Abstract

Abstract

Antidiabetic activity of Cocor Bebek leaves (Kalanchoe pinnata Lam.Pers.) ethanolic extract from Bogor city, Kabupaten Bogor and south of Tangerang city has been studied. The study was conducted in vitro using glucosidase inhibitor method. The results of the study showed that IC50 of the extract from Bogor city, Kabupaten Bogor, and Tangerang Selatan city is 40.94 ppm, 33.58 ppm and 16.12 ppm respectively. Meanwhile, IC50 of quersetin which has antidiabetic activity is 10.22 ppm. The results showed that Cocor Bebek leaves (Kalanchoe pinnata Lam.Pers.) ethanolic extract had antidiabetic activity with IC50 less than 100 ppm. However, the activity is lesser than quercetin. Keywords:Kalanchoe pinnata Lam.Pers, antidiabetic activity, glucosidase inhibitor

Abstract

Nyatoh plant (Palaquium amboinense B.) is native to Indonesia which is known to have the ability to produce potential secondary metabolites. The aim of this study was to evaluate the antibacterial and antioxidant activities from bioactive compounds produced by endophytic bacteria from nyatoh leaves. Five endophytic bacteria were tested against 4 targeted bacteria including Escherichia coli ATCC 8739, Pseudomonas aeruginosa ATCC 15442, Bacillus subtilis ATCC 19659, and Staphylococcus aureus ATCC 6538. Bacterial isolate which had the strongest antibacterial activity was then fermented and extracted. The corresponding extract was further tested on disc diffusion method, determination of Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) values, antioxidant evaluation, and analyzed for its bioactive compound components via GC-MS analysis. The results showed that one isolate, namely D1 can produce secondary metabolite which act as antibacterial and antioxidant potentials. This corresponding extract has the strongest MIC and MBC against B. subtilis with value of 39.5 and >78.1 µg/ml and exhibited antioxidant activity against 2,2-difenil-1-pikrilhidrazil (DPPH) with inhibition value of 85.37%. Based on GC-MS analysis, secondary metabolites derived from this potential extract contained some major active constituents including Tributyl acetylcitrate and bis (2-ethylhexyl) ester which might act as antibacterial and antioxidant agents.

Abstract

Chlorella sp. microalgae is a potential source of natural antioxidants and other bioactive compounds used in the food and pharmaceutical industries. Ultrasound pretreatments followed by enzymolysis are widely used to improve the functional properties of biological materials. This work aimed to investigate the effects of ultrasound pretreatments followed by bromelain hydrolysis on the proteins, phenolic content, and antioxidant activity of Chlorella sp. crude extract. The experimental design for the study involved two factors: ultrasound pretreatment time (0, 5, and 10 min) and bromelain hydrolysis time (0, 2, 4, and 6 hours). The protein was measured using Lowry assay while the total phenolic content was measured using a Folin–Ciocalteu assay. The highest protein content (7.09 ± 0.44 mg/mL) and phenolic concentrations (0.28 ± 0.01 ìg/mL) were obtained at 5 min ultrasound pretreatment and 6 hours enzymolysis. For antioxidant activity, 5 min ultrasound pretreatment followed by 4 hours of enzymolysis resulted in the highest DPPH (1,1-diphenyl-2-picrylhydrazyl radical) inhibitory activity (81.74%). Meanwhile, ultrasound pretreatment for 10 min followed by 6 hours of enzymolysis resulted in the highest ABTS (2,20 azinobis-(3-ethylbenzthiazoline-6-sulfonic acid) inhibitory activity (94.10%). This study showed that variation in ultrasound pretreatments followed by bromelain hydrolysis could be used to improve the protein content, phenolic content, and antioxidant activity of Chlorella sp. crude extract.
