Abstract

Ndusuk leaves (Melastoma malabathricum L.) are traditionally used by the Manggarai community to reduce the fatty appearance of pork dishes, yet the molecular basis of this practice has not been investigated using a GC-MS-based metabolomic approach. This study compared the metabolite profiles of Ndusuk leaf decoctions prepared without and with pork. Dried leaves (35 g) were decocted in 2.5 L of water at 100°C for 30 min under two treatments: leaves only (control) and leaves with 100 g pork (sample), each performed in triplicate (n = 3). Metabolites were analyzed by GC-MS and tentatively identified using the NIST library (similarity index ≥80%). The control contained 118 tentatively identified metabolites, whereas the pork-treated extract contained 126. Boiling with pork increased cis-vaccenic acid (+4.07%) and palmitic acid (+1.92%), while decreasing melezitose (−2.80%) and glycerol (−0.73%). These findings demonstrate that cooking with pork alters the metabolite profile of Ndusuk leaf decoctions without providing evidence of direct fat reduction. The observed metabolite shifts may suggest possible phenolic–lipid interactions, antioxidant-related processes, and Maillard reaction-associated changes; however, these mechanisms require confirmation using targeted metabolomics and authentic standards. This study provides the first metabolomic evidence supporting the traditional culinary use of Ndusuk leaves and establishes a scientific basis for Manggarai pork-cooking practices.

Abstract

Background: Red ginger (Zingiber officinale var. Rubrum) is rich in bioactive compounds, such as gingerol and shogaol, which are highly sensitive to type of extraction solvent. Therefore, a chromatogram profile analysis is required to determine most effective solvent. Purpose: This study aims to compare chromatogram profiles of 95% ethanol and ethyl acetate extracts of red ginger rhizome using High-Performance Liquid Chromatography (HPLC). Method: This descriptive laboratory study extracted 90 gr of red ginger crude drug (simplisia) powder from Ende Regency, Flores, via maceration using 95% ethanol and ethyl acetate for 3 × 24 hours. Resulting filtrates were evaporated to obtain thick extracts, which were then analyzed using HPLC to evaluate the number of peaks, retention time, and peak area. Results: 95% ethanol extract produced a more complex chromatogram profile with 30 peaks, whereas the ethyl acetate extract yielded 22 peaks. This difference indicates superiority of 95% ethanol in extracting secondary metabolite compounds across a wider polarity range compared to ethyl acetate. Conclusion: There is a significant difference in chromatogram profiles between two extracts. 95% ethanol solvent proved to be more effective in extracting bioactive compounds from red ginger rhizome than ethyl acetate, serving as a baseline for herbal raw material standardization and a foundation for future research.   Keywords: HPLC; Red ginger; Zingiber officinale var. Abstract

Background: Faloak (Sterculia quadrifida R.Br.) is a traditional medicinal plant used by people of East Nusa Tenggara that contains flavonoid compounds, one of which is rutin, which has potential as an antioxidant and anti-inflammatory. However, information regarding the distribution of rutin across various organs of Faloak plant and its antioxidant activity remains limited. Purpose: To determine rutin content in ethanol extract of Faloak (Sterculia quadrifida R.Br.) leaves using HPLC and to determine antioxidant activity of extract using DPPH (1,1-diphenyl-2-picrylhydrazyl) method. Method: This research is a descriptive study conducted in a laboratory by applying treatments to samples, including extraction using ethanol solvent and analysis of target compound using an HPLC instrument. Data generated consist of numerical values from measurement of rutin content and percentage inhibition of DPPH radicals in ethanol extract of Faloak leaves. Results: Calibration curve of rutin standard showed good linearity ( = 0.993). Ethanol extract of Faloak leaves contains rutin with an average content of 0.807 ± 0.094%. DPPH test results for Faloak leaf extract showed an  value of 102.17 ppm, which is classified as a moderate antioxidant category. Conclusion: Rutin was detected in quantifiable amounts in Faloak leaves. Faloak leaf extract also exhibited moderate antioxidant activity, consistent with detected rutin content, thereby demonstrating potential for development as a natural source of rutin with antioxidant activity.   Keywords: Antioxidant; DPPH; Faloak; HPLC; Rutin.

Abstract

Abstract

Background: Plant-based ethnomedicine practices remain a vital part of rural public health systems. Knowledge regarding the use of medicinal plants is passed down through generations, yet it has not been scientifically documented, placing it at risk of being lost as times change. Purpose: To identify the types of medicinal plants, the plant parts used, preparation methods, modes of administration, and the types of ailments treated by the village community. Method: This is a descriptive qualitative study employing an ethnographic approach. It was conducted in Camplong 2 Village, Fatuleu District, Kupang Regency, East Nusa Tenggara Province. Data were collected through observation, in-depth interviews, and documentation involving three key informants and 27 additional informants. Data analysis was performed through data reduction, data presentation, and conclusion drawing. Results: Forty seven species of medicinal plants utilized by the community were identified. The plant part most frequently used was the leaf (59%), the dominant preparation method was boiling (57%), and the most common method of administration was oral consumption (54%). These medicinal plants were used to treat 34 types of ailments categorized into 10 groups, with the category comprising fever and general bodily complaints being the most prevalent (19%). Conclusion: Village communities continue to maintain plant-based ethnomedicine practices rich in local knowledge. The diversity of medicinal plant species, along with the methods of their preparation and use, demonstrates that traditional medicine still plays a vital role in maintaining community health.   Keywords: Ethnomedicine; Medicinal Plants; Traditional Medicine.
