. The variables observed were plant height, number of branches, number of leaves, content of leaf chlorophyll, number of root nodules, disease intensity, disease intensity suppression, number of pods per plant, weight of pods per plant, dry weight of seeds per plant, dry weight of seeds per harvested area, and peanut yields per hectare. The results showed that all treatments of rhizobacteria used was able to promote the growth and resistance of the peanut against diseases so that peanut yields could be improved. Rhizobacteria RZ 35 and RZ 36 are rhizobacteria that have the best ability than other rhizobacterias. RZ 35 has more ability in improving growth and peanut resistance against diseases, whereas RZ 36 has more ability in improving peanut yields.

Abstract

Influence of 2,4-D to induce callus from strawberry leaves on alternative media of AB Mix hydroponic nutrientThis research was conducted to find out the most optimum 2,4-D concentration to induce callus from strawberry leaves on alternative media of AB Mix hydroponic nutrient. This research used Completely Randomized Design with 5 levels of treatment concentration of 2.4-D hormon, first: AB Mix hydroponic nutrients (as basic media) without 2,4-D, second: basic media + 0.5 ppm 2.4 -D, third: basic media + 1 ppm 2,4-D, fourth: basic media + 1.5 ppm 2,4-D, fifth: basic media + 2 ppm 2,4-D. The variables observed were the day explant curled up, day explant swelled, percentage of eksplan curled up (%), percentage of explants swelled (%), the day callus appears, and percentage of explant browning (%). The best percentage of the treatment in callus induction of the strawberry leaves is on the P3 treatment with the concentration of 2,4-D as much as 1.5 ppm, with the fastest curled up explants time of 3 days after culture, the highest curled up eksplants (93.3%), the fastest swelled explants (17.0 days after culture), and the highest swelled percentage (40.0%). Callus formed 32 days after swelling. Successful callus formed only in treatment basic media with 1,5 ppm 2,4-D concentration.
