Abstract

Myoglobin (MB) gene encodes the protein of myoglobin, which is a protein found in muscle tissues that plays a crucial role in binding and storing oxygen. This study aimed to analyze the polymorphism of the MB gene, examine its relationship with the meat mineral content, and analyze its expression in the liver and thigh muscle tissue of IPB-D2 chicken. A total of 55 IPB-D2 chickens were used in this study. Identification of the gene polymorphism, quantification of the mineral content, and gene expression were performed using the direct sequencing method, the Atomic Absorption Spectroscopy (AAS) method, and the qRT-PCR, respectively. Chicken thigh muscle and liver tissues were used as the source of mRNA in this study. The statistical analysis methods used were the chi-square test to test Hardy-Weinberg equilibrium, and the T-test for mineral association analysis and gene expression analysis. IPB-D2 chicken on average contains 17.81, 0.22, 16.50, and 0.34 mg/kg of Fe, Se, Zn, and Mn, respectively. Two SNPs were found in 5’UTR of the MB gene, namely SNP g.17 G>T and SNP g.25 T>C. Genotype TT (g.17 G>T) and genotype CC (g.25 T>C) were significantly (P<0.05) associated with high Fe content in IPB-D2 chicken. The gene expression analysis showed that MB mRNA expression in the liver was not statistically different between high Fe (TT/CC) and low Fe (GG/TT and GT/TC) genotype combination. Meanwhile, in the thigh muscle, MB mRNA expression in TT/CC genotype combination was significantly higher than GG/TT and GT/TC genotype combination (P<0.05). These results suggest that the identified polymorphisms in the IPB-D2 MB gene could serve as references for investigating similar gene in other chicken breeds, especially regarding Fe, Se, Zn, and Mn mineral content.

Abstract

Research on the 11 morphometrics of IPB-D chicken in Sukabumi district has not been conducted. This study aimed to evaluate the morphometrics of IPB D1, IPB D2 and IPB D3 chickens in Sukabumi District. The design of this study was a completely randomized design with 3 levels of treatment linesnamely IPB D1, IPB D2 and IPB D3 with 95% confidence interval. Treatments that had a significant effect were followed by the Tukey test. Measured morphometric parameters such as shank length, shank circumference, femur length, tibia length, breast length, breast width, breast depth, breast circumference and back length. IPB D3 chickens show that their genetic condition is not yet stable as meat type. At the age of 4 to 8 weeks, IPB-D3 chickens have a larger size in the variables of shank circumference, femur length, tibia length, breast length and breast circumference. However, at the age of 8 to 12 weeks IPB-D3 chickens have a smaller size in the variables of breast length, breast width, breast depth, breast circumference and back length. Meanwhile, IPB D1 chickens show that they are genetically stable and IPB D2 is able to adapt because of its resistance traits.

Abstract

The transforming growth factor-beta 2 (TGF-β2) gene is a group of growth hormone (GH) genes that control the nature of growth and belongs to the cytokine group. This study aimed to identify Single Nucleotide Polymorphism (SNP) g.640 T>C from the TGF-β2 gene then associated with the growth properties of tolaki chickens. Chicken blood samples used in this study amounted to 70 birds. Data on the nature of growth observed include; hatched weight, feed consumption, body weight, body weight gain and feed conversion. The TGF-β2 gene polymorphism was identified using the PCR-RFLP method. SNP g.640 T>C in the TGF-β2 gene was polymorphic and wasin Hardy-Weinberg balance. Identification results found 2 alleles, namely T and C and 3 genotype namely TT, TC, and CC. The association of TGF-β2 gene diversity in tolaki chicken generally showed a difference (P<0.05) on feed consumption, body weight and body weight gain. CC genotype has higher feed consumption, body weight, weight gain and more efficient feed conversion than TC and TT genotypes.

Abstract

The aim of this study was to identify the polymorphism of the CD1B gene in c.248 in exon 3 and its association with IgY and ND titers in sentul chicken Sensi-1 Agrinak line. IgY is the main antibody of chickens while the ND titer is an ND specific antibody, both of which have an important role in the response of disease resistance. The Cluster of differentiation 1B (CD1B) gene is one of the major genes of disease resistance that functions in representing antigens through specific immune responses. This study used 17 sensi-1 chicken lines. The study was conducted by PCR sequencing method, ELISA test, and HI test. The result showed mutation on c248 A>G in exon 3. The SNP were polymorphic and Hardy Weinberg equilibrium. Thre were two A and G alleles and 3 genotypes namely AG, GG, and AA. The frequency of the GG genotype was higher (0.41) compared to other genotypes. The AG genotype were associated with IgY production and were not associated with ND titers.

Abstract

The TGF-β3 gene is a subtype of TGF-β superfamily that plays a crucial role in skeletal muscle growth. The objectives of this study were to identify the TGF-β3|BslI locus polymorphisms and to evaluate their effects on carcass components in Kampung and broiler chicken cross. A total of 129 chickens used in this study, consisting of: Kampung chickens (44), parent stock broilers Cobb strains (10), F1 Kampung x broilers (35), and F2 Kampung x broilers (40). The PCR-RFLP and direct sequencing techniques were used to obtain mutations in the intron 4 of TGF-β3 gene using the BslI restriction enzyme. The TGF-β3|BslI locus was polymorphic in all populations and three genotypes were found: CC, AA, and CA. No association was found between the three genotypes with the carcass components in F2 Kampung x broiler cross.

Abstract

IPB D-1 chickens were produced from research on the application of molecular genetics with the aim of increasing the productivity and quality of Indonesian local chickens. This study aims to determine the perception of farmers at Sinar Harapan Farm Jampang Tengah Sukabumi about IPB D-1 chickens, which is viewed from the characteristics of innovations in IPB D-1 chickens. The research was conducted at Sinar Harapan Farm (SHF) which was located in Jampang Tengah District, Sukabumi Regency. Data collection was carried out in August 2020. Data collection was carried out by interviewing all 18 member farmers of Sinar Harapan Farm. SHF farmers’ perceptions of IPB D-1 chickens were measured from five characteristics of innovations in IPB D-1 chickens which included: relative advantage, compatibility, complexity, trialability, and observability. The results showed that the perception score for the aspect of relative advantage was 4.34, compatibility was 4.38, compatibility was 4.25, trialability was 4.13, and observability was 4.15. The farmers of Sinar Harapan Farm Jampang Tengah Sukabumi showed a good perception about IPB-D1 chickens.

Abstract

The BG1 gene is a disease resistance candidate gene in chicken, which is located in the MHC region. The mechanism of the BG1 gene is known as an inhibitor of T lymphocyte cell activation. This study aims to identify the diversity of the SNP gene BG1 in the IPB-D2 chicken line. IPB-D2 chicken is a candidate for a new line from a cross from IPB-D1 chicken, which was selected for high body resistance and good productivity. A total of 27 DNA samples of IPB-D2 (G0) chickens and 58 samples of IPB-D2 (G2) chickens were sequenced to identify the diversity of BG1 genes. The study found 6 point mutations (g.4489 G>T, g.4712 C>T, g.4741 T>C, g.4747 T>C, g.4749 T>C, g.4761 G>A) and deletion of 2 nucleotide bases g.4758 - g.4759 delAT. The combination of 6 SNPs forms 4 haplotypes. The SNPs that changed the amino acid composition were SNP g.4489 G>T cys> phen, g.4712 C>T gln>codon stop, g.4749 T>C val>ala and g.4761 G> A gly> glu. All mutations are classified as polymorphic, low heterozygosity value, and not in Hardy-Weinberg equilibrium.

Abstract

Quality carcass and non-carcass are two inseparable components that result from slaughtering animals with high economic value. The aims study is effect of polimorfisme DGAT1 gene on hot and cold carcass, neck and non-carcass traits in Indonesia sheep. A total of 50 rams used in this study were collected from 10 Barbados Cross Sheep (BCS), 10 Compass Agrinac Sheep (CAS), 15 Javanese Thin-Tailed Sheep (JTTS), and 15 Jonggol Sheep (JS). The sheep were slaughtered at 10-12 months old with an average body weight of 20.45 kg. Identification of the DGAT1 gene polymorphism was performed by Polymerase Chain Reaction-Restriction Fragment Length Polymorphism (PCR-RFLP). The effect of the DGAT1 gene and breed with carcass and non-carcass traits were described using a T-test. The result showed that we found two genotypes: CC (466 bp), and CT (76, 390, and 466 bp) in sheep. The DGAT1 gene polymorphisms (g.8539 C>T) were in Hardy-Weinberg Equilibrium (HWE). The DGAT1 gene polymorphisms had a significant (P<0.05) on carcass traits in neck. However, the DGAT1 gene had no significant association (P>0.05) with non-carcass traits. The CT genotype had the highest value of carcass traits compared to CC genotypes. Therefore, the quality of carcass and non-carcass in Indonesian sheep for the DGAT1 gene only affects the neck value.

Abstract

Sentul chicken is one of Indonesia’s native chickens that has high potential for meat and egg production performance. The production of Sentul chicken breeds with high body resistance is one of the solutions to fulfill this potential. This research aimed to ascertain the impact of various IgY concentrations (high, medium, and low) on the production of 90 Debu and 90 Kelabu Sentul chickens. Parameters observed included pre-laying and laying performance. Six replications of a completely randomized design were used to conduct the experiment. The Anova test was used to analyze the data that was collected. The findings revealed that different strains and sex of Debu and Kelabu Sentul chickens did not significantly affect the value of IgY concentration. In the entire population studied, only 12.09% of Debu Sentul chickens and 11. 32% of Kelabu Sentul chickens had low Igy concentration values. In pre-laying period, the difference in the types of chicken strains has no significant effect on the body weight gain and thefeed convertion. Chickens with high IgY concentrations performed worse during the laying phase than hens with low IgY concentrations. The research found that the population of Kelabu Sentul chickens with high IgY concentration value is more than Debu Sentul chickens, which makes them suitable to be used as breeders.

Abstract

The increasing demand for chicken meat in Indonesia has encouraged the development of superior localchickens such as IPB D1, which have the ability to utilize local feed and exhibit rapid growth. Thisstudy aimed to determine the role of zinc, chromium, and lemuru oil supplementation on the digestivesystem development and internal organs functionality of IPB D1 chickens. A total of 100 twelve-weekold IPB D1 chickens were assigned to four treatment groups: control (P0), feed supplemented withzinc and chromium (P1), feed supplemented with lemuru oil (P2), and feed supplemented with zinc,chromium, and lemuru oil (P3). A Completely Randomized Design (CRD) was applied, and significantdifferences were analyzed using Tukey’s post hoc test. The findings revealed no significant impact ofsupplementation on the relative length of the digestive tract. Nonetheless, a significant increase in thepercentage of gallbladder weight was observed in the P3 group (0.09%–0.13%). In contrast, the heartweight percentage was significantly reduced compared to the control group (0.48%–0.58%). Theseresults indicate that combined supplementation of zinc, chromium, and lemuru oil may modulate lipidmetabolism and cardiovascular function in IPB D1 chickens. Further investigations are required toestablish optimal dosages and assess the long-term implications on the performance of IPB D1 chickens.
