Abstract

Fungi endophytes are living microorganisms colonizing inside the internal tissue of a plant and provide ecological benefits for their host. Endophytes provide various metabolites for plant adaptation toward biotic and abiotic stresses and have tremendous pharmacological activities. Toona sureni (T. sureni) (Meliaceae) belongs to the Toona genera and is reported to have antimalarial activity. To determine the compounds produced by the endophytic fungus from this plant, we isolated a compound from Periconia pseudobyssoides (P. pseudobyssoides) K5 endophytes from the stem bark of T. sureni. Diketopiperazine cyclo-(S-Pro-R-Leu), a non-ribosomal peptide, was isolated from brown rice fermented at 28 ± 2 ℃ for 30 days. The structure was determined by spectroscopic methods including fourier-transform infrared spectroscopy, mass spectrometry, and 1D and 2D nuclear magnetic resonance techniques. This compound was evaluated for heme polymerization inhibition activity (HPIA) with an IC50 value of 9.89 ± 0.24 mmol/L compared with positive control chloroquine phosphate with an IC50 value of 3.08 ± 0.58 mmol/L. This compound has been categorized as having low activity three times lower than positive control chloroquine phosphate. This information provides new leads about the compound diketopiperazine cyclo-(S-Pro-R-Leu) produced by P. pseudobyssoides K5 endophytes having low activity in inhibiting heme polymerization. In the future, to explore the potency of this compound as antimalarial agent, the other antimalarial test such as lactate dehydrogenase assay might be useful.

Abstract

Three steroids, ergosterol (1), ergosterol peroxide (2) and stigmasterol (3) have been isolated from endophytic fungus, Lasiodiplodia theobromae derived from the root of Aglaia argentea Blume. The steroids were isolated by vacuum chromatography and column chromatography, the chemical structure was established following the analysis of 1D-NMR, 2D-NMR, IR, MS and by comparison with previously reported spectra data. Ergosterol peroxide (2) and stigmasterol (3) were reported for the first time isolated from Lasiodiplodia theobromae endophytic fungus. Cytotoxic activities of the compounds were tested with resazurin assay against HeLa cervical cancer cells, compound 2 displayed strongest cytotoxic activities against HeLa cervical cancer cells with IC50 values of 0.28 µM, while compounds 1 and 3 showed IC50 values of 0.34 µM and 27.32 µM, respectively

Abstract

ABSTRACT. Steroids are secondary metabolic derivatives of terpenes containing the tetracyclic ring system known to exhibit fascinating pharmacological activity. Steroids are distributed in various genera of endophytic fungi including Fusarium genus which lives inside a higher tree such as Chisocheton macrophyllus. The purpose of this research is to identify and characterize the chemical structure of steroids generated by F. phaseoli, an endophytic fungus obtained from C. macrophyllus roots, as well as to assess their antibacterial activity against Escherichia coli and Staphylococcus aureus. The brown rice medium was fermented with F. phaseoli for six weeks before extraction with ethyl acetate. The extracts yielded four compounds, identified using spectroscopic methods such as FTIR, HRTOF-MS, 1D, and 2D NMR, and then compared to previously described compounds. Compounds 1-4 were identified as ergosterol (1), ergosterol peroxide (2), atroside (3), and cerevisterol (4). The four isolated compounds were evaluated for antibacterial activity against Staphylococcus aureus ATCC 6538 and Escherichia coli ATCC 11229 and displayed activity with MIC50 values of 500 µg/mL. Abstract

ABSTRACT This study aims to describe the implementation of adiwiyata school at SD Negeri Serayu Yogyakarta which includes 3 components of the Movement for Care and Culture of the Environment at Schools (PBLHS). This research is a qualitative research. Collecting data used observation, interview, and documentation techniques. The research subjects were the principal, adiwiyata teams, class teachers, and students. The data obtained were analyzed using qualitative descriptive analysis techniques. Examination of the validity of research data using triangulation techniques and triangulation of sources. The results research on the implementation of adiwiyata schools in SD Negeri Serayu showed that: (1) The PBLHS Movement Planning was prepared based on the School Self-Evaluation Report (EDS) and the results of the Identification of Environmental Potentials and Problems (IPMLH) by involving various parties, namely the school principal, the board of educators, and school Committee. The One KTSP document contains the PBLHS Movement and the teacher includes the implementation of PRLH into the RPP; (2) The implementation of the PBLHS Movement has been carried out through learning, extracurricular and self-accustoming that has been integrated into the Application of Environmentally Friendly Behavior (PRLH), the application of PRLH for the community, establishing work and communication networks, publication of the PBLHS Movement, forming and empowering Adiwiyata Cadres; (3) Monitoring & Evaluation of the PBLHS Movement was carried out by involving the principal, adiwiyata team, teachers and students; (4) Supporting factors and inhibiting factors. The supporting factor was the motivation of each individual to protect the environment and the inhibiting factor was the difference in commitment and lack of communication. The implementation of Adiwiyata School at SD Negeri Serayu Yogyakarta was conducted well based on P.53/Menlhk/Setjen/Kum.1/9/2019 which covers the planning, implementation, and monitoring & evaluation of the PBLHS Movement.Keywords: Adiwiyata School, PBLHS Movement, PRLH Implementation

Abstract

Stroke merupakan penyebab utama kecacatan dan kematian di Indonesia. Identifikasi dan pengendalian faktor risiko melalui skrining dini menjadi strategi penting dalam pencegahan stroke, khususnya pada penderita hipertensi dan diabetes melitus. Namun, capaian angka skrining risiko stroke di Puskesmas Manyar, Kabupaten Gresik, masih belum optimal. Kegiatan pengabdian masyarakat ini bertujuan untuk meningkatkan pengetahuan kader kesehatan sebagai upaya mendukung peningkatan capaian skrining risiko stroke. Metode yang digunakan meliputi pembekalan kader kesehatan melalui penyuluhan, pemberian media edukasi (leaflet dan video), serta sosialisasi website SRIKANDI (Skrining Risiko Stroke Mandiri). Evaluasi dilakukan menggunakan desain pre-test dan post-test yang dianalisis secara statistik. Sebanyak 45 kader kesehatan (97,8% dari target) mengikuti kegiatan ini, seluruhnya berjenis kelamin perempuan dengan mayoritas berada pada usia produktif. Hasil analisis menunjukkan peningkatan rerata nilai pengetahuan kader dari 86,00 menjadi 94,89, dengan perbedaan yang bermakna secara statistik (p < 0,05). Kegiatan ini terbukti efektif dalam meningkatkan pengetahuan kader kesehatan mengenai stroke dan pentingnya skrining risiko stroke. Diharapkan kader kesehatan dapat berperan sebagai perpanjangan tangan Puskesmas dalam mengedukasi dan mendorong masyarakat untuk melakukan skrining risiko stroke secara berkelanjutan.
