Abstract

MDA-MB-231 is a breast cancer cell line derived from metastatic adenocarcinoma and classified as a triple-negative breast cancer (TNBC) subtype. This breast cancer subtype is highly aggressive, has a high recurrence rate, and offers limited treatment options due to the absence of estrogen, progesterone, and HER-2 receptor expression. Therefore, the development of advanced therapeutic strategies is urgently needed to inhibit the metastasis of TNBC cancer cells. This study aimed to investigate the potential of the methanol extract of Vernonia amygdalina leaves as a natural chemotherapeutic agent for metastatic breast cancer therapy, with a focus on inducing cell death through apoptosis mechanisms. This study was an experimental research that began with the collection and identification of raw materials, followed by the preparation of simplicia, extraction processes, and phytochemical screening. Subsequently, cytotoxicity testing was conducted using the MTT assay, cell cycle analysis was performed using the PI-staining assay, and apoptosis was assessed using the Annexin V/PI-staining assay, all of which were analyzed through flow cytometry. The methanol extract of V. amygdalina demonstrated a moderate cytotoxic effect with an IC50 value of 109.36 µg/mL. The extract induced S-phase cell cycle arrest in a dose-dependent manner, indicating its ability to reduce cell viability by inhibiting DNA replication. In the apoptosis assay, the highest percentage of cell death (3.12%) was observed at the concentration of 54.5 µg/mL, suggesting that this dose produced the strongest apoptotic response among the treatments tested. The methanol extract of V. amygdalina leaves shows potential as a natural chemotherapeutic agent for metastatic breast cancer. The extract can induce cancer cell death through apoptosis, indicating its promise for further development as a supportive anticancer therapy.

Abstract

The development of topical preparations from natural ingredients requires rigorous physical quality evaluation to ensure their safety, stability, and efficacy. This study aimed to formulate and evaluate the physical quality of a gel preparation containing the water fraction of Cyperus rotundus L. at various concentrations. The water fraction of the rhizome was formulated into gel preparations with concentrations of 5% (F1), 10% (F2), 15% (F3), and 20% (F4) using Carbopol 940 as a gelling agent. The physical quality evaluation included organoleptic tests, homogeneity, pH, spreadability, adhesion, and viscosity. The results showed that all gel formulas met the established physical quality requirements. An increase in the concentration of the water fraction significantly affected the physical characteristics of the preparation, such as a more intense color, as well as an increase in pH, adhesion, and viscosity. All formulas exhibited a pH suitable for the physiological range of the skin (5.53–6.80), good spreadability (5.49–6.77 cm), and adequate adhesion (>2 seconds). It was concluded that the nut grass rhizome water fraction gel has stable and acceptable physical characteristics, making it a potential candidate for further development as a topical preparation.

Abstract

Peningkatan kasus asam lambung pada remaja dipengaruhi oleh gaya hidup tidak sehat dan rendahnya pengetahuan mengenai pencegahan serta penanganannya, sehingga kegiatan ini bertujuan meningkatkan pemahaman remaja tentang penggunaan bahan alam dalam mengatasi asam lambung. Metode yang digunakan adalah edukasi melalui penyuluhan kesehatan kepada siswa SMA dengan media presentasi, brosur, serta evaluasi menggunakan pretest dan post-test. Hasil menunjukkan adanya peningkatan signifikan pengetahuan peserta dari rata-rata 65,15% menjadi 91,51% setelah diberikan edukasi. Remaja mampu memahami penyebab, gejala, pencegahan, serta pemanfaatan bahan alami seperti jahe, kunyit, sereh, madu, dan pepaya sebagai alternatif penanganan awal. Hasil analisis statistik menggunakan uji Paired T-Test menunjukkan adanya perbedaan yang signifikan antara nilai pretest dan post-test (p<0,001), sehingga edukasi kesehatan berbasis bahan alam dapat meningkatkan pengetahuan remaja mengenai pengendalian asam lambung.

Abstract

Background: Streptococcus mutans is the primary bacterium responsible for dental caries. Continuous use of synthetic antibacterial agents may cause side effects; therefore, natural antibacterial alternatives are needed. Kopasanda leaves (Chromolaena odorata L.) are known to contain bioactive compounds with potential antibacterial activity. This study aimed to evaluate the antibacterial activity of the ethyl acetate fraction of the ethanolic extract of kopasanda leaves (Chromolaena odorata L.) against Streptococcus mutans.Methods: An experimental laboratory study was conducted using the agar well diffusion method with ethyl acetate fractions at concentrations of 10%, 15%, and 20%. Gentamicin was used as the positive control and the solvent as the negative control. The inhibition zones were measured in millimeters and analyzed using One-Way ANOVA.Results: The ethyl acetate fraction showed antibacterial activity against Streptococcus mutans, with mean inhibition zone diameters of 11.33 mm, 13.49 mm, and 15.98 mm at concentrations of 10%, 15%, and 20%, respectively. The 20% concentration exhibited the highest antibacterial activity, and statistical analysis indicated a significant difference among treatments (p < 0.05).Conclusion: The ethyl acetate fraction of kopasanda leaf extract (Chromolaena odorata L.) exhibits antibacterial activity against Streptococcus mutans, with 20% being the most effective concentration.
