Abstract

In order to prevent oxidative stress, our body synthesizes catalase, an endogenous antioxidant enzyme that catalyzes hydrogen peroxide (H2O2) into water and oxygen. Until now, the level of MDA and catalase in aging population were still varied and those level at different age in elderly population has not been yet reported. Therefore, the purpose of this study was to analyse the profile of MDA level and catalase specific activity in plasma of elderly women based on increasing age. Methods: This research used 60 healthy elderly women as the subjects living in Jakarta. The subjects were divided into 2 groups based on age category, the younger group (60 – 70 years old) and the older group (>70 years old). MDA and specific activity of catalase were analyzed in plasma using spectrophotometer. Results: MDA level in the younger group (60-70 years old) was slightly higher than MDA levels in the older group (>70 years old) but it was not significant. Moreover, specific activity of catalase in the younger group was significantly lower than the older group. Conclusions: There was no difference in MDA level of elderly woman between younger and older group. However, catalase specific activity significantly increased with increasing age. Abstract

Several studies have predicted the molecular interactions of the active ingredient of Spirulina platensis as an anti-inflammatory compound. However, these interaction studies did not review the modulation of the NF-κB activation pathway, which involves various factors. This study demonstrated the potential of the bioactive compounds of S. platensis for modulating immune function by reducing inflammation through the inhibition of the NF-κB activation pathway. Phycocyanobilin was predicted to have good potential for molecular docking with multisubunit IκB kinase (IKK)1/IKKA, IKK2/IKKB, NF-κB-inducing kinase, and the IκBα/NF-κB complex. Furthermore, β-carotene exhibited good potential for interactions with NF-κB essential modulator/IKK and the NF-κB complex, and α-glucan had the potential for interactions with COX-2. Therefore, supplementation with S. platensisand its bioactive compounds is expected to provide optimal benefits. Abstract

Relative Expression of HIF-1α mRNA in Rat Heart, Brain and Blood During Induced Systemic Hypoxia. Hypoxia is a pathological condition in which the body as a whole or region of the body (tissue or cell) deprived of adequate oxygen supply. The transcriptional regulator hypoxia inducible factor-1 (HIF-1) is an essential mediator of O2 homeostasis. Unlike the β sub unit (HIF-1β), the activity of HIF-1α is controlled in an oxygen-dependent manner. It has been reported that the stability and expression of HIF-1α during hypoxia is remarkably higher than those under normoxic conditions. The aim of this study was to analyze the adaptive tissue responses during induced systemic hypoxia by comparation of relative expression of mRNA HIF-1α in rat heart, brain and blood. Twenty-five male Sprague Dawley rats were subjected to systemic hypoxia by placing them in the hypoxic chamber supplied by 8-10% of O2 for 0, 1, 7, 14 and 21 days, respectively. The relative expression level of HIF-1α mRNA in brain, heart and leucocyte cells were analyzed using quantitative RT-PCR assay (Real Time PCR) based on Pfaff’s formula. This study demonstrates that the increased of relative expression of HIF-1α mRNA during induced systemic hypoxia reached its maximum level at day 7 (in heart) or at day 14 (in brain), whereas in leucocyte cells the stimulation of HIF-1α expression was intensively maintained up to 21 days although the expression has reached the remarkably high level. We could conclude that HIF-1α as an oxygen sensing during systemic hypoxia has different capacity and sensitivity in brain, heart and blood tissues, due to the importance of oxygen homeostasis in each tissue

Abstract

Pro- and anti-inflammatory mediators need to be released in a balanced way to maintain a healthy state as we age. One important regulatory element in the equilibrium of pro- and anti-inflammatory mediators is NF-κB. The purpose of this study was to examine how S. platensis affected the control of inflammatory mediators in young, healthy, emerging adults and adults in rats. In this investigation, 200 mg/kg BW of S. platensis extract was administered to six groups of male Wistar rats, ages 12, 18, and 24 weeks, along with a control group. In both the treatment and control groups, NF-κB p65 protein expression was lower at 24 weeks than it was at 12 and 18 weeks. TNF-α and COX-2 proteins were lower in the treatment group than in the control group. All age groups in the treatment group had higher levels of IL-10 protein than the control group. The quantity of NF-κB p65 was positively correlated with COX-2 and TNF-α. By raising the concentration of NF-κB p65, the ethanolic extract of S. platensis altered a mediator of cellular immunity. A decrease followed this in TNF-α and COX-2 and a rise in IL-10 in the rat spleen at different ages.

Abstract

Flavonoids are naturally occurring bioactive compounds widely found in fruits, vegetables, tea, and various medicinal plants. Numerous studies have demonstrated that flavonoids exhibit biological activities involved in the regulation of cell growth, metabolism, and aging processes through their effects on the growth hormone (GH) and insulin-like growth factor-1 (IGF-1) signaling axis. The GH/IGF-1 pathway plays a crucial role in growth regulation, cellular differentiation, energy metabolism, and the development of various diseases, including cancer and metabolic disorders. Therefore, understanding the interaction between flavonoids and this signaling pathway is essential for the development of natural product-based therapeutic strategies. This study employed a Systematic Literature Review (SLR) approach to identify, retrieve, and synthesize scientific evidence regarding the role of flavonoids in modulating the GH and IGF-1 signaling pathways. Literature searches were conducted across several international scientific databases following the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) guidelines. Eligible articles were then qualitatively analyzed to evaluate the molecular mechanisms of flavonoids in the regulation of components within the GH/IGF-1 signaling pathway. The results indicate that various flavonoids, including quercetin, kaempferol, genistein, epigallocatechin gallate (EGCG), and luteolin, can modulate the GH/IGF-1 axis by regulating IGF-1 receptor expression and downstream signaling pathways such as PI3K/AKT/mTOR and MAPK. These mechanisms contribute to the regulation of cell proliferation, induction of apoptosis, reduction of oxidative stress, and improvement of metabolic homeostasis. Overall, these findings suggest that flavonoids hold potential as therapeutic and preventive agents in conditions associated with dysregulation of the GH/IGF-1 signaling pathway.

Abstract

Background: Free radicals in excessive concentrations damages cells and accelerate the aging process. Antioxidants found in Centella asiatica (CA) and Acalypha indica (AI) have the potential to prevent oxidative cellular damage. Objective: This study aimed to determine the effect of CA and AI on carbonyl and glutathione levels in the brain of older rats. Methods: 18-month age rats were treated using either AI, CA, or vitamin E. In addition, 18-month age and 2-month age untreated rats were used as a negative control. The brain carbonyl and glutathione levels were measured by Agustyanak and the Elmann method, respectively. Results: Treatment with CA significantly decreased brain carbonyl levels (2.87 nmol/mL) than the control rats (4.54 nmol/mL). Furthermore, treating AI did not reduce the brain carbonyl and GSH levels in aged brain rats. Conclusion: Centella asiatica can reduce the protein destruction that occurs with increasing age.

Abstract

Background: Hypertensive disorders, particularly preeclampsia, are major contributors to maternal mortality and neonatal morbidity. Angiogenic imbalance plays a critical role in placental ischemia, a hallmark of preeclampsia. Purpose: This study aimed to investigate the roles of endoglin, soluble endoglin (sEndoglin), and matrix metalloproteinase-14 (MMP-14) in the angiogenic imbalance observed in preeclampsia placentas compared to normal-term placentas. Method: A cross-sectional study was conducted using 68 placental samples: 34 from normal-term pregnancies and 34 from preeclampsia cases. Concentrations of endoglin, sEndoglin, and MMP-14 were measured using the sandwich ELISA method, and protein levels were determined using the Christian Warburg method. Data were analyzed using SPSS version 20. Results: The concentration of endoglin in preeclampsia placentas was slightly higher (1.37 [0.2–2.2] ng/μg protein) compared to normal placentas (1.12 [0.6–14.1] ng/μg protein), although the difference was not statistically significant. In contrast, sEndoglin (0.05 [0.0–0.01] ng/μg protein vs. 0.02 [0.0–0.3] ng/μg protein) and MMP-14 (0.14 [0.1–0.6] ng/μg protein vs. 0.11 [0.1–1.3] ng/μg protein) concentrations were significantly higher in preeclampsia placentas compared to normal placentas. All parameters showed a gradual decrease with advancing gestational age. sEndoglin and MMP-14 demonstrated a strong positive correlation (r = 0.658, p < 0.001), while endoglin and MMP-14 exhibited a moderate positive correlation (r = 0.554, p < 0.001). Conclusion: Endoglin, sEndoglin, and MMP-14 were differentially expressed in preeclampsia placentas, with sEndoglin and MMP-14 significantly elevated. These findings highlight their potential role in angiogenic imbalance and may provide insight into the pathophysiology of preeclampsia.

Abstract

Background: Oxidative stress tends to rise as age increases, with the liver being notably affected. Changes in liver function are closely linked to oxidative stress, which targets hepatocytic proteins. Free radicals that attack the liver can cause damage to its proteins, which can be measured through carbonyl level. Spirulina platensis, is a blue-green sea algae which grow in sea waters throughout the world and have been consumed as antioxidants. Objective: The objective of this study was to evaluate the effect of Spirulina platensis administration on carbonyl level in the liver tissues of young adult rats with different ages. Methods: Male Wistar rats aged 8 weeks, 14 weeks, and 20 weeks were given 200 mg/kg BW S. platensis extract orally once a day for 29 days until aged 12 weeks, 18 weeks, and 24 weeks. Control group of each age (only given aquadest) Results: The 12-week group given spirulina showed an increase in carbonyl level of 1,215 times compared to the control group. The 18-week group given spirulina showed a reduction in carbonyl level of 0.686 times compared to the control group. The 24-week group given spirulina showed an increase in carbonyl level of 0.924 times compared to the control group. Conclusion: Spirulina platensis administration decreased the carbonyl level in 18-weeks and 24-weeks old rat compared to control group.

Abstract

Traumatic brain injury (TBI) is a major global health problem that often leads to systemic complications beyond neurological damage, notably gastrointestinal (GI) dysfunction. The mechanisms linking TBI to GI complications remain inconclusive. We conducted a systematic review using current clinical evidence on the pathophysiological processes underlying post-TBI GI dysfunction, focusing on two principal mechanisms: inflammatory-oxidative processes and hypothalamic–pituitary–adrenal (HPA) axis alterations. A comprehensive search was conducted through PubMed, Cochrane Library, and Scopus to identify eligible studies. Evidence indicates that surges of proinflammatory mediators and chemokines, along with reduced anti-inflammatory mediators, drive systemic immune imbalance. Moreover, iNOS upregulation and gut microbiota dysbiosis contribute to mucosal injury. Concurrently, HPA axis dysregulation exerts a bidirectional impact. Elevated ACTH and cortisol reflect an intact stress response that may stabilise metabolism if combined with early enteral nutrition, whereas critical illness-related corticosteroid insufficiency (CIRCI) and hypergastrinemia are strongly associated with gastrointestinal bleeding and mortality. Together, these findings underscore the synergistic role of inflammatory and endocrine disturbances in driving gastrointestinal vulnerability after TBI. Understanding these mechanisms is crucial for developing biomarker-based monitoring and targeted interventions to improve prognosis and reduce GI-related complications in TBI patients.
