Abstract

The aim of this study was to  isolate bacteria that could potentially degrade the cellulose from of agricultural land in the village  Candikuning Bedugul and to determine the ability of isolates in degrade cellulose.These  bacteria are capable of utilizing cellulose as carbon source 11 only isolates that produce a clear zone with diameters above 12.07 mm up to 45.59 mm on Gram ’sIodine test.  Eleven isolates bacteria cellulolytic pure who afford occurrence bacterial growth on media selective and isolates produce clear zone that most highest is CK 18.Keywords: cellulose degrading bacteria, isolation, Gram's Iodine test.

Abstract

The aimed of this research were 1) determine the effect of maceration time and maltodextrin concentration on the characteristics of mandarin citrus peel extract powder, 2) find out the maceration time and maltodextrin concentration to obtain the best characteristics of the mandarin citrus peel extract powder. This study used a factorial randomized block design. The first factor was maceration time consists of 4 levels, namely: 6, 12, 18 and 24 hours, and the second factor was maltodextrin concentration which consists of 3 levels: 10, 15 and 20 %. Each treatment was done 3 times. The parameters tested include yield, solubility, sensory testing. The results showed that maceration time had no significant effect on the yield and solubility of mandarin citrus peel extract powder. Maltodextrin concentration effect had signifcant on the yield and solubility of mandarin citrus peel extract powder. Treatment interaction between maceration time and maltodextrin concentration had significant effect on taste preferences, the strength of taste, flavour preferences, and flavour strength. However, no significant effect on yield and solubility of mandarin citrus feel extract powder. 6 hours maceration time and 10% maltodextrin concentration produced the best characteristics of mandarin citrus peel extract powder, include: yield 42.91%, solubility 99.75%, taste preferences 6.13 (between like until very like), the strength of taste 2.07 (between strong to strong a bit), aroma preferences of 5.27 (between like a bit to like), the strength of the aroma of 1.53 (between very strong to strong).

Abstract

This study aims were to determine the effect of medium type on biomass of Tetraselmis chuii and to find out the best type of medium for producing biomass of Tetraselmis chuii. This study used a descriptive quantitative method with treatment of medium consisting of 3 medium types, namely Walne, Miquel-Allen (MQ), and Bold Basal Medium (BBM). Observation of cell biomass concentration Tetraselmis chuii was used haemacytometer with 3 replications. The results of this study showed that the medium type had significantly affect on the biomass concentration of Tetraselmis chuii. BBM was the best medium to produce the highest biomass of Tetraselmis chuii with a density 2.2 x 10 6 (cells/ml) on 10 days cultivation

Abstract

The aim of this study was to understand the effect of sodium nitrate and sodium phosphate concentration on Guillard medium to yields of biomass, and also to determine the best concentration of sodium nitrate and sodium phosphate to produce the highest yields of biomass on marine microalgae Nannochloropsis sp. Determining the effects of sodium nitrate and sodium phosphate on biomass was performed using Response Surface Methodology (RSM). The concentrations of sodium nitrate and sodium phosphate on Guillard medium affects the yields of biomass on marine microalgae Nannochloropsis sp. The optimum concentration of sodium nitrate and sodium phosphate to produce the highest yields of biomass were 178.14 g/l and 13.34 g/l respectively, with biomass concentration of 8.00 x 107 cell/ml. Abstract

This study aims to know the effect of concentration of Sodium Nitrate (NaNO3) and Sodium Dehidrogen Phosphate (NaH2PO4) on walne media on the biomass concentration and protein Nannochloropsis oculata and determine the best concentration of NaNO3 and NaH2PO4 on the Walne media to produce Nannochloropsis oculata biomass with highest protein content. This study was designed with a randomized block design with two factors, each treatment was divided into two by the time of cultivation.The data obtained were analyzed by Anova, if there was a real effect will be followed by Duncan test. The results showed that the concentration of NaNO3 and NaH2PO4 on media Walne had significant effect on the concentration of the Nannochloropsis oculatamicroalgae biomass and protein content, and no interaction of both factors.The concentration of NaNO3 of 200 g/L and NaH2PO4 of 25 g/L at Walne media producethe hight biomass concentration of Nannochloropsis oculata biomass amount of 7.66 x 106cells / ml and 7.13 x 106cells / mlrespectively. the highest protein content of Nannochloropsis oculata was produced at nitrate concentration of 200 g/L amount of 25.74% and 25 g/L phosphate consentration of amount 22.93%. Abstract

The determined of these research were to knowing optimal of temperature and carbon source of biodesulfurization isolate SBJ8. Isolate growth at Medium MSSF-TD with concentration of 200 ppm DBT in tetradecane which tested various temperature 37?C, 41?C, 45?C. The next step was to determine the best carbon source using glucose, sucrose and citric acid. The result of experiment showed isolate SBJ8 temprature at 37?C and pH 7 with carbon source glucose. In condition SBJ8 could growth (OD660) from 0,765, and 0,411 with DBT degradation rate in tetradecane each 76,7%, and 63,4%. Keyword: biodesulfurization, SBJ8, dibenzothiophene, tetradecane.

Abstract

The aims of this study were to know the effect of addition of NaNO3 and K2HPO4 on BG-11 medium on biomass and chlorophyll concentration of Tetraselmis chuii and to fine out the best concentration of NaNO3 and K2HPO4 on BG-11 medium to produce highest biomass and chlorophyll concentration of Tetraselmis chuii. This study used a factorial randomized block design.The first factor was NaNO3consists of three levels i.e. 0.5, 1.5 and 2.5 g/l. The second factor wasK2HPO4consists of three levels i.e. 0.020, 0.035 and 0.050 g/l. The experiments were grouped into 2 groups based on the time of culturitation, in order to obtain 18 experimental units. Data were analyzed by analysis of variance and if the treatment significantly affected the parameter followed by Duncan test. The result showed that addition of NaNO3 and K2HPO4had very significant effect on the concentration of biomass and chlorophyll content of Tetraselmis chuii. The addition of NaNO31.5 g/l and K2HPO40.035 g/l was produce the highest of biomass and chlorophyll concentration of 3.3 x 106cells/ml and 10.17 mg/g, respectively. Abstract

This study aimd toknowthe effect of addition of magnesiumsulfate heptahydrate and ferric chloride in Blue Green Medium-11 on the production biomass of Tetraselmis chuii and tofine out the concentration of magnesium sulfate heptahydrate and ferric chloride on Blue Green medium-11 which could produce the highest biomass of Tetraselmis chuii. The Experimens was designed by a completely randomized design (CRD) with factorial pattern two factors were designent in the experient. The first factor was magnesium sulfate heptahydrate addition which was consist of three levels i.e. 0, 2, and 4g/L. The second factor was ferric chloride addition which was consist of three levels i.e. 0, 12, and 24 µM. Experiement was repeated twice times to obtain 18 units experiment. Data were analyzed by analysis of variance and if the treatment significantly affected the parameter followed by Duncan test. The addition of magnesium sulfate heptahydrat e4g/L producedin the highest biomass concentration of 4.4x10 6 cells/ml while the addition ferric chloride 24 ?m produce the highest biomass concentration of 4.3x10 6 cells/ml.

Abstract

urpose of this study were 1). assessing the effect of tape inoculum addition and fermentation time on the characteristics of the cacao bean pulp, 2). determine the treatment could provide an indicator in the best process of destruction of cacao bean pulp. This study used randomized block design (RAK) with two factor. The first factor was the concentration of tape inoculum addition that consists of 5 levels ie 0% (no control), 0.5%, 1%, 1.5% and 2%. The second factor was the time of fermentation which consists of 5 levels namely 1 day, 2 days, 3 days, 4 days and 5 days. The experiments were grouped into 2 groups based on the time of implementation, in order to obtain 50 units experiment. The result showed that the treatment of tape inoculum addition had very significant effect on pH, total acid, pectin content and reducing sugar of cocoa bean pulp. The treatment of fermentation time had very significant effect on pH and reducing sugar of cocoa bean pulp, but did not affect on the total acid,and pectin content. Interaction of the treatmens had no effect on pH, total acid, pectin content, reducing sugar of cocoa bean pulp. The treatment of 2% tape inoculum addition and 4 days fermentation was the best treatment to destroy pulp during cacao bean pulp fermentation with the best characteristics was 2.10% pectin content, reducing sugar of 0.70%, total acid of 0.50 meq NaOH / g, and pH of 3.90. Abstract

The aims of this study was to determine the optimum substrate concentration of cassava peel and fermentation time on crude cellulase activity from Trichoderma viride FNCC 6013. Optimization on crude cellulase activity from Trichoderma viride were calculated using the response surface method. Central Composite Design was used to study the effect of optimum substrate concentration of cassava peel and fermentation time on maximum crude cellulase activity (filter paperase). The results showed that 3.14% substrate concentration of cassava peel and 7.26 days fermentation produces maximum crude cellulase activity (filter paperase), amounting to 0.099 Units/mL.
