Abstract

Detection of Ocurrence Citrus Vein Phloem Degeneration (CVPD) Disease on Citrus Plant with Comprehensive Symptoms using Polymerase Chain Reaction (PCR) Technique Citrus Vein Phloem Degeneration (CVPD) is one of very serious disease of citrus plant. The disease is caused by Liberobacter asiaticum bacteria and transmitted by Diaphorina citri Kuw insect. The outspread of disease can also caused by the use of citrus seedlings which have been infected by CVPD. Preliminary visual observations CVPD disease symptoms, there are two types of plants showing symptoms of partial and systemic. Partial symptoms are not all the leaves of plants showing symptoms CVPD, while the systemic symptom is on the whole leaves of plants showing symptoms CVPD. This research aimed to determine the outspread of L. asiaticum bacteria in every parts of the plant that made detection by PCR. The research has been conducted from in Batukaang, Bangli and Plaga, Badung then continued at Laboratory of Genetic Resources and Biology Molecular, Udayana University. Results of PCR visualization using 1% agarose gel showed that there were a DNA band at 1160 bp on the leaves, twigs, branches and trunks. Because the size of 1160 bp DNA bands is expression by L. asiaticum, then the citrus leaf samples were detected positive for the L. asiaticum and it can be ascertained that L. asiaticum haas been distributed to all parts of the plant.

Abstract

The Impact of Chemical Fertilization, Organic, Mineral, and The Combination of Fertilizer towards the Production Number and Quality of Sweet Corn (Zea mays saccharata Sturt.) in Inceptisol Experimental Garden Agriculture Faculty of Udayana University The study aims to figuring out the used of some fertilizers in order to increase the production and the quality of sweet corn in Inceptisols area, and also for figuring out chemicals characteristic and soil fertility in Pegok Village, Denpasar. This research was conducted on August until November 2016, the research took place in the Experimental Garden Faculty of Agriculture, and Soil Laboratory and Environmental Sciences, Faculty of Agriculture, Udayana University. The design method was used for this research was a randomized complete block resign (RCBD) using seven treatments and three-time repetition with the number of plots 21 consist of P0 = Control, P1 = Organic fertilizer, P2 = Mineral fertilizer, P3 = Chemical fertilizer, P4 = Combination Fertilizer (PO + PK), P5 = Combination Fertilizer (PM + PK), P6 = Combination Fertilizer (PO + PM + PK). When the result of variance statistical test shows different result, the procedure followed by LSD method (Least Significance Different) with 95% of confidence level using Costat program. The result of statistical analysis shows the role of chemical fertilizers was proven ability to increase the sweet corn production number, the highest result was 13,97 tons ha-1 which use the P3 treatment (200 kg Phonska and 200 kg Urea) ha-1 increased 110,08%, it showed a better result compared to P0 (Control) which only showing 6,65 tons ha-1. The best quality of sweet corn was shown by P6 formula: the combination of organic, chemical and mineral fertilizer.

Abstract

ABSTRACT Biological Fertilizer Usage Effectiveness (Enterobacter cloacae) to Increase Yield and Quality of Rice Seeds Varieties Cigeulis The purposes of this research were to find out the influence of biological fertilizer Egary which contains rhizobacteria Enterobacter cloacae related to the crop of Cigeulis rice, as well as to test the effectiveness of biological fertilizer to reduce the utilization of chemical fertilizer, and to obtain the quality of the seed production. The resullt indicated that the augmentation of biological fertilizer plus chemical fertilizer phonska and urea at 100 kg/ha each (Py) increased the production of the rice crop about 1.8 tons/ha or increased about 33.93% and the result of biological fertilizer plus chemical fertilizer phonska and urea at 200 kg/ha each (Pz) increased about 0.8 tons/ha or about 33.13% if compared with control. The research of Py treatment with biological fertilizer plus chemical fertilizer phonska and urea at 100 kg/ha each, produced Cigeulis rice crop variety for 6.3 tons/ha and improved the quality of the seeds both physical and physiological parameters, produced 1000 grain weight of seeds, germination and vigor highest power savings and has better endurance than Pz and control (30.28 g, 89.11%, and 85.89%, respectively). Abstract

ABSTRACT Efectivity Test for Some Extraction Techniques and Dry Heat Treatment on Tomato Seed (Lycopersicum esculentum Mill.) This experiment aimed to obtain the most effective extraction method and determine the effect of dry heat treatment to produce high quality tomato seeds. The design used was completely randomized design with 3 treatments and 9 replications. The treatments include: (1) Bl : fresh tomato seeds taken, immediately dried to 5% moisture content, (2) Ba: fresh tomato seeds taken, then soaked in water for 24 hours, dried to 5% moisture content, (3 ) Bh: fresh tomato seeds taken, then soaked in a solution of 2% HCl for 2 hours, then dried until the moisture content of 5%. Seeds obtained from each extraction technique is divided into two lots, one lot for Dry Heat Treatment (DHT) and the other one lot for Non Dry Heat Treatment (NDHT). The variables measured were the number of seed moisture content of 5% by weight of unity, purity of the seeds, the total viability, potential viability, vigor speed germination, and seedling growth vigor. The results showed the significant diference between extraction treatment to all of variables tested, while the granting of DHT on the seed does not provide a response to tomato seed quality, on all variables tested. Bh extraction treatment showed the best results with the highest values  on all of variables tested.   Key word : tomato seeds, extraction, dry heat treatment, viability and vigor.

Abstract

Soil Quality Evaluation of Paddy Soil Under “Simantri’s” and Non “Simantri’s” System in Subak Riang, Riang Gede Village, Penebel Subdistrict This experiment conducted to evaluate the differences between soil quality of paddy soil sunder “Simantri’s” and “non Simantri’s” activities, as well as to evaluate the differences among major physical, chemical and biological soil properties which determining their quality. There were three steps applied in this study including: field survey to determine the research area and sampling points; Minimum Data Set to determine the soil quality and t-test to determine the dominant factor which influencing the soil quality. The results of this study showed: the soil quality of faddy soil under “Simantri’s”activity is better than“non Simantri’s” conditions. Other Than That, it was found the soil total nitrogen significantly higher under “Simantri’s” than “non Simantri’s” conditions.

Abstract

Exploration, Inventorize, and Characterization of Durian Kunyit (Durio zibethinus Murr) Germplasm in Sub-District of Pupuan, Tabanan District This research aimed to explore, inventorize, and characterize durian kunyit (Durio zibethinus Murr) plants in the sub-district of Pupuan. The research coveredthe exploration of durian kunyit in each village, the inventorize of the dispersal of durian kunyit, and the characterization of durian kunyit. The variables observed were the agronomic characters, morphology characters, and molecular character of durian kunyit. The results showed the discovery of 8 durian kunyit mother trees scattered at an altitude of 370 – 661 meters above sea level. The agronomic character of durian kunyit fruit are small–medium sized, fruit weight 1,1 kg, fruit length 17,3 cm, fruit diameter 15,0 cm, number of seeds 17 per fruit, aryl texture quite soft, watery condition, taste sweet little bitter, the flesh is quite thick, aril is yellow, the aroma is quite strong, not sticky in the hand and the production reaches 200 pieces per tree at the harvest. The morphology characters of durian kunyit were plants aged 10 – 80 years, plant height 16 m, tree circumference 165 cm, petiole length 17,8 mm, petiolewidth 2,9 mm, length leaf strands 14,2 cm, leaf blade width 4,9 cm. The molecular character of durian kunyit forms 8 specific banding patterns at 350 – 1600 basepair using the PKBT ISSR Primer 2.

Abstract

ABSTRACT Antagonistic Test of Some Rhizobacteria Against Sclerotium rolfsii Sacc. Causes Damping Off Disease in Peanut Plant Damping-off disease is one of the important diseases on peanut plants caused by the fungi Sclerotium rolfsii Sacc. Control which is currently only with synthetic pesticides, which many negative impacts on ecosystems and humans. The many negative effects of pesticides would require alternative more environmentally friendly control. One alternative that can be recommended is the use of rhizobacteria which acts as a biological agent. The results showed that there were some rhizobacteria that effectively inhibit the growth of S. rolfsii Sacc. the inhibition test in vitro with the highest inhibition in the treatment of K. pneumoniae isolates KCX1GRA by 94.9% when compared with controls at 3 days after inoculation observation. Rhizobacteria filtrate tested in vitro showed that the treatment of the filtrate concentration of 10% - 20% on respectively filtrate rhizobacteria able to inhibit the growth of fungus S. rolfsii Sacc. Treatment of the filtrate rhizobacteria isolates 20M2 and KCX1GRA concentration of 20% -50% able to kill the fungi S. rolfsii Sacc., while the filtrate rhizobacteria isolates KCBS and Pi1 concentration of 30% -50% are also able to kill the fungi S. rolfsii Sacc. In the glass house study, the treatment was able to suppress rhizobacteria disease incidance in peanuts from 92% to 8% -10% compared with the control treatment was 92% at 6 weeks after planting observation. Abstract

ABSTRACT Efficacy of Essential Oil of Lemongrass (Cymbopogon citratus L.) to Diamond Back Moth (Plutella xylostella L.) on Cabbage in the Laboratory The use of synthetic pesticides continuously will be affect the biotic and non-biotic factors. Therefore, the others control measure which were practically, effectively and safely to human being and environmental - frendly must be created. Geographic condition of Indonesian has been known is rich in biodiversity is necessary to be studied to find the sources for pest control measure which is environmental-friendly. Study about “ Efficacy of Essential oil of Lemongrass (Cymbopogon citratus L.) to Diamond Back Moth (Plutella xylostella L.) on Cabbage was conducted in February, 4. to October, 18.2012 in the Laboratory of Pest and Diseases, Departement of Agroekotechnology, Faculty of Agriculture, Udayana University. The purpose of this study is to determine killing capacity of essential oil of Lemongrass (C. citratus) against Diamond Back Moth (P. xylostella) on Cabbage. The results showed that the essential oil of Lemongrass is effective to kill the larvae of DBM (P. xylostella). The level of concentrations that is used as treatment in this study are 1.5%, 1.25%, 1.%, 0.75%, and 0.5% that have a significant effect to kill the larvae of P. xylostella compare to the control treatment (tween 80 and water). Concretation 0.5% of lemongrass oil causing mortality in average 38.67%, while the concentrations 1.5% is 100%. The concretations 1.25%, 1%, and 0.75% their percentage of mortality are 94.67%, 74.67%, and 56%, respectively. Base on the results of this study, the optimum concretation which are give mortality to the larvae is 1,5% with the percentage of mortality reach to 100% on 6 days after treatment.
