Abstract

Potato is an important food crop in both developed and developing countries and it is the fourth most important food crop after wheat, maize, and rice. Potato production in Indonesia has decreased in recent years, mainly due to the use of low quality tuber seed. The objective of this research was to determine the effect of plant materials, types of nutrients, and planting density on mini tuber formation, as well as to determine the best aeroponics conditions for mini tuber formation of Granola cultivar of potato. This research has been carried out experimentally using a Split-split Plot Design. The main plot was planting materials which consisted of plantlet and knol. The subplot was the type of nutrition media which consisted of Farran nutrition, Otazu nutrition, and AB-commercial mix. The sub-sub-plot was planting density which consisted of 50, 75, and 100 plants/m2. The research results showed that knol planted in aeroponics system on AB-commercial mix at a planting density of 75 plants/m2 resulted in the best mini-tuber production. In this study, for the first time three important factors for potato mini-tuber production have thoroughly been investigated in Indonesia. This finding will be used by Kledung Horticultural Seed Garden, Temanggung, for mass production of high-quality mini-tuber seed, which in turn will provide sufficient tuber seed for the farmer in its surrounding areas. By increasing the production of high-quality tuber seed, it is expected that it will contribute to the improvement of potato productivity and increase Indonesian potato production.

Abstract

This research has been carried out with a view to induce rooting and  plantlet formation, followed by acclimatization.  Micro shoots of sugarcane cv. PS 881 were cultured on Murashige and Skoog medium supplemented with different types and concentration of auxins for root induction. This research conducted experimentally using a split-plot design. The main plots were three types of auxins, which consisted of IAA, IBA, and NAA. The subplots were auxin concentrations with four levels, i.e. 0 μM, 5 μM, 10 μM, and 15 μM. Significantly faster root emergence time and higher number of roots observed in the Murashige and Skoog basal medium supplemented with 10 μM NAA. The best root length obtained in the Murashige and Skoog basal medium supplemented with NAA 0 μM. Plantlets derived from NAA 10 μM treatment showed the best performance during acclimation with a 100% survival rate. NAA at a concentration of 10 µM considered to be the best treatment in  plantlet formation and acclimatization of sugarcane cv. PS 881. This study showed that the use of MS medium with 10 uM NAA is able to increase the growth of PS 881 sugarcane plantlets. The results of this study can increase the availability of high quality seedlings and increase national sugar production.

Abstract

Tagetes sp. merupakan salah satu tanaman hias yang memiliki nilai yang cukup tinggi. Tagetes sp. sering dimanfaatkan secara beragam. Penelitian tahun pertama ini dilakukan dengan tujuan untuk: mempelajari pengaruh interaksi paclobrutazole dan gibberellin terhadap pembungaan Tagetes sp.dan untuk menentukan konsentrasi paclobrutazole dan gibberellin terbaik untuk memacu pembungaan Tagetes sp. Metode penelitian yang digunakan adalah metode eksperimental dengan pola perlakuan RAL faktorial. Sebagai faktor pertama adalah konsentrasi gibberellin terdiri dari 4 taraf yaitu 0 ppm; 50 ppm; 100 ppm; 150 ppm. Sebagai faktor kedua adalah konsentrasi paclobrutazole, yang terdiri dari 3 taraf terdiri dari  0 ppm; 100 ppm; 200 ppm. Setiap kombinasi perlakuan diulang 10 kali sehingga diperoleh 120 unit percobaan. Hasil yang didapat menunjukan bahwa interaksi antara paklobrutazol dan GA3 tidak menunjukan hasil yang berbeda nyata pda semua parameter yang diujikan. Penggunaan GA3 dan paklobrutazol secara mandiri dapat mempengaruhi pertumbuhan Tagetes sp. Perlakuan GA3 100 ppm menunjukan tinggi tanaman dan laju pertumbuhan relatif sebesar 24,40 dan 3,08. Perlakuan paklobrutazol pada konsentrasi 0 ppm dan 200 ppm menunjukan rataan jumlah bunga terbanyak yaitu 10,80, sedangakan untuk rataan jumlah akar terbanyak ditunjukan pada perlakuan paklobrutazol 0 ppm yaitu sebanyak 49,06.

Abstract

AbstractBambu merupakan tumbuhan yang termasuk dalam subfamilia Bambusoidae dari familia Poaceae. Keanekaragaman Bambu di dunia tergolong tinggi, dengan pemanfaatannya yang beragam. Penelitian ini bertujuan untuk mengetahui keanekaragaman bambu berdasarkan karakter morfologi dan pemanfaatannya di wilayah Kabupaten Banyumas. Penelitian ini dilakukan di beberapa wilayah di Kabupaten Banyumas dengan menggunakan metode survei dengan teknik pengambilan sampel secara purposive sampling. Variabel yang digunakan yaitu morfologi dan bagian bambu yang dimanfaatkan dengan parameter yang diamati yaitu karakter morfologi akar, batang/buluh dan daun bambu serta pemanfaatannya oleh masyarakat. Data yang diperoleh dianalisis secara deskriptif untuk mengetahui keanekaragaman, hubungan kemiripan dianalisa dengan menggunakan Software MEGA 6.0 metode Unweight Pair Group Method Arithmatic Average (UPGMA), pemanfaatan bambu dianalisa secara deskriptif berdasarkan hasil wawancara. Berdasarkan hasil penelitian, terdapat 6 spesies dengan 2 varietas bambu yaitu yaitu Bambusa vulgaris Schrad. Ex Wendl. var. vulgaris (Bambu Ampel), Bambusa vulgaris Schrad. Ex Wendl. var. striata (Bambu kuning), Bambusa multiplex (Lour.) Raeusch. ex J. A. J. H. Schult (Bambu cina/cendani/gendani), Dendrocalamus asper (Schult.) Backer ex Heyne (Bambu Petung), Gigantochloa atter (Hassk.) Kurz (Bambu asak/ater), Gigantochloa apus (J.A. J. H Schult.) Kurz (Bambu Tali) dan Gigantochloa atroviolacea Widjaja (Bambu Wulung/Ireng). Hubungan kemiripan terdekat dimiliki oleh G. apus dan G. atroviolacea dengan nilai disimilaritas 0,11. Hubungan kemiripan terjauh dimiliki oleh B. vulgaris var. vulgaris dengan D. asper dengan nilai disimilaritas 2,19. Masyarakat di Kabupaten Banyumas memanfaatkan bambu sebagai buluhnya bahan bangunan dan kerajinan serta rebung dan daunnya untuk konsumsi. Kata kunci : Agroekonomi, Bambu, Banyumas, Keanekaragaman.

Abstract

Microfloriculture refers to souvenirs and Living Room Ornaments made from the in vitro cultured ornamental plants, which are innovative, unique, environmentally friendly, and can be produced across the year. In vitro pitcher-forming Nepenthes mirabilis will increase the attractiveness and market value of microfloriculture products. Two experiments have been carried out to stimulate in vitro pitcher formation in N. mirabilis using a completely randomized design (CRD) of a two-factorial treatment pattern with three replicates. The first experiment tested different Murashige and Skoog (MS) media formulations (full-strength, 3/4MS, 1/2MS, and 1/4MS) and phytagel concentrations (0, 2.5, 3.5, and 4.5 g l-1). The same media formulations and 6-Benzylaminopurine (BAP) concentrations (0, 0.5, 1.0, and 1.5 μM) were tested in the second experiment. The results showed that the explants cultured on 1/4MS media produced the greatest number of shoots (3.83-8.25) and leaves (26.17-26.75), the earliest pitcher formation (27.75±2.06 days), the highest number of pitchers (6.50±2.65), and pitcher-forming leaves (23.30±5.10%). Liquid media enhanced leaves (28.33±4.97) and pitchers (6.42±3.36) formation as well as pitcher-forming leaves percentage (21.37±8.01%). A 1/4MS supplemented with 0.5 μM BAP maximized pitcher-forming leaves (21.08±7.78%) and pitcher number (5.67±2.52). Adding 1.5 μM BAP on 1/4MS media produced the highest number of leaves (38.67±5.69). Full-strength MS produced the lowest number of shoots, leaves, pitchers, and pitcher-forming leaves. This study developed an affordable and effective method to improve in vitro pitcher development in N. mirabilis, highlighting how nutrient levels and BAP concentration influence pitcher formation, which aids in optimizing culture techniques for ornamental carnivorous plants.

Abstract

The pitcher plant (Nepenthes mirabilis (Lour.) Druce) is a unique plant listed in the Convention on International Trade in Endangered Species of Wild Fauna and Flora (CITES) Appendix II and is protected in Indonesia. Conventional propagation of N. mirabilis is difficult and takes a longer time. Therefore, in vitro culture method is proposed. This study aimed to determine the best and most economical media formulation and agar concentration for N. mirabilis micropropagation. This research has been carried out experimentally using a completely randomized two-factor factorial design. The first factor was the media formulation (full-strength Murashige and Skoog (MS), half-strength MS, half-strength MS + AB mix, and AB mix) and the second factor was agar concentrations (6, 8, and 10 g l-1). Twelve treatment combinations were obtained and repeated 5 times to produce 60 experimental units. The explants were apical microshoots (1.5 cm long with 5 leaflets). The cultures were incubated at 24 ℃ under continuous light for 16 weeks. The parameters measured included shoot emergence time, number of shoots, number of leaves, and shoot length. The data were analyzed using variance analysis followed by Duncan’s multiple range test at a 95% confidence level. The results showed that half-strength MS medium resulted in the highest number of shoots and leaves and the longest shoot length, whereas adding 8 g l-1 agar resulted in the fastest shoot emergence time. Half-strength MS medium solidified with 8 g l-1 agar could produce many N. mirabilis (Lour.) Druce microshoots to support both conservation and microfloriculture development.
