Abstract

The availability of source material to multifunctional tree Toona sp. have constraints for macro and micro propagation, in this regard, this study aims to:1)observe the formation of buds on T. sinensis and T. sureni stem cuttings in an effort to sustain the availability of explants and 2)analyze the effect of GA4, IBA and BAP growth hormone on regeneration and propagation of the axillary bud initiation and callogenesis through tissue culture. Isolation of stem cuttings was conducted in a greenhouse with number of shoots per nodule and nodule distance as observed.Variables Tissue culture techniques were used to study the effect of exogenous GA4 hormones application on the initiation of axillary buds. The application of IBA hormone on plantlet rooting and callogenesis. The results showed that T. sinensis has 2-7 buds per nodule and nodule distance of 2-5 mm, while T. sureni has one bud per nodule and nodule distance of 5-10 cm. The average length of T. sinensis shoots were 15.8 cm and 17.4 for T. sureni. Enrichment with 1 mg/l GA4 gave the highest axillary shoot length on T. sinensis (8.5 cm ± 0.7228) and (9.8 cm ± 0.1022) for T. sureni. Two mg/l IBA enrichment gave the highest root length of T. sinensis (10.9 cm ± 1.8392) and for T. sureni (7.9 cm ± 0.7633). Three mg/l concentration BAP application gave the best effect in term of callus weight of T. sinensis (1.4 g ± 0.3833). Scoring of callus weight showed moderate response categories to both T. sinensis and T. sureni.

Abstract

This study aims to observe acid tolerance of Vitex pubescens culture growth. Planlet cultures were established from seeds on Murashige & Skoog medium supplemented with 1 µM benzylaminopurin; 0,1 µM kinetin and 0,01 µM naphtalenaceticacid. The pH was adjusted to 5,7;4,0 and 3,0. The mean height of planet were 4,44 cm ± 0,32 for pH 5,7; 2,88 cm ± 0,54 for pH 4 and 1,10 cm ± 1,38 for pH 3. The mean length of root were 13,72 cm ± 0,47 for pH 5,7; 18,88 cm ± 0,60 for pH  and 19,27 cm ± 0,73 pH 3. The mean number of branch roots per cm were 1,65 ± 0,058 for pH 5,7;3,88 ± 0,18 for pH 4 and 0,57 ± 0,30 for pH. After 12 weeks acclimatization in Yellow Red Podzolic (YRP) soil, the mean height of plant with pH 5,7 culture were 29,86 cm ± 0,64 and 26,72 cm ± 0,40 for pH culture. In vitro acid tolerance of Vitex pubescens were at pH 4 and can grow in ex vitro condition in YRP soil with pH 4,48.

Abstract

Falcataria moluccana (Miq.) is severely attacked by Uromycladium tepperianum (Sacc.) McAlpine, which is a gall-forming (neoplasmic) and parasitic obligate pathogen. Phenolic compounds have the ability to function as co-factors of pathogenicity determinant of pathogens development and chemical defenses of plants. The purpose of this study was to determine the content of phenolic compounds on gall rust tolerance sengon by observation of tolerant sengon height, quantitative analysis of total phenolic compounds, wood anatomy analysis and tolerance test of sengon callus from tissue culture with filtrate gall rust as chemical agent. The results of the research were (1) inoculated sengon have lower total phenolic compounds than the control (no inoculation), (2) tolerant sengon have lower content of phenolic compounds than sensitive one, (3) microscopic wood anatomy observation shows that tolerant sengon have darker substrat, and (4) in the same concentration of gall rust filtrate incubation media, the tolerant sengon have higher survival cell calli. The highest concentration that callus cells can survive was 25% (v/v).

Abstract

Malapari (Pongamia pinnata) seeds have low sensitivity to desiccation and freezing storage. In the effort to improve the viability of seed storage, preliminary test of the vacuum ultradry storage techniques were important to be done. The purpose of this study was to attain lengthen periode of viability seeds with vacuum ultradry technics. All parameters of the treatment with a vacuum ultradry technique showed better results than the control. Interaction between humidity and incubation period have significantly influnces on shoots length. Interaction between moisture with incubation period and intercation beetwen humidity with temperatures has significant effect on root length. Interaction between moisture, incubation period and temperature have significantly affect to vigor index. The highest vigor index was 8.1050 at 20% humidity, 12 weeks incubation period with temperature 5C,86% higher than in the control.

Abstract

Effect of Gibberellins (GA4) on Germination Time and Height of Cendana (Santalum album Linn.) The hemiparasite Santalum album Linn. (cendana) grows very slow, in nature the rare and difficult seeds need stimulation to germinate. Gibberellins (including GA4) are growth regulators, usually used to increase growth as well as to break seed dormancy. The objectives of this research were to investigate the influence of gibberellins on germination percentage and height of cendana growth. Experiment was laid out in a Completely Randomized Design with 3 replicates of 300 seeds for germination percentage and 3 replicates of 10 seeds for seedling growth. Gibberellin was applied as treatment with 100, 300, and 500 ppm. The seed germination was recorded until 9 weeks, and height of plants measured until 8 months at the greenhouse. The results showed that the addition of gibberellins at all treatment increased the percentage of germination and caused the seeds germinated four weeks earlier than the control. In the first 4 months, 500 ppm gibberellins gave the highest acceleration of germination, afterward all treatments have relatively the same influences. Gibberellins gave positive effect on height of cendana growth. After 7 months, the growth decreased although all gibberellin treatments gave higher growth than the control.
