Abstract

Background: Helicobacter pylori is a well known pathogen which is recognized as the most frequent infection of individuals. Helicobacter pylori plays important role in the etiology og gastritis, gastric and duodenal ulcers. We investigated the presence of Helicobacter pylori as an extragastric reservoir in the tonsilectomy specimens. Aim: To determine the presence of Helicobacter pylori in cases of chronic tonsillitis in tonsilectomy specimens. Methods: This is a descriptive study with cross sectional which the sample will be obtained using consecutive sampling method. We collect 43 cases with chronic tonsilitis were examined in tonsillectomy specimens for which available tissue for routin staining could be retrieved were histopathology chronic tonsillitis. The colonization of the helicobacter pylori have been evaluated with hematoxylin & Eosin and Giemsa stains under the light microscope. Results: With H&E staining, Helicobacter pylori has been detected in 23 cases (53,48%) of the tonsil-lectomy specimens in total. No colonization has been observed in the remaining 20 cases (46,51%). With giemsa staining Helicobacter pylori has been detected in 29 cases (67,74%) of the tonsillectomy specimens in total. No colonization has been observed in the remaining 14 cases (32,55%). Abstract

ABSTRAKLatar BelakangMatrix Metalloproteinase 9 (MMP-9) merupakan enzim proteolitik yang diduga berperan pentingdalam proses progresifitas lesi prakanker menjadi kanker serviks. Kadar MMP-9 yang tinggi akanmenyebabkan proses degradasi jaringan serviks menjadi lebih cepat dan mempermudah prosesinvasi sel kanker. Penelitian ini mengevaluasi level immunoekspresi dari MMP-9 secaraimmunohistokimia pada berbagai spektrum lesi prakanker dan lesi karsinoma serviks invasif.Bahan dan CaraLima puluh sampel jaringan serviks dibagi dalam tiga kelompok yaitu lesi prakanker Low gradeSquamous Intraepithelial Lesion (LGSIL), lesi prakanker High Grade Squamous IntraepithelialLesion (HGSIL), dan lesi karsinoma invasive. Sampel akan dievaluasi intensitas dan kuantitasekspresi immunohistokimia MMP-9, kemudian diamati dan dianalisa dengan uji korelasi Spearmen.HasilJumlah sel yang terwarnai diffuse pada stroma serviks ditemukan pada 23 (46 %) kasus, 12 (26%)kasus pada karsinoma invasif dan 9 (18%) kasus HGSIL. Stroma pada lesi LGSIL lebih banyakyang tidak menampilkan IHK MMP-9. Pada stroma neoplasia intraepitel dan karsinoma invasifserviks ditemukan korelasi positif dan signifikan dengan ekspresi imunohistokimia MMP-9 baik darisegi intensitas pewarnaan maupun jumlah sel yang positif terwarnai.KesimpulanSemakin tinggi nilai positif ekspresi imunohistokimia MMP-9 yang dinilai berdasarkan intensitas dankuantitas, semakin tinggi derajat keparahan lesi neoplasia serviks. Peningkatan ekspresiimunohistokimia MMP-9 berperan penting dalam proses karsinogenesis tumor serviks

Abstract

Background Prostate cancer is the fourth ranked malignancy and the eighth leading mortality worldwide. The Gleason grading system is used to evaluate the tumor grade. The highest the score, the worst and the most aggressive the nature of the tumor, which associated with invasive behaviour. In prostate carcinoma, Matrix Metalloproteinase-9 (MMP-9) is involved in all stages of cancer cell progression include proliferation, angiogenesis, apoptosis, epithelial-mesenchymal transition (EMT) and metastasis. MMP-9 degrade the surrounding tumor matrix and neural tissue during perineural invasion in prostate carcinoma which is a prerequisite for invasion and metastasis. The aim of this research is to evaluate the correlation of MMP-9 immunohistochemical expression with the Gleason Grading System International Society of Urological Pathology (ISUP) of prostate adenocarcinoma. Methods Cross sectional analytic study of 32 samples were diagnosed with prostate adenocarcinoma at H. Adam Malik General Hospital Medan. Staining was performed using a polyclonal antibody rabbit MMP-9 immunohistochemistry (Bioenzy) which would appear positive in the cytoplasm and in stroma of the tumor cell at a dilution of 1: 100 (overnight). MMP-9 expression was assessed semi quantitatively based on intensity and tumor cell distribution with four-tier expression categories. Data analysis was tested with the Kruskal-Wallis test. Results Most cases (56,25%) were strongly positive with MMP-9 and MMP-9 immunohistochemical expression has a significant relationship with Gleason Grading ISUP System of prostate adenocarcinoma (p:0.001). Conclusion There was a significant association between MMP-9 immunohistochemical expression and the Gleason Grading ISUP System of prostate adenocarcinoma.
