Abstract

Periodontitis is an inflammation disease of the periodontal tissues surrounding the teeth, include periodontal ligament fibroblast cell. The recently researches developed tissue engineering concept to accelerate regeneration and proliferation periodontal ligament cells. One of the major components of tissue engineering is scaffold. The hydroxyapatite is biomaterial that can be used as scaffold. The egg shell has the potential biowaste to be a source of hydroxyapatite biomaterial. Before being applied as a scaffold in the medical field, cytotoxicity testing needs to be done. This study aimed to examine toxicity effect of egg shell hydroxyapatite in various concentration toward human periodontal ligament fibroblast cells. This study used human periodontal ligament fibroblast primary cells (5x103) in microplate 96 wells which divided into 7 groups, control cells, control media without cells and the treatment groups with concentration egg shell hydroxyapatite100 μg/ml, 75 μg/ml, 50 μg/ml, 25 μg/ml dan 25 μg/ml and incubated for 24 hours. Cytotoxicity test was conducted using MTT assay. Optical density values describe the viability of living cells and the readings were done using ELISA readers, subsequently the data were analyzed using One way ANOVA. The number of human periodontal ligament cells at all concentration showed that the percentage of cell life is more than 90%. Egg shell hydroxyapatite at all concentrations tested is not toxic toward human periodontal ligament fibroblas cells.

Abstract

Burn therapy involves elimination of causative factors and symptomatic therapy. Kenitu leaves (Chrysophyllum cainito L.) are known to contain β-amylin acetate, Gallic acid, ursolic acid, quercetin, β-sitosterol, and other substances that have the potential to be antibacterial, antioxidant, and anti-inflammatory. The use of kenitu leaves (Chrysophyllum cainito L.) as an alternative to wound healing has low side effects and is easy to obtain, especially in Jember Regency. The aim of this study was to examine the effect of kenitu leaves (Chrysophyllum cainito L.) leaf extract gel on re-epithelialization in buccal mucosal burns of wistar rats. This type of research is experimental laboratories. This research used 32 samples which divided into four control groups and four treatment groups. Burns are made on the left buccal mucosa. In the control group the CMC placebo gel was given and the treatment group was given 2% leaf extract gel. Euthanasia was performed on days 4, 7, 10, and 13 then histological preparations were made. Measurement using software optilab viewer 2.2 and image raster 3.0. The average yield of epithelial thickness was lowest in the control group on day 4 and highest in the treatment group on day 13. The results of the One Way Anova statistical test showed a significant difference in epithelial thickness between the control group and the treatment group. This study proved that the 2% leaf extract can accelerate the process of reepithelialization in the healing of wistar mucosal mucosal burns.

Abstract

Orthodontic appliance used to correct malocclusion which may include dental abnormalities, jaw relationship abnormalities, growth disorder of bone-forming facial or oral soft tissues disorder. Mechanical force of orthodontic appliance applied to suppress the teeth and periodontal tissue including gingiva, periodontal ligament and alveolar bone. This process involves the alveolar bone remodeling. Orthodontic mechanical force will cause the periodontal area is divided into two areas: pressure and strain area. In the pressure area, mechanical force will stimulate osteoclasts to perform alveolar bone resorption. Whereas strain area, osteoblasts will perform new alveolar bone formation. Orthodontic tooth movement has three phases process. The phase is initial phase, lag phase and postlag phase. The third phase of tooth movement mechanism involving cellular and molecular reactions in the periodontal tissues.

Abstract

Background: The prevalence of tooth extraction was 7.29% according to RISKESDAS 2018. Post-tooth extraction will cause injury and bone resorption. Scaffold material combination of hydroxyapatite gypsum puger (HAGP) and aloe vera (AV) can be used for preservation of extraction socket. Objective: To analyze fibroblast and type 1 collagen cells in the extraction socket after being induction by a combination of hydroxyapatite gypsum puger scaffold and aloe vera. Materials and Methods: Making a combination of gypsum puger hydroxyapatite scaffold and aloe vera, divided into four groups, namely negative control, aloe vera scaffold, HAGP scaffold, and HAGP+AV scaffold. Extraction of the mandibular 1st molar in Wistar rats, the application on the extraction socket was waited for 3, 5, 7, and 14 days. Preparation of preparations using Hemaktocillin Eosin staining method. Results: LSD test on the HAGP+AV scaffold group between days 3 and 5, days 3 and 7, days 3 and 14, days 5 and 7, days 5 and 14, days 7, and 14 were obtained (p=0.000), this shows a significant difference between the treatment groups. The results of type 1 collagen in the HAGP+AV scaffold group between days 3 and 14 were obtained (p=0.005) showing a significant difference. Each group on day 14 showed the highest number of fibroblast and type 1 collagen cells. Conclusion: The combination of HAGP+AV+gelatin scaffold can increase fibroblast cells and type 1 collagen after tooth extraction. The socket healing process is getting faster on day 14.
