Abstract

Lactobacillus plantarum S34 dilaporkan mempunyai aktivitas antibakteri yang terkait dengan produksi bakteriosin. Bagian dari gen yang menyandikan salah satu lokus bakteriosin yang diproduksi oleh L. plantarum S34, disebut dengan plantarisin W (plnW), diamplifikasi dari plasmid dan dikloning menggunakan sistem vektor pGEM®-T Easy ke dalam Escherichia coli DH5?. Sekuens nukleotida plnW (± 405 pb) diidentifikasi sebagai protein integral membran. Lebih lanjut, plnW diekspresikan secara heterologus sebagai fusi protein dengan His(6)-tag tioredoksin menggunakan vektor ekspresi pET-32a(+) ke dalam E. coli BL21 (DE3) pLysS. Protein fusi rekombinan plnW terdapat dalam sitoplasma sel, tetapi selain fraksi terlarut terdapat juga fraksi tidak terlarut berupa badan inklusi. Purifikasi parsial dilakukan menggunakan kromatografi afinitas ligan Co2+ untuk fraksi terlarut dan metode elektroelusi gel poliakrilamid untuk fraksi tidak terlarut. Massa molekul berukuran kurang lebih 33 kDa terdeteksi berdasarkan pemisahan SDS-PAGE dan dikonfirmasi dengan Western blot sebagai protein fusi rekombinan plnW. Protein yang sudah terpurifikasi bermanfaat untuk mengetahui kaitan antara struktur dan fungsi bakteriosin.

Abstract

On November 11, 2017, MUI (Indonesian Council of Ulama), DMI (Indonesian Mosque Council) have launched the national program named eco masjid. Eco masjid is a program of sustainable mosque through efforts to preserve the environment and natural resources oriented to on aspects of idarah (management), imarah (prosperity activities), and ri’ayah (maintenance and facilities) which dominantly related to its building. In Indonesia, sustainable building principles are widely promoted by GBCI (Green Building Council Indonesia) and have been applied to various types of buildings. However, it has been realized that the study on the implementation of sustainable principle in the mosque is very rare. The research was carried out qualitatively by using direct observation on the pilot mosque to capture all implemented sustainable building principle. Based on the questionnaire to 44 participants which reflected 44 DKM (Dewan Kemakmuran Masjid), the result came up that applying water efficient faucet, using the LED lamp and sticker posting are the most achievable sustainable practices in the existing mosques. The authors suggest to all related bodies to determine the standard on how to assess the implementation of eco Masjid program

Abstract

Penentuan isoflavon asal limbah tahu telah dilakukan.Ekstrak kasar isoflavon asal limbah tahu diperoleh dengan cara menghidrolisis menggunakan HCl 4 N diperoleh rendemen sebanyak 4,81 g/l. Kadar isoflavon diukur menggunakan metode Graham (1999) yang sebelumnya dilakukan optimasi enzim NADH oksidase diperoleh panjang gelombang optimum pada 327 nm pada suhu 350C, pH 5, konsentrasi substrat optimum 100 μM, konsentrasi enzim optimum 0,3 U/ml. Kadar isoflavon dalam limbah tahu 0,123x103 ppm sedangkan kadar isoflavon kedelai sebesar 0,5921x103 ppm. Sehingga kadar isoflavon limbah tahu adalah sebesar 20% dibanding kadar isoflavon kedelai.

Abstract

Red yeast rice (RYR) and rice bran (RB) have been reported to have many biological activities, such as anticancer, antioxidant, antidiabetes and antihypercholesterolemia. This research aimed to determine the best fraction of RYR extract as a HMG CoA reductase inhibitor and the best ratio of the mixture of RYR and RB extract fraction as a HMG CoA reductase inhibitor and an antioxidant using in vitro assays. RYR and RB were individually extracted using ethanol 95%, followed by liquid-liquid fractionation using n-hexane, dichloromethane, etyhl acetate, and water. The RYR extract fraction with the best HMG CoA reductase inhibition was then mixed with RB extract in various ratios for HMG CoA reductase inhibition test and antioxidant activity by FRAP method, as well as analysis of total phenolics and flavonoids content. The results showed that the strongest inhibitory effect to the HMG CoA reductase was found in the water fraction of RYR extract (37.37%). The mixture of RYR and RB water extract fractions with a ratio of RYR:RB 1:3 had the strongest inhibition (51.85%) as compared to other ratios, followed by a ratio of 2:2 (51.03%) and 3:1 (42.36%). The best antioxidant activity was found in the RYR:RB ratio of 1:3 (24.76 mg TE/g). The highest total phenolic and flavonoid content was found in the RYR:RB ratio of 2:2 (11.17 mg GAE/g) and the RYR:RB ratio of 3:1 (7.44 mg QE/g). Overall, the addition of RB extract into the RYR extract was able to provide a significantly better HMG CoA reductase inhibitory effects and antioxidant acti-vity than RYR extract alone.

Abstract

Dextransucrase is a glucasyltransferase that catalyzes the transfer of an alpha-D-glucopyranosyl group from sucrose to dextran and releasing the fructose. Enzyme activity was determined by measuring the initial reaction rate of fructose released from sucrose. A standard unit (U) is defined as the amount of enzyme that catalyzes the formation of 1 umol of D-fructose per mg of protein per minute. Dextransucrase was produced by fed-bath cultures of isolate bacterial from sugar cane in a mixture containing sucrose (10 % b/v) in themedium fermentation. Extracellular dextransucrase was obtained from bacterial isolate from sugar cane. Final culturedextransucrase activity was 0,2776 Ulmg of protein. Dextransucrase purified by ethanol 80 % showed activity of 4,2077 Ulmg of protein or activity increase 1515,75 %. Temperature optimum was 40 OC and pH optimum was 7.6. All of the metal increased the activity, except HgC12. Dectransucrase kinetics constants, Km is 3,00 mM and Vmaks is 3,5559 unit/mg of proteinlmin. Molecular weight is 95.155 kDa.

Abstract

The aim of this research was to produce tannin acylhydrolase (tannase) from Aspergillus niger isolated from cacao pod. The first step of the study included determination of optimal pH, temperature, and incubation period to produce tannase. Optimal conditions obtained for tannase production were pH 5.5, a temperature of 28 oC and an incubation period of 3 days. Optimization of production medium was conducted. The media tested were solid and liquid wheat flour media with a concentration of tannic acid as inducer at 0, 3, 5, and 7% (wt/vol). The best production medium was solid medium with tannic acid concentration of 5% (wt/vol).

Abstract

A high rate and massive illegal logging cases from the past until to date, and the low rate of law compliance in the sense of Indonesia law and policy implementation, awoke some issues that encourage Indonesia and Indonesian jurists to build a better system. Abstract

The prevalence of hypercholesterolemia in Indonesia reaches 11.2%. Hypercholesterolemia can occur due to lack of exercise, consume too much food containing fat and cholesterol, and lack of fiber intake. Rice bran and red yeast rice contain fiber and phytochemical compounds that can reduce cholesterol, including alkaloids, saponins, tannins, flavonoids, and phenolics. The purpose of this study is to use rice bran and red yeast rice as an alternative snack that can potentially bind cholesterol. Rice bran and red yeast rice are added to agar to obtain gelatin snacks with high fiber content and phytochemical compounds. Agar is made into three formulas. Hedonic rating tests have been carried out, so that obtain the formula that is most preferred by consumers. The test results show that the formula with the addition of sodium citrate of 0.3% is most preferred, with the addition of bran and red yeast rice each of 30 and 0.8 grams and the use of agar for 7 grams. Gelatin products contain 14.80% food fiber and total flavonoids by 0.02%. The total phenolic was not detected in agar products. The results of the cholesterol-binding test in vitro showed that gelatin products can bind cholesterol by 60.11%.

Abstract

Angkak dan bekatul merupakan pangan fungsional yang berpotensi sebagai agen terapi untuk mengendalikan gula darah, dikarenakan kandungan antioksidan alaminya. Tujuan penelitian adalah untuk mengevaluasi pengaruh campuran ekstrak angkak dan bekatul dalam menghambat peroksidasi lipid serta efeknya terhadap organ pankreas, hati dan ginjal tikus hiperglikemik secara histopatologi. Tikus dibagi ke dalam 7 kelompok yaitu kelompok kontrol normal, kontrol negatif, kontrol positif, formulasi ekstrak angkak bekatul kelompok I, II, III dan IV (20:1400, 40:700, 30:1050, dan 60:2100 mg/kg BB). Parameter pengujian yang dilakukan adalah konsentrasi malondialdehid (MDA) dan aktivitas enzim aminotransferase (ALT/AST) serum darah tikus dan histopatologi organ pankreas, hati dan ginjal. Campuran terbaik dalam menghambat pembentukan MDA dan peningkatan enzim ALT/AST adalah kelompok III dengan konsentrasi MDA (3,.31±0,.53 nmol/ml) dan aktivitas enzim ALT (35,.80±5,.63 U/l) tidak berbeda nyata dengan kontrol normal (4,.85±2,.32 nmol/ml) dan (27,.85±8,.13), sementara aktivitas enzim AST (67,.221±16,.05) mengalami penurunan namun berbeda nyata dengan kontrol normal (4,.365±0,.00). Kondisi histopatologi organ pankreas, hati, dan ginjal tikus semua kelompok perlakuan menunjukkan kemampuan dalam memperbaiki kerusakan organ. Kesimpulannya, campuran angkak dan bekatul 30:1050 mg/kg BB memiliki kemampuan terbaik dalam meghambat peroksidan lipid.Angkak and Rice Bran Extract to Prevent Lipid Peroxidation in Spague-Dawley Rats HyperglycemicAbstractAngkak and rice bran is a functional food that has potential as a therapeutic agent to control blood sugar. This potential is due to the content of natural antioxidants in red yeast rice and rice bran. The objective of this research was to evaluate the effect of angkak and rice bran extract as lipid peroxidation inhibition. The effect on the pancreatic, liver and kidney organs in hyperglycemic rats by histopatological study was also analyzed. Rats were divided into 7 groups namely normal control group, negative control, positive control, rice bran extract formulation on gorup I, II, III, IV (20: 1400, 40: 700, 30: 1050, and 60: 2100 mg/kg BW). The analysis parameters carried out were the concentration of malondialdehyde (MDA), the activity of the serum alanine/aspartat aminotransferase (ALT/AST) enzymes and rat histopathology of pancreatic, liver and kidney organs. The best mixture of angkak and rice bran to inhibit the formation of MDA and increase the ALT/AST enzyme was group III with MDA concentration (3,.31±0.,53 nmol/ml) and ALT enzyme activity (35.,80±5,.63 U/l) was not significantly different from normal control (4,.85±2,.32 nmol/ml) and (27.,85±8,.13), while the enzyme activity of AST (67,221±16,.05) decreased but was significantly different from normal controls (4,365±0,.00). Histopathological conditions of pancreatic, liver, and kidney organs of rats in all treatment groups showed the ability to repair damaged organs. In conclusion, the mixture of red yeast rice and rice bran 30:1050 mg/kg BW had the best ability to inhibit lipid antioxidants.

Abstract

Belimbing wuluh merupakan tanaman jenis buah dan obat tradisional. Tanaman belimbing wuluh sudah sering dimanfaatkan masyarakat salah satunya untuk mengobati penyakit seperti batuk dan radang rektum. Tujuan penelitian ini adalah menguji senyawa fitokimia, menganalisis kandungan total fenolik dan flavonoid, serta aktivitas antioksidan dan antiinflamasi secara in vitro pada ekstrak etanol daun belimbing wuluh. Metode yang dilakukan pada penelitian ini adalah ekstraksi daun belimbing wuluh, skrining fitokimia, perhitungan total fenol dan flavonoid, uji aktivitas antioksidan, dan uji aktivitas antiinflamasi. Senyawa fitokimia yang terkandung pada ekstrak etanol daun belimbing wuluh adalah saponin, tanin, steroid, flavonoid, dan alkaloid. Kandungan total fenolik dan flavonoid ekstrak etanol daun belimbing wuluh secara berturut-turut sebesar 39,03 dan 97,28 µg QE/mg. Ekstrak etanol daun belimbing wuluh memiliki aktivitas antioksidan yang tergolong sangat kuat, sementara aktivitas antiinflamasinya terutama ditunjukkan pada konsentrasi ekstrak 200 µg/ ml, memiliki nilai persen inhibisi hemolisis yang paling tinggi. Kesimpulannya, ekstrak etanol daun belimbing wuluh dapat menjadi salah satu sumber antioksidan dan antiinflamasi alami.Ethanol Extracts of Averrhoa Bilimbi Leaf Demonstrated Antioxidative and Anti-inflammatory ActivityAbstractAverrhoa bilimbi, a fruit-bearing plant, has been traditionally used for medicinal purposes such as treatment of cough and rectal inflammation. In this current work, ethanol extract of Averrhoa bilimbi leaf was analyzed, with regard to phytochemical composition, i.e. total phenolic, and flavonoid, antioxidant activity, and in vitro antiinflammation activity. Extraction of carambola leaves, phytochemical screening, total phenolic and flavonoid contens, assay of antioxidant activity, and assay of anti-inflammatory activity were done in this research. As a result, the leaf extract positively contained some phytochemical compounds, i.e. saponin, tannin, steroid, flavonoid, and alkaloid. Furthermore, total phenolic and flavonoid of the leaf extract was found at 39.03 and 97.28 µg QE/mg extract, respectively. Additionally, antioxidant activity of the leaf extract was classified as very strong, while its anti-inflammatory feature at extract concentration of 200 µg/ml exhibited the highest inhibition of hemolysis. In summary, the ability of Averrhoa bilimbi leaf ethanol extracts to act as antioxidative and anti-inflammatory agents was determined and this may open the use for natural antioxidant and anti-inflammatory agents.
