Abstract

We evaluated the effectiveness of vitamin C and Aeromonas salmonicida vaccine in grouper (Epinephelus fuscoguttatus) for increasing immune responses and protection against A .salmonicida. The vitamin C used was polyethoxylated ascorbic and tocopherol. The vaccine was prepared from formalin-killed cells and concentrated extracellular products of a single isolate A. salmonicida. Bath immersion vitamin C and vaccine trials were conducted for 60 min. Fish used had a mean weight 25 g. Control groupers were injected with tryptic soy broth. The results showed that vitamin C enhanced phagocytic activity in head kidney leucocytes of grouper 7, 14, 28 and 36 days after treatments. A significant different of the antibody titre was found between control fish and the treated fish at 42 days after treatments. In addition, at day 42, Relative Percent Survival (RPS) for control group was 53.3 %, vitamin C-treated group was 80.0 % and vaccinated group was 90.0 %. The results of this study suggest that bath immersion of vitamin C provided an increasing of phagocytic activity (non-specific immune responses), titre antibody (specific immune responses) and protection against A. salmonicida infection in grouper. A. salmonicida vaccine also en-hanced titre antibody and protection against A. salmonicida infection in grouper.

Abstract

Blue swimming crabs (Portunus pelagicus) from Makassar Strait have some variations in color and pattern of spots on the carapace. The purpose of this study was to analyze the habitat preferences blue swimmer crab species complex from Macassart Straits.  Crabs were grouping based on the difference of color and pattern of spots on the crab carapace to identify the variety of crabs.  Habitat was observed salinity, depth, and substrate type. The results of PCA analysis showed a tendency of each population occupies a different base substrates (P<0.01). Substrate type also correlated with depth and salinity. The conclusion of this study is the difference in the color and pattern on the carapace thought to be different species or varieties and they occupy different habitats.  Preferences of the different habitats for different population groups are very important information for the development of blue swimming crab aquaculture in the future. Keywords: Crab, Portunus pelagicus, morphometric, habitat, aquaculture

Abstract

Kegiatan pembesaran ikan kakap putih di beberapa Kabupaten di Sulawesi Selatan yang menggunakan benih yang diproduksi dari balai perikanan di Sulawesi Selatan dan Ambon telah berkembang selama beberapa tahun terakhir. Akan tetapi, benih yang dihasilkan dari kegiatan pembenihan tersebut memiliki kualitas yang masih rendah yang diduga karena keragaman genetik yang rendah. Penelitian ini menggunakan metode Random Amplified Polymorphic DNA, RAPD untuk menganalisis variasi genetik ikan kakap putih tipe liar dari Bulungan Kalimantan Utara dan ikan hasil domestikasi dari panti pembenihan di Ambon yang dibesarkan di tambak rakyat di Desa Borongkalukua Kabupaten Maros Sulawesi Selatan. Hasil penelitian menunjukkan bahwa ikan kakap putih tipe liar memiliki nilai heterozigositas yang lebih tinggi dibandingkan dengan ikan kakap putih hasil domestikasi. Jarak genetik antar individu-individu di dalam populasi tipe liar lebih tinggi dibandingkan dengan jarak genetik individu-individu hasil domestikasi. Dendogram dan pohon filogenetik yang dibentuk berdasarkan evolutionary dissimilarity menunjukkan bahwa pada tingkat ketidakmiripan 10% ikan kakap putih tipe liar dari Bulungan Kalimantan Timur membentuk klaster yang terpisah dengan ikan kakap putih hasil domestikasi dari Ambon yang dipelihara di tambak Dusun Borongkalukua Kabupaten Maros. Keragaman genetik ikan hasil domestikasi lebih rendah dibandingkan ikan tipe liar dan menunjukkan pola evolusi yang pholyphyly Hasil yang diperoleh tersebut menunjukkan bahwa marka RAPD dapat digunakan untuk menganalisis perubahan keragaman genetik antara populasi ikan kakap putih tipe liar dan hasil domestikasi serta bermanfaat sebagai informasi dalam menyusun strategi pemanfaatan wild population sebagai sumber induk dan konservasi plasma nutfah ikan kakap putih.

Abstract

One sub-district that has the potential for marine and fisheries as well as superior tourism in Pangkajene and Islands regencies is Liukang Tupabbiring Utara sub-district. Superior fisheries potential in Mattiro Baji Village and Mattiro Bombang Village in Liukang Tupabbiring Utarasub-district is an opportunity to invest. Small islands in these two villages generally face various obstacles and problems as a consequence of isolation and the difficulty of access to service centers, education, and the economy. Therefore, UNHAS in collaboration with the Universitas Muhammadiyah Makassar and the Regional Government of Pangkep Regency agreed to try to overcome and reduce this problem through the Regional Partnership Program (PKW) in Liukang Tupabbiring Utara sub-district, Pangkep Districts. The Regional Partnership Program (PKW) has produced several activities, namely: Training in making environmentally friendly fishing equipment and sustainable fisheries, post-harvest handling & processing of seaweed and fish products, PIRT management, product packaging and exhibitions, business development of souvenirs and coverage in online media, simple bookkeeping training and marketing for handicraft businesses on Saugi Island, health data mapping, training in waste processing, advocacy for the formation of cooperatives. The implementation of this program can not only improve the economy and quality of public health, but also can encourage environmental quality improvement, so that natural resources can be managed sustainably by better human resources.

Abstract

Penelitian ini bertujuan untuk menganalisis diferensiasi morfologi yang terjadi di antara stok ikan kakap putih, barramundi (Lates calcarifer Bloch, 1790) di perairan pantai Kabupaten Bone, Wajo, Takalar, dan Kalimantan Utara. Diferensiasi karakter morfometrik dianalisis menggunakan fungsi dikskriminan. Hasil penelitian menunjukkan bahwa ikan kakap putih dari perairan Teluk Bone, Selat Makassar dan pesisir Pantai Sekatak Kalimantan Utara bersifat monophyletic. Aliran gen dan migrasi di antara populasi atau stok ikan kakap putih sangat terbatas atau hampir tidak ada. Stok ikan kakap putih di pesisir pantai Desa Akkotengeng Kabupaten Wajo dan stok di pesisir pantai dan daerah aliran sungai (DAS) Sungai Saro Kabupaten Takalar melakukan rekruitmen sendiri. Terdapat satu individu ikan kakap putih di perairan pantai Siwa Kabupaten Wajo dan Cenrana Kabupaten Bone (kedua stok berlokasi di Teluk Bone) yang memiliki fenotipe (morfologi) yang mirip dengan ikan kakap putih di perairan pantai Bulungan Kalimantan Utara. Analisis kontribusi 15 karakter morfometrik terhadap fungsi kanonik menunjukkan bahwa karakter yang menjadi penciri di antara stok adalah tinggi badan, caudal peduncle, dan diameter mata. Hasil discriminant function analysis (DFA) menunjukkan bahwa terdapat tiga populasi lokal yang signifikan berbeda secara fenotipe, sehingga dalam pengelolaannya dibutuhkan manajemen yang berbeda.Kata kunci: Asian seabass, barramundi, karakter morfometrik, Lates calcarifer

Abstract

Tilapia (Oreochromis niloticus) is one of the freshwater fish that is of great interest to the public, because its protein content is quite high, namely 55.58%. One of the problems in tilapia cultivation is that the need for feed is very high, where only 25% of the feed given is converted as a result of production and the rest is wasted as waste. This greatly affects the costs in the cultivation business. One alternative to overcome this problem is the use of Recombinant Growth Hormone (rGH). This study aimed to evaluate the effect of rGH administration on feed conversion ratio and survival of Tilapia (O. niloticus) given rGH through a combination method of submersion and feed rGH (oral boosting). The test animals used tilapia seeds with a size of 2-3 cm with 40 days of maintenance. A plastic jar with a volume of 10 L filled with 8 L of water and stocking density of 40 tilapia seeds. Test animals were given feed commercial made feed at a dose of 5% of body weight. This study used a completely randomized design (RAL) consisting of 4 treatments with 3 replications each, namely A (control), B (submersio), C (feed rGH oral boosting), and D (submersio+feed rGH oral boosting). The results showed that the effect of rGH administration gave the best results in treatment D, submersion+ feed rGH (oral boosting), the ratio feed conversion (FCR) was (0.77 ± 0.02) and survival rate was 92.50%. Provision of rGH through the combination method of submersion + feed rGH (oral boosting) can increase the efficiency of good utilization and high survival. Abstrak Recombinant Growth Hormone (rGH) merupakan salah satu metode alternatif untuk mengatasi masalah pertumbuhan dan tingginya kebutuhan pakan dalam budidaya ikan nila. Penelitian ini bertujuan untuk mengevaluasi pertumbuhan, nisbah konversi pakan dan sintasan benih ikan nila (O. niloticus) yang diberikan rGH melalui metode kombinasi perendaman dan pakan buatan. Ikan uji yang digunakan yaitu benih ikan nila ukuran 2-3 cm sebanyak 480 ekor dengan lama pemeliharaan 40 hari. Ikan dipelihara pada wadah plastik bening dengan volume 10 L yang diisi air sebanyak 8 L dengan padat tebar 40 ekor setiap wadahnya. Ikan diberi pakan buatan dengan dosis 5% dari bobot tubuh. Penelitian ini menggunakan rancangan acak lengkap yang terdiri atas 4 perlakuan dengan masing-masing 3 ulangan, yaitu A (kontrol), B (perendaman), C (pakan), dan D (kombinasi perendaman + pakan). Aplikasi rGH ke pakan buatan komersil dengan dosis 30 mL rGH dalam satu kg pakan, sementara untuk perendaman dengan dosis 100 mL larutan rGH untuk 120 ekor ikan dalam 10 L air. Hasil penelitian menunjukkan bahwa perlakuan perendaman + pakan rGH memberikan hasil yang terbaik, yaitu pertumbuhan spesifik sebesar 5,29 %/hari, nisbah konversi pakan sebesar (0,77 ± 0,02) dan sintasan 92,50%. Pemberian rGH dengan kombinasi perendaman dan oral berpotensi dalam meningkatkan produktivitas dan efisiensi biaya dalam budidaya ikan nila.

Abstract

Penelitian ini bertujuan untuk mengkaji keragaman genetik populasi ikan beronang di perairan Selat Makassar dan Teluk Bone dengan metode random amplified polimorphic DNA (RAPD). Sampel ikan beronang diperoleh dari tiga lokasi di perairan Selat Makassar (Polman, Barru, dan Takalar) dan satu lokasi di perairan Teluk Bone (Malili). Sebanyak 8 ekor ikan beronang dari tiap-tiap lokasi dipreservasi dengan larutan TNES urea buffer. Ekstraksi DNA genom dengan metode Phenol-Chloroform. Di antara tujuh screening primer, lima primer diseleksi untuk analisis RAPD. Keragaman genetik dianalisis menggunakan software TFPGA (Tools For Population Genetic Analysis). Kedekatan hubungan kekerabatan ditampilkan dalam dendrogram. Hasil Penelitian menunjukkan bahwa ikan beronang populasi Malili, Teluk Bone menghasilkan polimorfik tertinggi yaitu 63,24% dan terendah dari populasi Barru, Selat Makassar yakni 40,30%. Indeks similaritas tertinggi (0,83) diperoleh antara populasi Polman dan Barru, dan indeks similaritas terendah (0,65) antara populasi Barru dengan populasi Malili. Hubungan kekerabatan berdasarkan jarak genetik ikan beronang pada penelitian ini diperoleh dua kelompok utama yaitu populasi Selat Makassar dan Teluk Bone. Ketiga populasi ikan beronang asal perairan Selat Makassar merupakan satu kelompok dan terpisah dari ikan beronang populasi Malili, Teluk Bone.

Abstract

Edible films are environmentally biodegradable materials used for food packaging. The edible green alga Caulerpa racemosa has antimicrobial properties; however, its chlorophyll-based bioactive compounds can be damaged when heated so it is prepared in microcapsules. Our research evaluated the effect of Caulerpa microcapsules on an alginate-based edible film on film properties and food spoilage. The microcapsules were used at concentrations of 0%, 0.5%, 1%, and 1.5%. The edible film was measured for film properties, total phenolic content (TPC), antioxidant activity (DPPH), functional group (FTIR) and microstructure (SEM). Food spoilage was evaluated on a popular fish-based product (fish bubble snacks). Bacterial strains of Escherichia coli and Staphylococcus aureus were used to evaluate antimicrobial effectiveness of the edible films. The addition of Caulerpa microcapsules had no significant negative effect on physical properties of the alginate-based film, while the smoother and more homogenous surface should enhance the barrier properties of the film. The slow and evenly distributed release of active compounds from the microcapsules increased resistance to Rhizopus sp. and significantly reduced the proliferation of E. coli but not S. aureus on coated fish snacks. Caulerpa racemosa can be used to enhance the effectiveness of alginate-based films in delaying spoilage and could extend product shelf-life

Abstract

This study aims to evaluate the existing condition of waste sorting place in Makkassar City and the existing regulations in supporting the handling of waste in Makassar City in order to produce formulation of waste sorting system from household to TPS. Methods of data collection used in this study are: (1) study of literature and regulation, and waste management system in several cities and major countries to be adopted in Makassar; (2) field survey to find out the complaints in the community around the place of sorting of garbage; (3) Focus Group Discussion (FGD) from various stakeholders including the community around the place of waste segregation and related SKPD. Likert scale (as one of attitude scale) for the development of statistical analysis technique where ordinal data is converted to interval data, the result will be analyzed using Analytic Hierarchy Process (AHP) to formulate policy about septic and environmentally friendly waste separation place in Makassar City. The result of the research shows that: (1) Waste Bank and TPS3R in Makassar city can improve the economy of society and reduce unemployment, (2) Waste Bank in Makassar city acceptable to society and not cause negative impact, (3) TPS3TR in Makassar City according to people's perception around the location can make dirty air due to dust and environmental smoke, (4) TPS3TR in Makassar City according to the perception of the people around the location can disrupt the general traffic around the TPS3R location, (5) Garbage Bank and TPS3R in Makassar City can improve people's economy and reduce unemployment.
