Abstract

This research was done to identify the body size and shape  among three types of local sheeps of Garut fighting type, Garut meat type and their crossbred fighting and meat type in Garut district (Margawati, Sukawening and Wanaraja) and Bogor district (Ciomas and Cinagara).  The total number of sheep from Garut district used in this study were 520 head consisting of crossbred sheep from Margawati (71 head), fighting sheep from Wanaraja (79 head), meat sheep from Wanaraja (84 head), fighting sheep from Sukawening (87 head) and meat sheep from Sukawening (62 head). Whereas the total number of sheep from Bogor district were 137 head consisting of those of fighting sheep from Ciomas (66 head) and from Cinagara (70 head). Some body measurenments measured in this study were body weight, body length, wither height, chest width, chest circumfrence, cranium, tail length and tail width.  Analysis of all those sizes were based on principal component analysis (PCA) then visualized into the group of crowded diagram.  Different group of crowed builded on the base of scoring in body size and body shape derivated from covarian matrix were able to identifed morphologycal penotypic differences among garut sheep studied.   The results from canonical  analyses showed that body lenght, chest girth,chest width, and wither height were the most discriminant variables had impact to the diferences score size between  types of Garut sheep. However, the tail length and tail width were variables had impact to diferences score shape. The  crowed diagram showed that an intersection among three types (fighting, meat and crossbred) of sheeps suspected as resulted from the same genetic resource, namely Garut sheep.  Genetic flow was suspected from crossbred in Margawati to meat sheep in Cinagara as well as the sheeps from Sukawening and Wanaraja to fighting sheep in Ciomas.  Close genetic distances that were found among fighting and meat sheeps in Sukaweining as well as fighting and meat sheeps in Wanaraja could be considered that these two groups of sheeps as the genetic resource of fighting sheep in Ciomas. Abstract

The objective of this study was to estimate heritability of birth and weaning weights of the fighting type of Garut sheep. The data of birth and weaning weight of 175 SR (Super lambs, i.e. cross between fighting type Garut rams and selected Garut ewes) and 61 SB (Sukabumi lambs, i.e. cross between fighting type Garut rams and Sukabumi ewes) were used. The data were collected from March 2001 to August 2002. The results showed that the means of all traits of SR sheep group were larger (P < 0.01) than those of SB sheep group. Estimated heritability of birth weight and weaning weight of SR sheep were 0.67  ± 0.19 and 0.95  ± 0.16 respectively. Estimated heritability value of birth and weaning weight of SB sheep were 0.53  ± 0.33 and 0.57  ± 0.37 respectively. The heritability of birth and weaning weight were considered as high which means that the selection programme will be more effective and efficient in improving the genetic merits. Abstract

The aim of this study was to estimate genetic and phenotypic parameters of growth traits consisting of birth weight (BW), weaning weight (WW), 6 mo body weight (6WM), 12 mo body weight (12WM), and 18 mo body weight (18WM) of Ettawa Grade goats. The number of goat used to determine growth traits of BW, WW, 6WM, 12WM, and 18WM were 316; 316; 259; 259 and 165 heads, respectively. Data were analyzed using General Linear Model (GLM) to identify non-genetic effect. Estimation of genetic and phenotypic parameters including heritability, repeatability, genetic and phenotypic correlation were calculated using Restricted Maximum Likelihood and GLM. Genetic trends were calculated using the regression of mean breeding values on birth year. The results showed that parity and type of birth had significant (P<0.05) influence on all growth traits. Estimated heritability of birth, weaning, 6WM, 12WM, and 18WM were 0.54±0.12; 0.35±0.07; 0.37±0.09; 0.68±0.16 and 0.63±0.19, respectively. Estimated repeatability of WW, 6WM, 12WM and 18WM, WW, 6WM, 12WM and 18WM were 0.98±0.01; 0.97±0.01; 0.94±0.03; 0.71±0.12 and 0.91±0.04, respectively. The genetic trends for traits of BW and 18MW were decreased fluctuatively. However, the WW, 6MW, 12MW were increased fluctuatively. The high and positive genetic correlations between all growth traits and 12WM traits in this study indicated that selection for high 12WM will improve genetic merit in Ettawa Grade goats. Key words: body weight, EBV, Ettawa Grade goat, genetic and phenotypic parameters

Abstract

Calpastatin (CAST) gene is well known as an inhibitor of muscle protein degradation and relates to muscle growth and meat tenderness. The objective of this study was to determine the association of CAST gene with growth traits and carcass characteristics in Bali cattle. A number of data from 35 Bali bulls were collected from BPTU Bali Cattle to obtain growth traits, carcass characteristics, and blood samples. Polymorphism of CAST gene in Bali bulls was analyzed by using PCR-RFLP and DNA sequencing. The association of CAST gene with growth traits and carcass characteristics were analyzed by using General Linear Model (GLM). The result showed that there were two genotypes (GG and AG) of CAST gene with allele frequencies of 0.857 and 0.143, respectively, for G and A. Notably, mutation A to G occurred in 253 bp CAST fragment gene in Bali Cattle. Genotypes GG and AG of CAST gene significantly affected (P<0.05) the back-fat thickness and longissimus dorsi without a significant effect on the growth traits. It could be concluded that CAST gene had a potency as a marker gene for carcass quality in Bali cattle.  Key words: Bali cattle, CAST gene, PCR-RFLP, polymorphism

Abstract

Myostatin (MSTN) gene plays a key role in skletal muscle homeostasis such as inducing muscle athrophy, poliferation of myoblast, increasing ubiquitin-proteasomal, downregulating IGF pathway, and glucolysis. Myostatin gene expression is controled by CpG island located in promoter region. The objectives of this research were to identify polymorphism of MSTN promoter gene and to associate the polymorphism of SNP with growth and muscling traits in Bali cattle. A total of 48 Bali cattle from BPTU-HMT Bali island was screened to identify genetic polymorphisms in MSTN promoter region using sequencing method. The growth and muscling traits were measured at 12 months of age. The muscling traits were evaluated using ultrasound console with linear transducer having frequency 6.5 Hz and scaning we conducted at 130 mm in deep. Analysis of polymorphism was conducted by using PopGen 1.32 software. The association of MSTN with growth and muscling traits were analyzed by using General Linear Model (GLM) procedure. This result showed that a total of 20 polymorphic SNPs (seven SNPs in CpG island) were detected in this region. Although, only 3 SNPs (g.-8078C>T, g.-7996G>C, and g.-7930A>G) had equilibrium condition in Hardy-Weinberg analysis. The association result showed that 2 SNPs (g.-7799T>C and g.-7941C>T) were significantly associated with intramuscular fat percentage (P≤0.05) in Bali cattle. Although the 2 SNPs were nominally significant at nominal P≤0.05 threshold, they were not significant after Bonferroni correction for multiple testing. It could be concluded that MSTN promoter gene was polymorphic in Bali cattle and there were 2 SNPs associated with carcass quality.

Abstract

Information on animal genetic resource is needed in conservation program of Indonesian local buffalo. This study was aimed to analyze the quantitative traits diversity and genetic distance of local buffalo of Southeast Sulawesi local based on morphological approach. The total of 271 heads of swamp buffaloes were used in the study from four sub-populations, namely Bombana island’s, Bombana mainland, Kolaka and Konawe. Duncan Multiple Range test at 5% probability was used to compare the quantitative traits between the buffalo sub-populations. Genetic distance was analyzed by using the simple discriminant function. The results showed that the average body weight of male buffalo between sub-populations was not significantly different (P>0.05), but the average body weight of females buffalo from Bombana Island’s (465.22 ± 103.25) was significantly (P<0.05) higher than those of the other sub-populations. The highest variation of quantitative traits of local buffalo was found in Konawe sub-population, i.e. in body weight of female buffalo (35.36%) and body weight of male buffalo (22.05%), rump length in males buffalo (20.44%), and rump width in females buffalo (14.58%). Southeast Sulawesi local buffalo can be grouped into three clusters based on the phylogeny tree, namely 1) Kolaka and Konawe cluster, 2) Bombana island’s cluster, and 3) Bombana mainland cluster. The distinguishing variables of the group of Southeast Sulawesi local buffalo were rump length, body length, and chest circumference. The highest phenotype similarity value was shown by Bombana island’s buffalo (68.57%), while the lowest was found at Konawe buffalo (26.76%). The farthest genetic distance was found between Bombana Islands and Konawe buffalo (3.71703), while the closest genetics distance was identified between Kolaka and Konawe buffalo (0.86616).

Abstract

Abstrak Metode yang digunakan dalam penelitian ini ialah menggunakan Penelitian Tindakan Kelas (PTK) yang berusaha memecahkan atau menjawab masalah yang dihadapi pada subjek penelitian. Subjek penelitian ini adalah peserta didik Kelas X Multimedia SMK Karsa Mulya Palangka Raya dengan jumlah 34 peserta didik. Teknik dalam pengumpulan data dalam penelitian ini adalah observasi dan tes. Analisis yang digunakan yaitu analisis kualitatif dan analisis kuantitatif. Hasil penelitian ini menunjukan bahwa menggunakan media video dapat meningkatkan hasil belajar peserta didik pada pembelajaran Desain Grafis kelas X Multimedia SMK Karsa Mulya Palangka Raya. Hal ini berdasarkan hasil penelitian hasil penelitian yang didapatkan nilai dalam kegiatan pembelajaran yang dilakukan selama dua siklus, bahwa dapat disimpulkan: penggunaan media video dapat meningkatkan hasil belajar peserta didik, dapat dilihat dari rata-rata pra tindakan dengan nilai 43,82, rata-rata siklus I dengan nilai 74,56 dan pada siklus II memperoleh nilai 91,03 dengan ketuntasan klasikal 100%. Abstract

Dalam dunia kerja, komitmen seseorang terhadap organisasi atau perusahaan seringkali menjadi isu yang sangat penting. Saking pentingnya hal tersebut, maka beberapa organisasi seringkali memakai unsur komitmen sebagai salah satu syarat untuk memegang suatu jabatan/posisi yang ditawarkan dalam iklan-iklan lowongan pekerjaan. Sayangnya meskipun hal ini sudah sangat umum namun tidak jarang pegawai masih belum memahami arti komitmen secara sungguh-sungguh. Tujuan yang ingin dicapai di dalam penelitian ini adalah untuk mengetahui pengaruh faktor – faktor yang mempengaruhi komitmen kerja terhadap kepuasan serta dampaknya pada kinerja pada Perusahaan Daerah Bank Perkreditan Rakyat Kabupaten Cirebon. Metode penelitian yang digunakan dalam penelitian ini adalah metode survey. Hasil kesimpulan penelitian ini yaitu bahwa thitung untuk variabel komitmen normatif sebesar 2.886. Jadi nilai thitung variabel adalah lebih besar dari ttabel (2.886 > 1.670), sehingga secara parsial variabel komitmen normatif mempunyai pengaruh terhadap kepuasan kerja pegawai, bahwa thitung untuk variabel komitmen afectif sebesar 4.845. Jadi nilai thitung variabel adalah lebih besar dari ttabel (4.845 > 1.670), sehingga secara parsial variabel komitmen afectif mempunyai pengaruh terhadap kepuasan kerja pegawai, bahwa thitung untuk variabel komitmen kontinues sebesar 3.960. Jadi nilai thitung variabel adalah lebih besar dari ttabel (3.960> 1.670), sehingga secara parsial variabel komitmen kontinues mempunyai pengaruh terhadap kepuasan kerja pegawai, dan bahwa thitung untuk variabel kepuasan kerja sebesar 4.717. Jadi nilai thitung variabel adalah lebih besar dari ttabel (4.717 > 1.670), sehingga secara parsial variabel kepuasan kerjai mempunyai pengaruh terhadap kinerja pegawai. Kata kunci : Komitmen Normatif, komitmen afectif, komitmen continues, kepuasan kerja dan kinerja pegawai.

Abstract

The purpose of this research was to determine the effect of heat stress on thermoregulation, haematology, and productivity of Holstein Friesian (HF) dairy cows raised in different elevations. A total of 63 HF in a normal lactation period were used in this study. The research was conducted for 3 months during dry season in three different areas, which were at Pondok Ranggon (97 m.a.s.l) which categorized as a lowland, Ciawi (576 m.a.s.l) which categorized as a lower-upland, and Lembang (1241 m.a.s.l) which categorized as an upland. Observation on microclimate aspects which includes environmental temperature (Ta), relative humidity (RH) and Temperature-Humidity Index (THI) was done by recording each variable for every 2 hours starting from 08.00 to 16.00 WIB. The thermoregulation analysis was done based on the given physiological responses which consisted of the skin temperature (Ts), rectal temperature (Tr), body temperature (Tb), heart rate (Hr), respiratory rate (Rr), Heat Tolerance Coefficient (HTC), plasma cortisol level and haematological profile observation. The Ta, Rr, and THI measurements showed that in the lowland and lower-upland, the HF experienced moderate heat stress, while the HF raised in the upland area experienced less heat stress. The results showed that the dairy cows which raised in lowland had the highest HTC, Tr, Ts and Tb (P<0.05) and lowest Hr (P<0.05). All of the physiological and haematological parameters in the three study area showed a normal value.Furthermore, the erythrocyte, Hb and PVC concentration in a lowland raised HF were higher (P<0.05), while the plasma cortisol levels were not significantly different. The milk production of the observed dairy cows in different elevations was significantly different (P<0.05), with the highest milk yields, were found in the upland raised HF (13.1±3.52 kg), followed by the lower-upland (11.3±4.73 kg) and lowland (7.0±3.36 kg). In general, all of the HF raised in different land elevations was exposed to heat stress during dry seasons, even though the cows showed the ability to physiologically adapt and cope with the conditions.
