Abstract

We evaluated the effectiveness of vitamin C and Aeromonas salmonicida vaccine in grouper (Epinephelus fuscoguttatus) for increasing immune responses and protection against A .salmonicida. The vitamin C used was polyethoxylated ascorbic and tocopherol. The vaccine was prepared from formalin-killed cells and concentrated extracellular products of a single isolate A. salmonicida. Bath immersion vitamin C and vaccine trials were conducted for 60 min. Fish used had a mean weight 25 g. Control groupers were injected with tryptic soy broth. The results showed that vitamin C enhanced phagocytic activity in head kidney leucocytes of grouper 7, 14, 28 and 36 days after treatments. A significant different of the antibody titre was found between control fish and the treated fish at 42 days after treatments. In addition, at day 42, Relative Percent Survival (RPS) for control group was 53.3 %, vitamin C-treated group was 80.0 % and vaccinated group was 90.0 %. The results of this study suggest that bath immersion of vitamin C provided an increasing of phagocytic activity (non-specific immune responses), titre antibody (specific immune responses) and protection against A. salmonicida infection in grouper. A. salmonicida vaccine also en-hanced titre antibody and protection against A. salmonicida infection in grouper.

Abstract

The accumulation of organic matter at the bottom of the pond greatly affects nutrient metabolism, feed efficiency, growth and productivity in giant tiger prawn cultivation. One of the causes is improper pool bottom construction The purpose of this study was to compare the growth performance and efficiency of protein utilization in giant tiger prawns consuming artificial feed supplemented with multi-enzymes and reared in tarpaulin ponds constructed between not and using the faecal chamber (FAMBER) or the space of metabolic waste and unconsumptioned feed (SOMECOF) at the bottom of the pond with a recirculating aquaculture system (RAS). A tarpaulin pond with a diameter of 3 m and a height of 100 cm filled with 80 cm of seawater. The tarpaulin pond is equipped with a transparent plastic roof, seawater and aeration installation, and a filter in the water pump tank. The average body weight of the experimental prawn used were 8-9 g. The feed given is pellet type artificial feed supplemented with multi-enzymes at a dose of 30 cc/kg of feed. The frequency of feeding were four times a day, namely at 07:00; 12:00; 17:00 and 22:00 with a dose of 5%. The parameters measured were growth performance including growth, condition factors and feed efficiency, while the efficiency of protein utilization included protein content, protein efficiency ratio, amino acid profile, index and score of essential amino acids of feed and experimental prawn. Data analysis was done descriptively. The results showed that all parameters of growth performance and efficiency of protein utilization in tiger prawns reared in tarpaulin ponds using FAMBER were better than those without FAMBER. Based on the results of the study, it was concluded that keeping giant tiger prawns in a tarpaulin pond using a FAMBER is better than not using a FAMBER.

Abstract

Bacterial infectious diseases are a serious obstacle in the cultivation of tilapia (Oreochromis niloticus). The use of herbal ingredients such as cat whisker leaves (Orthosiphon aristatus) is an alternative to environmentally friendly treatment because of its antibacterial compound content. This study aims to analyze the antibacterial activity of cat whisker leaf extract against Pseudomonas aeruginosa bacteria and its effect on the histopathology of liver organs and gills of tilapia. This study used an experimental method with 4 extract dose treatments (50 ppm, 75 ppm, 100 ppm, and 125 ppm) and negative controls, each with 3 replicates. The first stage included the extraction of cat whisker leaves with ethanol solvent, determination of minimum inhibition concentrations through Optical Density (OD) observation using UV/Vis spectrophotometry, and LC-50 24-hour toxicity test. The second stage involves bacterial infection in fish, followed by soaking in the extract for 24 hours and maintenance for 7 days. The parameters observed included histopathology of the liver and gills and survival rate (SR). The results showed that cat whisker leaf extract was able to inhibit the growth of Pseudomonas aeruginosa, which can be seen from the difference in the degree of histological damage to the liver and gills between treatments. The highest survival of tilapia at the end of the maintenance period was found at a dose of 100 ppm with a value of 90% which is the optimal dose to improve the survival of infected fish.
