Abstract

Botryococcus sp. is an economically important microalgae as it contains carbohydrates, lipids, proteins, and a number of phytochemicals. The high concentrations of saturated fatty acids (SFAs) and monounsaturated fatty acids (MUFAs) in this strain also a valuable source for production of biofuel. Mutagenic engineering using irradiation has been known to increase the content of these biomass. The aim of this study is to know the effect of electron-beam irradiation to the growth profile and fatty acid composition of Botryococcus sp. In this study, Botryococcus sp. adjusted an optical density 1.0 after being exposed to different doses of electron-beam irradiation (160 kGy, 240 kGy, and 320 kGy) and induced random mutagenesis for strain improvement was observed based on high energy (1.5 MeV and 2 mA). Several mutants obtained were designated as strain B160, B240, and B320, respectively. The profile growth was significantly different between control (0 kGy) and irradiated microalgae strain. The highest growth is found in B320 mutant. Fatty acid of Botryococcus sp. control produced 7 fatty acids, B160 produced 7 fatty acids, B240 produced 6 fatty acids, whereas B320 produced 9 fatty acids. B320 produced hydrocarbon and phtalic acid as well as fatty acids. Total SFAs and MUFAs of B240 was increased 1.6 times compared to the wild type. The results would give some implications to improve the quality of biodiesel from Botryococcus sp.

Abstract

Endophytic Fungi is one of the natural antibacterial sources that has not been studied. Jamblang (Syzygium cumini L.) is a plant of the Myrtaceae family that has many benefits for health. Endophytic shoots located on the leaves and stems of Jamblang can be one of the natural sources of antibacterial. This study was conducted to obtain endophytic fungi isolates from Jamblang stems and leaves, that have antibacterial activity against Bacillus cereus. The method used at the stage of endophytic shell isolation from Jamblang stem and leaves is Direct Dilution Plate method. At the antibacterial test stage of Bacillus cereus was used agar block diffusion method. Observations were made of the clear zone formed. Isolate Endophytic Fungi which has a clear zone that is D2 Yellow isolate, D4 White Ash, D4 gray Ash, Black B2, B2 White and B3 Yellow. The largest clear zone is shown by B2 Black isolate with a diameter of 14.75 mm.

Abstract

Telah dilakukan penelitian mengenai aktivitas biosurfaktan dari jamur eksogenous terhadap penurunan Total Petroleum Hydrocarbon (TPH) oily sludge asal Balongan. Penelitian ini bertujuan untuk mengetahui kemampuan jamur eksogenous untuk menghasilkan biosurfaktan dan menurunkan TPH oily sludge asal Balongan. Metode yang digunakan adalah metode deskriptif dengan dua kali pengulangan yang terdiri dari tiga tahap. Tahap pertama adalah seleksi 9 isolat jamur penghasil biosurfaktan dengan Oil Spreading Assay, tahap kedua yaitu optimisasi pH untuk pertumbuhan jamur yang paling baik pada medium padat, dan tahap terakhir yaitu degradasi oily sludge dengan fermentasi padat atau Solid State Fermentation (SSF) selama 15 hari. Hasil penelitian menunjukkan bahwa dari 9 isolat jamur eksogenous, yang menghasilkan biosurfaktan terbaik adalah Cladosporium sp. dan Penicillium sp. Derajat keasaman yang optimum untuk pertumbuhan berbeda untuk tiap jamur, yaitu Cladosporium sp. optimum pada pH 5,5 sedangkan Penicillium sp.7 optimum pada pH 4,5. Setelah fermentasi selama 15 hari, Penicillium sp. mampu menurunkan kadar TPH sebesar 19,40% sedangkan Cladosporium sp. menurunkan kadar TPH sebanyak 17,92%.

Abstract

Penelitian dimaksudkan untuk mengisolasi dan menidentifikasi jamur yang terdapat pada karang lunak yang berada di lokasi Pantai Barat Cagar Alam Pananjung Pangandaran, dari penelitian ini bertujuan mengetahui jenis jenis jamur yang ditemukan dari sampel karang lunak yang diperoleh. Metode yang digunakan dalam penelitian ini adalah metode deskriptif dengan menggunakan tiga tahapan. Tahap pertama pengambilan sampel dilapangan dengan metode memotong bagian tubuh karang lunak. Tahapan kedua isolasi jamur yang ada pada bagian karang lunak kedalam medium agar dengan dua jenis penanaman  yaitu penanaman permukaan luar tubuh sampel (epifit), atau penanaman bagian dalam tubuh sampel (endofit). Tahap ketiga  identifikasi jamur dengan menggunakan metode  moist chamber yang kemudian di cocokan dengan buku identifikasi. Dari hasil yang didapatkan terdapat dari karang lunak jenis Euphyllia Sp. didapatkan sembilan isolat dengan tiga isolat pada sampel yang diisolasi secara epifit, dan enam spesies yang terdapat pada isolat pada sampel yang diisolasi secara endofit. Jamur yang isolasi secara epifit adalah  Saccharomyces Sp. dan Cladosporium Sp. dan jamur yang berasal dari isolasi secara endofit adalah Neurospora Sp., Apergillus Sp., Saccharomyces Sp., dan Geotrichum Sp.

Abstract

Phycocyanin is Spirulina’s content which has antioxidant activity to inhibit oxidation process or formation of free radicals that play a role in the initiation high-degenerative diseases. This research is aim to determine the best antioxidant activity of S. platensis with S. fusiformis through the best extraction methods of Spirulina cell between distilled water and PBS (Phosphate Buffer Solution). This research method existed Spirulina cell extraction used distilled water and PBS, and then followed by fractionation and dialysis phycocyanin extract, afterward tested the antioxidant activity used DPPH method. The result of extraction used PBS with a salt concentration of ammonium sulfate 30% produced the best purity value of phycocyanin extract of both types of Spirulina. Results of tested the antioxidant activity, S. fusiformis has the best antioxidant activity compared with S .platensis with a value of 311.43 ppm and 387.36 ppm, respectively.Keywords: Antioxidant, DPPH, Phycocyanin extracts, S. fusiformis, S. Abstract

Various ornamental fish, such as koi and comet is a mainstay commodity from Nagrak Village, Cisaat District, Sukabumi Regency, West Java. The ornamental fish farmers in this village have been grouped in the form of pokdakan called “Pokdakan PBC Fish Farm”, which is chaired by Ikral Panutan. The main problem faced by members of this group is the color of the ornamental fish which still in low grade in quality. The purpose of this activity was to improve the quality of koi ornamental fish by using additional powder feed spirulina so that it can give added value to the production of ornamental fish produced. Community empowerment activities have been conducted for 4 months from July to October 2016. Steps of implementation include: 1) Socialization and initiation stage; 2) Training about fish culture management, spirulina culture method and feeding method using discussion method and demonstration plots; and 3) Evaluation and monitoring phase. The results obtained are generally these activities get the response and good results from the extension participants, as evidenced by the ability of farmers in cultured spirulina henceforth be applied as a mixture of fish feed can result in improved color quality of koi. Use of spirulina powder feed additional material on Cyprinus carpio of PBC Fish Farm proven to improve color quality, and provide value-added economy by 50–100 with the selling price ranging from IDR 3.000, per head to IDR 4.500–6.000 per tail.

Abstract

Biodegradation of rice straw was performed by solid state fermentation using two isolates Penicillium to assess degradation time and effect of particle size on the growth and enzyme  production. Biodegradation of the straw was conducted over 12 days using three straw particle size of 4, 8, and 12 mesh. The results showed that particle size affects the degradation process of rice straw. Delignification influenced by activity of ligninase optimum size of 12 mesh with the degradation time in 2-4 days. Penicillium sp1 produced ligninase with the highest activity 1140 U/mL (lignin content 55.2%) and cellulase 140 U/m (content of cellulose 27.6%)., while highest ligninase and cellulase activity of Penicillium sp2. 882 U/mL (lignin content 49.4%) and 102 U/mL (content of cellulose 15.9%).Keywords: biopulping, biodegradation, rice straw, Penicillium spp.ABSTRAKBiodegradasi jerami padi telah dilakukan secara fermentasi media padat dengan menggunakan dua isolat Penicillium untuk mengkaji waktu degradasi dan pengaruh ukuran partikel jerami terhadap pertumbuhan Penicillium spp. dan produksi enzim lignoselulolitiknya. Biodegradasi jerami padi dilakukan selama 12 hari dengan menggunakan tiga ukuran partikel jerami 4, 8, dan 12 mesh. Hasil penelitian menunjukkan bahwa ukuran partikel berpengaruh terhadap proses degradasi jerami padi. Delignifikasi dipengaruhi oleh aktivitas ligninase yang optimum pada ukuran 12 mesh dengan waktu degradasi 2-4 hari. Penicillium sp1. menghasilkan ligninase dengan aktivitas tertinggi 1140 U/mL dan selulase 140 U/mL, sehingga dapat mendegradasi lignin sebanyak 55,2% dan selulosa 27,6%, sedangkan aktivitas tertinggi dari ligninase dan selulase dari Penicillium sp2., masing-masing adalah 882 U/mL dan 102 U/mL dengan kadar lignin yang terdegradasi adalah 49,4% dan selulosa 15,9%.Kata kunci: biopulping, biodegradasi, jerami padi, Penicillium spp.

Abstract

Introduction: One strategy for molecular cancer therapy is to know the key mechanism of cytotoxic compounds that can kill cancer cells. Ramie (Boehmeria nivea L. Gaud.) leaves contain active compounds that have important effects on cancer chemoprevention. Objective: To obtain the active fraction of a Ramie leaf extract in inhibiting the proliferation of MCF-7 breast cancer cell lines and to determine the mechanism of apoptosis induction using MCF-7 and Saccharomyces cerevisiae strains 1140, 1353, and 1138. Method: Fractions were prepared using n-hexane, dichloromethane (CH2Cl2), ethyl acetate, and n-butanol as solvents. All fractions were tested qualitatively through phytochemical. The MTT-based cytotoxicity assay used MCF-7 (in vitro) to obtain the IC50 value, whereas the model system that targets the enzymatic (topoisomerase) used a yeast-based bioassay to obtain the IC12 value. Apoptotic induction of the active fraction in MCF-7 was performed using flow cytometry and qPCR (2-ΔΔCt method). Results: The phytochemical analysis indicated that the extract fraction consisted of alkaloids and steroids. The smallest IC50 value was obtained from the CH2Cl2 fraction as 3.79 g/mL, potentially act as an anticancer. A higher percentage of apoptosis than that of necrotizing cells and live cells was observed through flow cytometry. The CH2Cl2 fraction with an IC12 value < 8000 in strains 1140, 1353, and 1138 consistently showed the mechanism of apoptosis induction as topoisomerase I and II inhibitors. Also, another mechanism could be through the intrinsic pathway, indicated by the highest expression level in p53. Conclusions: The CH2Cl2 fraction of Ramie leaves can inhibits the proliferation of MCF-7 cells in the active and strong categories. The CH2Cl2 fraction induces apoptosis by increasing p53 gene expression and inhibiting topoisomerase I and II. Thus, it showed potential as an anticancer drug candidate.

Abstract

AbstrakSaat ini, perbanyakan rami menggunakan rizoma sebagai bahan tanamnya, tetapi dalam produksinya membutuhkan waktu yang lama dan sebagai bahan tanam umur simpannya singkat. Kultur jaringan merupakan salah satu teknologi untuk mendapatkan bahan tanam yang seragam dan sehat dalam waktu yang singkat. Tujuan dilakukannya penelitian ini adalah untuk mengetahui pengaruh konsentrasi BAP terhadap pertumbuhan tunas aksilar pada rami klon lokal Wonosobo dan konsentrasi mana yang memberikan pengaruh terbaik. Percobaan dilakukan di Laboratorium Kultur Jaringan, Teknologi Benih, Fakultas Pertanian, Universitas Padjadjaran. Nodus batang dari rami klon lokal Wonosobo diklturkan pada media MS dengan penambahan berbagai konsentrasi BAP selama 8 minggu dan diamati pertumbuhan dan perkembangannya. Desain penelitian yang digunakan adalah Rancangan Acak Lengkap (RAL) dengan 6 perlakuan yang diulang sebanyak tiga kali. Adapun perlakuannya adalah kontrol (tanpa penambahan BAP), BAP 0,5 mg/L, BAP 1,0 mg/L, BAP 1,5 mg/L, BAP 2,0 mg/L, dan BAP 3,0 mg/L. Hasil percobaan menunjukkan penambahan sitokinin berpengaruh terhadap pertumbuhan tunas aksilar rami klon lokal Wonosobo. Penambahan 0,5 mg/L berpotensi memberikan pengaruh terbaik terhadap pertumbuhan tunas aksilar rami klon lokal Wonosobo dilihat dari waktu muncul tunas, jumlah tunas, jumlah daun, dan tinggi plantlet. Kata Kunci: Benzylaminopurine (BAP) ∙ Rami ∙ Tunas aksilar AbstractCurrently, ramie propagation used the rhizome as the planting material. However, it took long time to produce and the longevity of rhizome as planting material is short. Therefore, another technology approach is needed. Tissue culture is one of alternative technologies that could to produce uniform and healthy planting material within short time. The objective of this research was to determine the effect of BAP concentration on the axillary bud growth of rami local clone of Wonosobo and also determine which concentration gives the best effect. The research was conducted at The Tissue Culture Laboratory of Seed Technology, Faculty of Agriculture, Universitas Padjadjaran. Nodes of ramie local clone of Wonosobo were cultured for 8 weeks on Murashige & Skoog (MS) medium added with various concentrations of BAP and then observed growth and development. The research design was Completely Randomize Design (RCD) with 6 treatments in terms of BAP concentrations and 3 replications. The treatment was ,control (without BAP); BAP 0,5 mg/L, BAP 1,0 mg/L, BAP 1,5 mg/L, BAP 2,0 mg/L, and BAP 3,0 mg/L. The result showed that there was different effect on axillary bud growth. The treatment of MS medium added with 0,5 mg/L BAP potentially showed as the best effect for bud appearance, number of shoots, number of leaves, and plantlet’s length.Keywords: Benzylaminopurine (BAP) ∙ Ramie ∙ Axillary bud
