Abstract

Colorectal cancer has provided a paradigm for the connection between inflammation and cancer. Modulation of intracellular signaling involved in chronic inflammatory response by anti-inflammatory agents represents an important strategy in molecular target-based chemoprevention. The aim of this research is to study the effect of buah merah oil on  IL-6  serum level and histopathology of colon in colitis-associated cancer (CAC) mice model. BALB/c mice were randomly divided into four groups. The negative control and buah merah control were given aquabidest and buah merah, respectively, without CAC induction. The AOM/DSS control and buah merah treatment were given azoxymethane (AOM) followed by dextran sulfate sodium (DSS) to induce CAC. The AOM/DSS control was given aquabidest while the buah merah treatment was given buah merah. Data were analyzed by One-Way ANOVA continued with Tukey-HSD. The result showed that IL-6 level in mice administrated with buah merah was significantly lower than the AOM/DSS control (p=0.000). Histopathological analysis scores  of colon were analyzed by Kruskal-Wallis continued with Mann-Whitney. The result showed that histopathological analysis score in buah merah treatment was significantly lower than in the AOM/DSS control (p=0.002). Taken together, buah merah oil lowered IL-6 level and histopathological analysis score of colon in CAC mice model. Keywords: CAC, buah merah oil,  IL-6 level, histopathological  analysis scores of  colon

Abstract

BACKGROUND: Inflammation is a key component of the tumor microenvironment. Cancer-associated inflammation is marked by the presence of specific inflammatory cells and inflammatory mediators. Moreover, immune cells in the tumor microenvironment not only fail to mount an effective antitumor immune response, but also interact intimately with the tumor cells to actively promote oncogenesis.CONTENT: Their roles in regulating cytokine-dependent inflammation and immunity make the signal transducer and activator of transcription (STAT) proteins critical players in determining whether immune responses in tumor microenvironment promote or inhibit cancer. Recent evidence suggests a crucial role for STAT family proteins, especially STAT3 (signal transducer and activator of transcription 3), which is a point of convergence for numerous oncogenic signaling pathways, in selectively inducing and maintaining a procarcinogenic inflammatory microenvironment, both at the initiation of malignant transformation and during cancer progression. The persistent activation of STAT3 increases tumor cell proliferation, survival, and invasion while supressing antitumor immunity. SUMMARY: STAT3 signaling pathway constitutes a potential preventive and therapeutic target for cancer immunotherapy.  KEYWORDS: STAT3, inflammation, tumor microenvironment, cancer immunotherapy

Abstract

Background: Routine physical exercise is often used as an intervention to improve lung function and optimal lung function will improve physical fitness. This study aimed to examine the effect of rope jumping and Tabata exercise in lung function as represented in Forced Expiratory Volume in 1 Second (FEV1), Forced Vital Capacity (FVC) and physical fitness index. Methods: This is an analytic comparative experimental study with pretest and posttest design, involving 60 adult male, separated into 2 groups, which are rope jumping or Tabata exercise for 2 weeks period. The study was conducted at the Faculty of Medicine of Maranatha Christian University Bandung in December 2017 to September 2018. The measured data were FEV1 and FVC using spirometry; and physical fitness index using Harvard Step Test. Data with normal distribution was analyzed using paired T Test, data with abnormal distribution using Mann-Whitney Test, and between group analysis using unpaired T Test. Result: We found a significant increase in FEV1 (P=0.017) but not FVC (P=0.151) in rope jumping grup, and a significant increase in FEV1 (P=0.010) and FVC (P=0.014) in Tabata exercise group. Both physical fitness index in rope jumping group and Tabata exercise group increase significantly (P=0.0005). There are no significant difference between rope jumping and Tabata exercise group in the effect towards FEV1 (P=0.853), FVC (P=0.876) and physical fitness (P=0.545). Conclusion: Rope jumping and Tabata exercise increase lung function and physical fitness index.

Abstract

BackgroundSkin aging is a complex biological phenomenon influenced by intrinsic and extrinsic factors. Photoaging can be prevented by applying phytochemicals that have sun-protective properties. This study aimed to evaluate the effect of saffron serum to restore collagen density and autophagy processes and reduce inflammatory gene expression in UVB-exposed Wistar rats. MethodsAn experimental laboratory study was conducted involving 20 male Wistar rats that were divided into 4 groups: control, UVB, UVB + base serum, UVB + saffron serum exposed to UVB radiation for 5 weeks with a total dose of 3100 mJ/cm2. The skin was extracted then underwent Masson Trichrome staining and real-time PCR to obtain collagen density and gene expression. ResultsThe gene expression of MMP1, IL6, TNFá, LC3, and p62 was significantly increased in the UVB group compared to the control group. Topical administration of saffron serum significantly increased collagen density (p=0.001). Induction by UVB significantly increased LC3 (p=0.020) and p62 (p=0.030) gene expression, indicating an inhibition of autophagy. The saffron serum might modulate autophagy by increasing LC3, but not significant (p=0.495) and significantly decreasing p62 gene expression (p=0.001). As for MMP1, IL6, and TNFá, no significant decrease in gene expression was found in the UVB + saffron serum group compared to the UVB group. ConclusionSaffron serum increases collagen density and modulates autophagy in the skin of UVB-exposed Wistar rats. Inflammatory markers were increased after UVB induction, but no differences were found after saffron serum topical administration.

Abstract

Aging is influenced by several factors, including Reactive Oxygen Species (ROS). An imbalance of higher ROS and lower antioxidants may lead to oxidative stress in cells that is manifested as wrinkles, dark spots, and fine lines. Antioxidants can prevent the effects of ROS. Safe and abundant sources of antioxidants can come from natural ingredients. One of the natural ingredients widely studied is Clitoria ternatea flower extract (CFE). This study aims to report the CFE potential as an anti-aging and antioxidant agent. Gallic acid and quercetin standards were used to measure the total flavonoid and phenolic content. Antioxidant activity was evaluated by hydroxyl (OH) and Nitric Oxide (NO) scavenging, while anti-aging activity was evaluated by collagenase and elastase inhibition tests. CFE showed high phenol and flavonoid content (14.49 μg GAE/mg and 9.00 μg QE/mg). CFE also has OH and NO scavenging activity with IC50 18.39 and 31.11 μg/mL, which exhibited antioxidant potential. CFE also showed potential to inhibit collagenase (IC50 = 177.48 μg/mL) and inhibit elastase (IC50 = 30.88 μg/mL). CFE may serve as a promising candidate with antioxidant and anti-aging properties.
