Abstract

Forty four clones of Acacia hybrid  propagated from selected stoolplants were tested together with a control of pure species seedling and clone in a clonal test established at Wonogiri, Central Java. The design of clonal test was Incomplete Block Design, which was laid-out as single treeplot of 20 replications with spacing of 3 m x 3 m. Measurements of tree height were conducted at age 12 months. There were significant differences between the clones in height growth. Eleven clones of 44 were more superior than the original population at SSO F1 (Control 4th) which superiority ranging from 1.81-35.35 % and 3 of them were more superior than the pure species (control 1th, control 2th and control 3th) which superiority ranging from 3.51 – 17.28%. Meanhile, clone repeatability for height at age 12 months was, while individual repeatability for height was 0.59 +- 0.1

Abstract

The research aim is to observe tissue culture method for Sandalwood using node explants. The explants were cultured on Murashige and Skoog (MS) medium solidified with agar and supplemented with varies combination of hormones: BAP (benzyl-amino-purine), NAA (napthalene-acetic-acid), IAA (indole-acetic-acid) and Kinetin (furfuril-amino-purine) for shoot induction, multiplication and rooting. The results of study showed that the medium of MS+1 mg/l BAP+0.01 mg/ lNAA provided a good response for shoot induction of Sandalwood clones number A.III.4.14 at around 85%. The medium of MS+0.5 mg/l BAP+0.01 mg/l NAA provided a good response for shoot multiplication of the clones number A.III.4.14 (number of shoot and shoot elongation). The rooting medium of ½MS+20 mg/l IAA+1 mg/l IAA and 0.01 mg/l NAA resulted rooting percentage across the clones at around 37%. The highest survival rate after acclimatization was found at clone number WS6 at around 56%.

Abstract

The first generation of Acacia hybrid between A. mangium and A. auriculiformis potential to be developed for plantation because of its superiority. The superiority of hibrid Acacia were fast growth, straight bole, light branching, more tolerant to pest/disease attack and better wood property than the parent trees. The only way to maintain their superiority is to propagate it vegetatively. The objective of the research was to observed characteristics of hibrid Acacia based on seed viability,sprouting and rooting ability. Materials used in this study were collected seed from control pollination at breeding garden which verified by molecular marker. The result showed that the character of seed viability on hybrid Acacia was still low (48.1%) meanwhile the sprouting and rooting ability were 16.5 ± 0.7 shoots and 78,4% respectively.

Abstract

The aims of this study were to assess genetic diversity within populations and genetic relationship among populations of C. inophyllum. Leaf samples as template DNA were collected from 10 natural populations and 1 plantation. Five random amplified polymorphism DNA (RAPD) markers consisted 30 loci were conducted to genetic analysis. Results showed genetic diversity within populations were in low to moderate level (mean HE=0.186). There is no private allele in any populations. The analysis of molecular variance (AMOVA) showed that genetic differentiation among Islands was insignificant; but the differentiation was siginificant among populations and individual trees. Genetic distance among populations was in low to moderate level (mean Da=0.250). Cluster analysis clearly divided the 11 populations into 2 clusters; cluster I consisted Selayar, Lombok, Gunung Kidul and Padang populations; cluster II consisted Way Kambas, Madura, Ketapang, Dompu, and Yapen populations. The genetic relationships did not associate with their geographical locations. In conclusion, genetic diversity and genetic relationship among populations of C. inophyllum was in moderate level.

Abstract

Falcataria moluccana (Miq.) is severely attacked by Uromycladium tepperianum (Sacc.) McAlpine, which is a gall-forming (neoplasmic) and parasitic obligate pathogen. Phenolic compounds have the ability to function as co-factors of pathogenicity determinant of pathogens development and chemical defenses of plants. The purpose of this study was to determine the content of phenolic compounds on gall rust tolerance sengon by observation of tolerant sengon height, quantitative analysis of total phenolic compounds, wood anatomy analysis and tolerance test of sengon callus from tissue culture with filtrate gall rust as chemical agent. The results of the research were (1) inoculated sengon have lower total phenolic compounds than the control (no inoculation), (2) tolerant sengon have lower content of phenolic compounds than sensitive one, (3) microscopic wood anatomy observation shows that tolerant sengon have darker substrat, and (4) in the same concentration of gall rust filtrate incubation media, the tolerant sengon have higher survival cell calli. The highest concentration that callus cells can survive was 25% (v/v).
